Atopic dermatitis (AD) is a common relapsing inflammatory skin disorder characterized by immune-mediated inflammation and epidermal barrier dysfunction. The pathogenesis of AD is multifactorial and has not been fully elucidated to date. This study aimed to evaluate whether serum IgG from adult AD patients could modulate the thymic maturation of IL-22-producing T cells and CLA+ T cells of non-atopic infants. Given that miRNAs regulate immune response genes, we evaluated whether miRNA expression is also altered in cultured thymocytes. Thymocytes were cultured with purified IgG from AD patients or control conditions (mock, Intravenous-IgG (IVIg), non-atopic IgG, or atopic non-AD IgG). Using flow cytometry analysis, we assessed the expression of CLA and intracellular levels of IL-4, IFN-γ, and IL-22 on double-positive T cells (DP T), CD4 T cells, or CD8 T cells. We also investigated the frequency of IgG isotypes and their direct interaction with the thymic T cells membrane. The miRNA profiles were evaluated by the Illumina small RNA-seq approach. MiRNA target gene prediction and enrichment analyses were performed using bioinformatics. Increased frequencies of IL-22 and CLA+ producing CD4+ T cells cultured with IgG of AD patients was seen in non-atopic infant thymocytes compared to all control conditions. No alterations were observed in the frequency of IgG isotypes among evaluated IgG pools. Evidence for a direct interaction between IgG and thymic DP T, CD4 T, and CD8 T cells is presented. The small RNA-seq analysis identified ten mature miRNAs that were modulated by AD IgG compared to mock condition (miR-181b-5p, hsa-miR-130b-3p, hsa-miR-26a-5p, hsa-miR-4497, has-miR-146a, hsa-let-7i-5p, hsa-miR-342-3p, has-miR-148a-3p, has-miR-92a and has-miR-4492). The prediction of the targetome of the seven dysregulated miRNAs between AD and mock control revealed 122 putative targets, and functional and pathway enrichment analyses were performed. Our results enhance our understanding of the mechanism by which IgG can collaborate in thymic T cells in the setting of infant AD.  相似文献

Amphiphilic copolymers are appealing materials because of their interesting architecture and tunable properties. In view of their application in the biomedical field, the preparation of these materials should avoid the use of toxic compounds as catalysts. Therefore, enzymatic catalysis is a suitable alternative to common synthetic routes. Pentablock copolymers (CUC) were synthesized with high yields by ring-opening polymerization of ε-caprolactone (ε-CL) initiated by Pluronic (EPE) and catalyzed by Candida antarctica lipase B enzyme. The variables to study the structure–property relationship were EPEs’ molecular weight and molar ratios between ε-CL monomer and EPE macro-initiator (M/In). The obtained copolymers were chemically characterized, the molecular weight determined, and morphologies evaluated. The results suggest an interaction between the reaction time and M/In variables. There was a correlation between the differential scanning calorimetry data with those of X-ray diffraction (WAXD). The length of the central block of CUC copolymers may have an important role in the crystal formation. WAXD analyses indicated that a micro-phase separation takes place in all the prepared copolymers. Preliminary cytotoxicity experiments on the extracts of the polymer confirmed that these materials are nontoxic.  相似文献

Soy protein fractions rich in β-conglycinin (7S) or glycinin (11S) were freeze dried or spray dried at temperatures of 120, 150 or 180 °C. The fractions were characterized for their particle size distribution, sorption isotherms and by scanning differential calorimetry. The gelling capacity of the protein fractions was studied at pH values of 3 and 7 using oscillatory measurements, mechanical properties and water holding capacity. The rheological measurements showed that viscous modulus (G″) predominated at low temperatures and the elastic modulus (G′) at high temperatures. At pH 3, the G′–G″ crossover occurred at lower temperatures when compared to pH 7. This behaviour was more accentuated for the 11S fractions due to its capacity to form stronger gels. An increase of drying temperature led to a displacement of the gel point to higher temperatures and decreased the elasticity modulus or gelling capacity of protein fractions. These results were confirmed by the mechanical properties, since at higher temperatures the gels were more fragile and brittle, especially when formed at pH 7.  相似文献

The efficiency of different surface pretreatments (four standard chemical etchings and four diamond powder abrasive procedures) on silicon nitride (Si3N4) substrates for chemical vapor deposition (CVD) of diamond has been systematically investigated. Blank Si3N4 samples were polished with colloidal silica (∼0.25 μm). Diamond nucleation and growth runs were conducted in a microwave plasma chemical vapor deposition apparatus for 10 min and 6 h, respectively. Superior results concerning nucleation density ( N d∼ 1010 cm−2 after 10 min), film uniformity, and grain size (below 2 μm after 6 h) were obtained for the mechanically microflawed samples, revealing that chemical etchings (hot and cold strong acids, molten base or CF4 plasma) are not crucial for good CVD diamond quality on Si3N4.  相似文献

The use of microcapsules has increased in several different areas, namely, textile applications. They have been used as a possible means of introducing new properties, namely, in medical care by antibiotics, skin moisturizers, and other drugs and for thermal comfort. In this study, we examined the influence of dielectric barrier discharge (DBD) plasma treatment on the adhesion of phase‐change material (PCM) microcapsules on wool fabric. Several experimental techniques were used to evaluate the wool surface modification after plasma treatment and the influence of the microcapsules' resistance to washing conditions, namely, the determination of the static and dynamic contact angles, surface energy, and adhesion work; X‐ray photoelectron spectroscopy; Fourier transform infrared spectroscopy; differential scanning calorimetry; and scanning electron microscopy. Chemical and physical characterization of the wool fiber in the fabric confirmed significant surface modification. The plasma treatment greatly increased the hydrophilicity, surface energy, and adhesion work of the wool fabric; this proved that more microcapsules were adsorbed on the fabric and more microcapsules remained on the fabric surface after the washing procedures. © 2012 Wiley Periodicals, Inc. In this work, the morphology and linear viscoelastic behavior of PMMA/PP blends to which a graft copolymer PP‐g‐PMMA has been added was studied. The copolymer concentration varied from 1 to 10 wt % relative to the dispersed phase concentration. The rheological data were used to infer the interfacial tension between the blended components. It was observed that PP‐g‐PMMA was effective as a compatibilizer for PMMA/PP blends. For PP‐g‐PMMA concentration added below the critical concentration of interface saturation, two rheological behaviors were observed depending on the blend concentration: for 70/30 blend, the storage modulus, at low frequencies, increased as compared to the one of the unmodified blend; for 90/10 blend, it decreased. For 90/10 blend, the relaxation spectrum presented an interfacial relaxation time related to the presence of the compatibilizer (τβ). For PP‐g‐PMMA concentrations added above the critical concentration of interface saturation, the storage modulus of all blends increased as compared with the one of the unmodified blend. © 2012 Wiley Periodicals, Inc. This study presents a nanocomposite manufacturing route starting from non‐functionalized nanosilica and SAN surface modification via “grafting from” ATRP up to processing of transparent polycarbonate and glycol modified poly(ethylene terephthalate) nanocomposites. Synthesis limitations of low molecular weight graft SAN derived from “classical” ATRP are overcome by employment of the emerging ARGET ATRP. Mechanical investigations of polycarbonate and glycol modified poly(ethylene terephthalate) nanocomposites present up to 80% enhancement of creep performance and additionally 70% enhancement against wear abrasion for glycol modified poly(ethylene terephthalate) nanocomposites. Strength and tensile modulus are only moderately influenced by the addition of nanoparticles. Morphological studies confirm that the present modification route is able to create uniform dispersions of single particles and small particle aggregates in polymer matrices.

ABSTRACT

This paper presents a simulation study of air flow inside dryers. An adaptive finite element methodology for two-dimensional problems was developed. This study is the first application of nadaptive techniques to such tupe of problems. The formultion is generic and can be easily extended to three-dimensional analysis. Several dryer configurations were studied in order to verify the efficiency and accuracy of the proposed methodology. Results agreed well with experimental data and show significant imporvements when compared to the conventional finite elements analysis.  相似文献
