The Helicobacter pylori Neutrophil Activating Protein (HP-NAP) is endowed with immunomodulatory properties that make it a potential candidate for anticancer therapeutic applications. By activating cytotoxic Th1 responses, HP-NAP inhibits the growth of bladder cancer and enhances the anti-tumor activity of oncolytic viruses in the treatment of metastatic breast cancer and neuroendocrine tumors. The possibility that HP-NAP exerts its anti-tumor effect also by modulating the activity of innate immune cells has not yet been explored. Taking advantage of the zebrafish model, we examined the therapeutic efficacy of HP-NAP against metastatic human melanoma, limiting the observational window to 9 days post-fertilization, well before the maturation of the adaptive immunity. Human melanoma cells were xenotransplanted into zebrafish embryos and tracked in the presence or absence of HP-NAP. The behavior and phenotype of macrophages and the impact of their drug-induced depletion were analyzed exploiting macrophage-expressed transgenes. HP-NAP administration efficiently inhibited tumor growth and metastasis and this was accompanied by strong recruitment of macrophages with a pro-inflammatory profile at the tumor site. The depletion of macrophages almost completely abrogated the ability of HP-NAP to counteract tumor growth. Our findings highlight the pivotal role of activated macrophages in counteracting melanoma growth and support the notion that HP-NAP might become a new biological therapeutic agent for the treatment of metastatic melanomas. 相似文献

Well-dispersed concentrated aqueous suspensions of Al2O3-doped Y-TZP (AY-TZP), AY-TZP with 5.4 vol% of CaO–P2O5–SiO2 (CaPSi) glass (AY-TZP5) and 10.5 vol% CaPSi glass (AY-TZP10), with ammonium polyacrylate (NH4PA) dispersant were prepared to produce slip cast compacts. The rheological properties of 35 and 40 vol% slips were studied. The densification, microstructure as well as hardness and fracture toughness were investigated as a function of CaPSi glass content at 1300°C-1500°C. The optimum NH4PA concentration of 35 vol% AY-TZP5 and AY-TZP10 slips at pH ~9 was found to be about 43% and 67% greater than that of AY-TZP slips; this behavior was related to the greater amounts of Ca2+ ions leached out from the CaPSi glass surface. The viscosity of stabilized 40 vol% slips with NH4PA attained a minimum value at 5.4 vol% CaPSi glass addition, and resulted in a more dense packing of cast samples. AY-TZP5 can be sintered at a lower temperature (1300°C) compared to that of AY-TZP. AY-TZP5 exhibited a fine microstructure of tetragonal ZrO2 (grain sizes below 0.3 µm), and ZrSiO4–Ca2P2O7 particles homogeneously distributed within the zirconia matrix. Amyloses from selected corn starches were used to measure the glucoses involved in the branching points. The number of glucose α 1,6 glycosidically linked were determined by two different methods: 1) The enzymatic one measures directly quantiatively and specifically the glucoses α, 1,6 linked. 2) The other, a chemical one that measures the different n-methylated glucoses involved in the terminal, α 1,4 linked and α1,6 linked positions. In this study, amaranth flour was used as an ingredient to prepare gluten-free cookies. The production process and attributes of amaranth cookies were characterised, and the potential use of amaranth flour as a functional ingredient was analysed. Cookies exhibited a non-uniform reddish brown colour and a cookie factor ratio of 4.5 ± 0.6. Storage studies indicated that after 3 weeks at room temperature cookies presented slight variations in the texture. Simulated gastrointestinal digestion of this product was able to release peptides capable of exerting potential antithrombotic and antihypertensive activities, IC50 values of 0.22 ± 0.04 and 0.23 ± 0.03 mg mL−1 protein, respectively. This work demonstrates for the first time that food made with amaranth flour exerts potential antithrombotic and antihypertensive activity. In conclusion, these amaranth cookies could be an alternative way of incorporating potentially health beneficial products for people who choose a conscious diet, including coeliac or vegan consumers. 相似文献

Extracellular matrix (ECM) of myocardium plays an important role to maintain a multilayered helical architecture of cardiomyocytes. In this study, we have characterized the structural and biomechanical properties of porcine myocardial ECM. Fresh myocardium were decellularized in a rotating bioreactor using 0.1 % sodium dodecyl sulfate solution. Masson's trichrome staining and SEM demonstrated the removal of cells and preservation of the interconnected 3D cardiomyocyte lacunae. Movat's pentachrome staining showed the preservation of cardiac elastin ultrastructure and vascular elastin distribution/alignment. DNA assay result confirmed a 98.59 % reduction in DNA content; the acellular myocardial scaffolds were found completely lack of staining for the porcine α-Gal antigen; and the accelerating enzymatic degradation assessment showed a constant degradation rate. Tensile and shear properties of the acellular myocardial scaffolds were also evaluated. Our observations showed that the acellular myocardial ECM possessed important traits of biodegradable scaffolds, indicating the potentials in cardiac regeneration and whole heart tissue engineering. 相似文献

Perfluorinated alkylated substances (PFAS) are of global interest due to their occurrence and persistency in the environment. This study includes surface waters and sediments for the analysis of eleven PFAS. The PFAS studied can be grouped in perfluoroalkyl carboxylates (PFCAs), perfluoroalkyl sulfonates (PFS) and perfluoroalkyl sulfonamides (PFSA). The two most important compounds are perfluorooctanoate (PFOA) and perfluorooctane sulfonate (PFOS). These two substances showed the most significant values for surface water samples with maximum concentrations of 21 ng l−1 for PFOA and 37 ng l−1 for PFOS. Sediment samples from seven Austrian lakes and the river Danube were studied. Whereas PFSA and PFS were not detected in any sediment sample PFCAs were detected in most of the lake samples in concentrations up to 1.7 μg kg−1 dry wt. PFOA, perfluorohexanoic acid (PFHxA) and perfluoroheptanoic acid (PFHpA) were detected in all Danube river sediment samples in concentrations varying from 0.1 up to 5.1 μg kg−1 dry wt. For the various sampling points the proportional mass flows deriving from wastewater discharges were calculated. Whereas only up to 10% of the average flow is discharged wastewater up to more than 50% of the PFAS mass flows in the rivers can be attributed to wastewater discharges. Besides wastewater different other pathways as emissions from point sources, further degradation of precursor products, runoff from contaminated sites or surface runoff as well as dry and wet deposition have to be considered as relevant sources for PFAS contamination in surface waters. 相似文献

Local authority planning departments within the United Kingdom are required to undertake gender mainstreaming as a result of European Union and domestic government requirements. However, research undertaken for the Royal Town Planning Institute demonstrates that few local authorities are doing so. A mêlée of competing equality and diversity considerations at local government level limits the attention given to gender considerations. A review of national policy guidance and planning law indicates that central government is taking an equivocal stance towards gender. It is concluded that before effective gender mainstreaming can take place institutional difficulties and underlying conceptual ambiguities need to be resolved. 相似文献

Baseline concentration levels of As, B, Ca, Cd, Cr, Cu, Co, Fe, Mg, Mn, Mo, Ni, P, Pb, Se, V, and Zn were determined for Porphyra columbina and Ulva sp. collected from three locations along San Jorge Gulf, in Patagonia Argentina. Elements were quantified by inductively coupled plasma-optical emission spectrometry, with the exception of lead and cadmium in some samples which were determined by electrothermal atomic absorption spectrometry. Three stations with different exposure degree to human activities, Bahía Solano, the mouth of Arroyo La Mata stream and Punta Maqueda, were selected as sampling points. The results showed a wide range of metal retention capacity between the two studied species. Regarding the levels of pollutants found in the researched sites, Punta Maqueda seemed to be less influenced by anthropogenic activities than the other two sites except for Cd. Taking into account their toxicities seasonal variations in Pb and Cd levels were studied in both algae in Punta Maqueda. Maximum concentrations of Cd (9.8 microg g(-1) dry wt.) were observed in P. columbina during winter, and maximum levels of Pb (0.82 microg g(-1) dry wt.) were detected in Ulva sp. during summer.
