The seed oil of Carapa guianensis Aublet (Andiroba) has been used in folk medicine for its insect-repelling, anti-inflammatory, and anti-malarial activities. This study aimed to examine the triglyceride (TG) reducing effects of C. guianensis-derived limonoids or other commercially available limonoids in human hepatoblastoma HepG2 cells and evaluate the expression of lipid metabolism or autophagy-related proteins by treatment with 7-deacetoxy-7-oxogedunin (DAOG; 1), a principal limonoid of C. guianensis. The gedunin-type limonoids, such as DAOG (% of control at 20 μM: 70.9 ± 0.9%), gedunin (2, 74.0 ± 1.1%), epoxyazadiradione (4, 73.4 ± 2.0%), 17β-hydroxyazadiradione (5, 79.9 ± 0.6%), 7-deacetoxy-7α-hydroxygedunin (6, 61.0 ± 1.2%), andirolide H (7, 87.4 ± 2.2%), and 6α-hydroxygedunin (8, 84.5 ± 1.1%), were observed to reduce the TG content at lower concentrations than berberine chloride (BBR, a positive control, 84.1 ± 0.3% at 30 μM) in HepG2 cells pretreated with high glucose and oleic acid. Andirobin-, obacunol-, nimbin-, and salannin-type limonoids showed no effect on the intracellular TG content in HepG2 cells. The TG-reducing effect of DAOG was attenuated by the concomitant use of compound C (dorsomorphin), an AMPK inhibitor. Further investigation on the detailed mechanism of action of DAOG at non-cytotoxic concentrations revealed that the expressions of autophagy-related proteins, LC3 and p62, were upregulated by treatment with DAOG. These findings suggested that gedunin-type limonoids from Andiroba could ameliorate fatty liver, and that the action of DAOG in particular is mediated by autophagy.  相似文献

There has been an increasing prevalence of neurodegenerative diseases with the rapid increase in aging societies worldwide. Biomarkers that can be used to detect pathological changes before the development of severe neuronal loss and consequently facilitate early intervention with disease-modifying therapeutic modalities are therefore urgently needed. Diffusion magnetic resonance imaging (MRI) is a promising tool that can be used to infer microstructural characteristics of the brain, such as microstructural integrity and complexity, as well as axonal density, order, and myelination, through the utilization of water molecules that are diffused within the tissue, with displacement at the micron scale. Diffusion tensor imaging is the most commonly used diffusion MRI technique to assess the pathophysiology of neurodegenerative diseases. However, diffusion tensor imaging has several limitations, and new technologies, including neurite orientation dispersion and density imaging, diffusion kurtosis imaging, and free-water imaging, have been recently developed as approaches to overcome these constraints. This review provides an overview of these technologies and their potential as biomarkers for the early diagnosis and disease progression of major neurodegenerative diseases.  相似文献

Porous alumina whose pores were aligned in one direction was fabricated by the unidirectional solidification method under a pressurized hydrogen atmosphere. The porous structure is formed at the solid–liquid interface during solidification due to a hydrogen solubility gap at the melting point. The hydrogen gas is dissolved into molten alumina according to Sieverts' law and insoluble gas that corresponds to the amount of solubility gap evolves from the solid phase at the solid–liquid interface during the unidirectional solidification to form the pores. The porosity and pore size of the solidified samples decreased with increasing total pressure where the environmental gas consisted of pure hydrogen or hydrogen–argon mixed gases. There is a reverse proportion relation between the pore diameter and the total pressure according to Boyle's law.  相似文献

To elucidate the insolubilization mechanism of iodinated polystyrene (IPS) upon exposure to an electron beam and deep UV, the effects of additives on exposure characteristics of IPS and its parent polymer, polystyrene, are clarified. Results indicate the following mechanism. Iodine atoms produced by homolytic dissociation upon exposure undergo competitive reactions: the radical recombination which is the reverse of dissociation and the formation of other radicals that leads to insolubilization. The proposed reaction mechanism can explain the relationship between the sensitivity and degree of iodination. The G values for crosslinking of IPS, G(X), are also obtained according to Charlesby's theory. The calculated lithographic characteristics on the basis of G(X) are discussed.  相似文献

Mining frequent itemsets from large databases has played an essential role in many data mining tasks. It is also important to maintain the discovered frequent itemsets for these data mining tasks when the database is updated. All algorithms proposed so far for the maintenance of discovered frequent itemsets are only performed with a fixed minimum support,which is the same as that used to obtain the discovered frequent itemsets. That is, users cannot change the minimum support even if the new results are unsatisfactory to the users. In this paper two new complementary algorithms, FMP (First Maintaining Process) and RMP (Repeated Maintaining Process), are proposed to maintain discovered frequent itemsets in the case that new transaction data are added to a transaction database. Both algorithms allow users to change the minimum support for the maintenance processes. FMP is used for the first maintaining process, and when the result derived from the FMP is unsatisfactory, RMP will be performed repeatedly until satisfactory results are obtained. The proposed algorithms re-use the previous results to cut down the cost of maintenance. Extensive experiments have been conducted to assess the performance of the algorithms. The experimental results show that the proposed algorithms are very resultful compared with the previous mining and maintenance algorithms for maintenance of discovered frequent itemsets.  相似文献

The performance of an indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) based on a monoclonal antibody (mAb) for ochratoxin A (OTA) detection was evaluated in a comparative study with high-performance liquid chromatography (HPLC) analysis using 68 freshly harvested coffee samples from the North of Paraná State, Brazil. The anti-OTA mAb showed high specificity and low cross-reactivity with OTA analogues (OTB and OTalpha), but cross-reacted with OTC. This ic-ELISA showed a detection limit of 3.75 ngg-1 sample, when compared to 0.80 ngg-1 by HPLC, with an ic-ELISA/HPLC correlation coefficient of 0.90. As regards OTA analysis of these coffee samples, natural contamination was detected in 10 samples (14.7%) by both methods, where the ic-ELISA values (range 3.9-7.3 ngg-1) were 1.1 to 1.6-fold higher than HPLC data (2.7-4.7 ngg-1). Five samples (7.4%) were OTA positive (range 0.84-1.30 ngg-1) only by HPLC assay, probably due to the higher detection limit reached by ic-ELISA. OTA was undetectable in 53 samples (77.9%) by both methods, while all positive samples (range 0.84-7.30 ngg-1) showed OTA levels lower than 8 ngg-1 (maximum limit recommended by the European Union). The matrix interference of green coffee was minimized by dilution of sample extracts before carrying out the ELISA assay. This mAb-based ic-ELISA can be effectively applied for OTA screening in coffee, because it is simple, sensitive and sample preparation is easy.  相似文献

We have performed scanning tunneling microscopy/spectroscopy (STM/STS) on single crystals of heavily Pb-doped Bi 2 Sr 2 CaCu 2 O y (Pb-Bi2212). The obtained STM images clearly demonstrated phase separation into the and phases with lamella-type and new zigzag-like microstructures. From the site-specified tunneling measurements, it was directly confirmed that the phase possesses higher T c . This suggests that the phase boundaries act as strong pinning sites especially at higher temperatures.  相似文献
