We have shown previously that the diphtheria toxin transmembranedomain (T) may function as a membrane anchor for soluble proteinsfused at its C-terminus. Binding to membranes is triggered byacidic pH. Here, we further characterized this anchoring device.Soluble proteins may be fused at the N-terminus of the T domainor at both extremities, without modifying its membrane bindingproperties. This allows one to choose the orientation of theprotein to be attached to the membrane. Maximum binding to thecell surface is reached within 1 h. Anchoring occurs on cellspreviously treated with proteinase K, suggesting that T interactswith the lipid phase of the membrane without the help of cellsurface proteins. Binding does not permeabilize cells or affectcell viability, despite the fact that it permeabilizes liposomesand alters their structure. When attached to L929 fibroblasts,the proteins are not internalized and remain displayed at theirsurface for more than 24 h. When bound to K562 myeloid cells,the molecules are internalized and degraded. Thus, dependingon the cell type, soluble proteins may be anchored to the surfaceof cells by the T domain for an extended time or directed towardsan internalization pathway.  相似文献

Anaerobic digestion enables waste (water) treatment and energy production in the form of biogas. The successful implementation of this process has lead to an increasing interest worldwide. However, anaerobic digestion is a complex biological process, where hundreds of microbial populations are involved, and whose start-up and operation are delicate issues. In order to better understand the process dynamics and to optimize the operating conditions, the availability of dynamic models is of paramount importance. Such models have to be inferred from prior knowledge and experimental data collected from real plants. Modeling and parameter identification are vast subjects, offering a realm of approaches and methods, which can be difficult to fully understand by scientists and engineers dedicated to the plant operation and improvements. This review article discusses existing modeling frameworks and methodologies for parameter estimation and model validation in the field of anaerobic digestion processes. The point of view is pragmatic, intentionally focusing on simple but efficient methods.  相似文献

This paper presents a fuzzy tuning system for real-time industrial PID (proportional-integral-derivative) controllers. The algorithm set the proportional gain, integral time and derivative time of a classical PID structure according to the set point, error and error derivative of the process, respectively. The tuning of the PID controller is based on a fuzzy inference machine. The set of rules of the fuzzy inference machine was obtained by experts engineering. The system is tested in an austempering process but can be applied in any industrial plant. Besides, an analysis between the response of the process with a PID controller and the system of fuzzy auto-tuning for P1D proposed was made.  相似文献

A new protocol is implemented to demonstrate the presence of blood in the patina of African art objects from Mali. Divided into three steps, the protocol first consists in demonstrating the presence of proteins and localizing them in the sample's cross sections using time-of-flight secondary ion mass spectrometry (TOF-SIMS) and synchrotron-based infrared microspectrometry (microFT-IR). In a second time, TOF-SIMS is used to investigate heme, which is a blood marker. If heme is missing, which could mean that it is too degraded to be detected, X-ray microfluorescence (microXRF) and X-ray absorption near-edge microspectroscopy (microXANES) are used to prove the presence of iron in the protein area and to get a fingerprint of its chemical environment. This permits us thus to demonstrate that iron is indeed linked with proteins and not with mineral phases of the sample. Coupled with the ritual context of the objects, this constitutes a proof of the use of blood. Thanks to this protocol, which has the major advantage of avoiding false positive results, the presence of blood has been demonstrated in seven out of the eight studied samples.  相似文献

A 13C n.m.r. study of the microstructure of ethyl vinyl ether (EVE) and isopropyl vinyl ether (IPVE) polymers prepared under various experimental conditions and using chemical or γ-ray initiation has been made. Long stereo-sequence assignments were conducted for poly EVE allowing determination of the configurational statistic of propagation in the radiation-induced polymerization of EVE in low polar solvents. Stereo-sequence intensities are found in good agreement with the Markovian model. The results are considered as further support for the occurence of a specific interaction between the growing active centres and the polymer chains. In the case of IPVE polymers much more important overlappings of the resonance patterns are observed thus preventing any similar detailed study.  相似文献

We report a novel and very handful strategy for the functionalization of diamond nanoparticles (NDs), based on the ultrasound-assisted grafting of aryl groups from the electroless reduction of diazonium salts. For this study, 4-nitrobenzenediazonium salt was used as a model molecule and the reaction was investigated in neutral and acidic aqueous media. Spectroscopic evidence for the successful attachment of aryl groups to nanodiamonds (NDs) was given by IR and XPS which clearly detect characteristic NO2 peaks. Moreover, the absence of any peaks from the +N≡N group in the IR spectra is a clear indication of the chemical reduction of the parent diazonium salt at the surface of NDs. This spontaneous chemical modification of NDs by aryl diazonium salts was confined to the surface of the ND particles; indeed, XRD measurements have shown that the crystalline structure of the bulk of the particles was unaffected. It opens up new possibilities towards the control of the surface chemical composition of NDs using simple protocols operated in very soft conditions, i.e. in water at room temperature. It shows conclusively that the chemistry toolbox of experts interested in nanodiamonds should contain aryl diazonium salts, given their versatility in forming active platforms.  相似文献

Abstract— Aviation increasingly conducts night operations using night‐vision devices. However, design of some aspects may limit performance. As pointed out by Kotulak, vergence/accommodation mismatch in NVDs, usually due to eyepiece focus misadjustments, is sometimes a source of visual acuity (VA) decrement. The increased separation between sensors, existing in some binocular helmetmounted displays, was also identified to be potentially responsible for decreased VA at short distances. Based upon knowledge pertaining to vergence and accommodation, a study was performed to better understand the problem of dissociation accommodation/convergence. Different conditions of interocular separation and viewing distances were used. Twelve subjects participated and were asked to resolve Landolt C charts using night‐vision goggles. The results show that, with the eyepiece focus fixed at 10 m, the decrease in VA is roughly proportional to the interocular separation, when looking at short distances. A fixed eyepiece focus at 4 m considerably reduces the conflict and results in improved VA. An additional experiment was conducted to investigate the effect of fixing the objective lenses focus at infinity. With this setting, the decrease in VA at a short distance was such that effects of the mismatch accommodation/convergence are no longer apparent regardless of the interocular separation.  相似文献

Two metabolically engineered Escherichia coli strains have been constructed to produce the carbohydrate moieties of gangliosides GM2 (GalNAcbeta-4(NeuAcalpha-3)Galbeta-4Glc; Gal = galactose, Glc = glucose, Ac = acetyl) and GM1 (Galbeta-3GalNAcbeta-4(NeuAcalpha-3)Galbeta-4Glc. The GM2 oligosaccharide-producing strain TA02 was devoid of both beta-galactosidase and sialic acid aldolase activities and overexpressed the genes for CMP-NeuAc synthase (CMP = cytidine monophosphate), alpha-2,3-sialyltransferase, UDP-GlcNAc (UDP = uridine diphosphate) C4 epimerase, and beta-1,4-GalNAc transferase. When this strain was cultivated on glycerol, exogenously added lactose and sialic acid were shown to be actively internalized into the cytoplasm and converted into GM2 oligosaccharide. The in vivo synthesis of GM1 oligosaccharide was achieved by taking a similar approach but using strain TA05, which additionally overexpressed the gene for beta-1,3-galactosyltransferase. In high-cell-density cultures, the production yields for the GM2 and GM1 oligosaccharides were 1.25 g L(-1) and 0.89 g L(-1), respectively.  相似文献

Very long aging times, up to 15,100 h (629 days) at 110°C, were achieved on flame‐retardant printed circuit board laminates commonly used in automotive design. This composite was fabricated from glass fibers embedded in an epoxy resin. Aging was performed in an oven under an air atmosphere at a temperature lower than the glass‐transition temperature. Temperature‐modulated differential scanning calorimetric analysis was used to investigate the influence of such aging on the glass‐transition phenomena. A new amorphous phase appeared during aging. By extending the analysis to samples collected at different thicknesses, we demonstrated the existence of a time‐dependent gradient of the properties. A skin–core structure was evidenced, and this slowed down oxidation and allowed physical aging to occur in the bulk sample. GluN2B‐containing NMDA receptors are involved in many important physiological functions and play a pivotal role in mediating pain as well as in several neurodegenerative disorders. We aimed to develop fluorescent probes to target the GluN2B subunit selectively in order to allow better understanding of the relationships between receptor localisation and physiological importance. Ifenprodil, known as the GluNR2B antagonist of reference, was chosen as the template for the elaboration of probes. We had previously reported a fluorescein conjugate that was shown (by confocal microscopy imaging of DS‐red‐labelled cortical neurons) to bind specifically to GluN2B. To elaborate this probe, we explored the influence of both the nature and the attachment point of the spacer between the fluorophore and the parent compound, ifenprodil. We performed chemical modifications of ifenprodil at the benzylic position and on the phenol ring by introducing secondary amine or amide functions and evaluated alkyl chains from two to 20 bonds either including or not including secondary amide functions as spacers. The previously developed probe was found to display the greatest activity in the inhibition of NMDA‐induced Ca2+ influx by calcium imaging experiments on HEK293 cells transfected with the cDNA encoding for GluN1‐1A and GluN2B. Further investigations revealed that this probe had a neuroprotective effect equivalent to that of ifenprodil in a standard test for neurotoxicity. Despite effects of lesser amplitude with these probes relative to ifenprodil, we demonstrated that they displaced [3H]ifenprodil in mouse brain slices in a similar manner.  相似文献
