The precise role that nitric oxide (NO) plays in the mechanisms of ischemic brain damage remains to be established. The expression of the inducible isoform (iNOS) of NO synthase (NOS) has been demonstrated not only in blood and glial cells using in vivo models of brain ischemia-reperfusion but also in neurons in rat forebrain slices exposed to oxygen-glucose deprivation (OGD). We have used this experimental model to study the effect of OGD on the neuronal isoform of NOS (nNOS) and iNOS. In OGD-exposed rat forebrain slices, a decrease in the calcium-dependent NOS activity was found 180 min after the OGD period, which was parallel to the increase during this period in calcium-independent NOS activity. Both dexamethasone and cycloheximide, which completely inhibited the induction of the calcium-independent NOS activity, caused a 40-70% recovery in calcium-dependent NOS activity when compared with slices collected immediately after OGD. The NO scavenger oxyhemoglobin produced complete recovery of calcium-dependent NOS activity, suggesting that NO formed after OGD is responsible for this down-regulation. Consistently, exposure to the NO donor (Z)-1-[(2-aminoethyl)-N-(2-ammonioethyl)amino]diazen-1-iu m-1,2-diolate (DETA-NONOate) for 180 min caused a decrease in the calcium-dependent NOS activity present in control rat forebrain slices. Furthermore, OGD and DETA-NONOate caused a decrease in level of both nNOS mRNA and protein. In summary, our results indicate that iNOS expression down-regulates nNOS activity in rat brain slices exposed to OGD. These studies suggest important and complex interactions between NOS isoforms, the elucidation of which may provide further insights into the physiological and pathophysiological events that occur during and after cerebral ischemia.  相似文献

The thermoluminescent behaviour of diamond films subjected to UV irradiation was studied by using an UV lamp of 254 nm wavelength. The UV irradiation was achieved by placing the samples 15 cm away from an UV source for different periods. The thermoluminescent signal was integrated from 0 to 350°C at a linear heating rate of 10°C/s in a N2 atmosphere. The corresponding luminescence spectra show an excitation band centered at 450 nm while the emission band is centered around 500 nm at room temperature. The diamond films were synthesized on molybdenum substrates by the combustion flame technique and characterized by Raman spectroscopy and scanning electron microscopy. Breakdown rates and colonisation of leaves from four tree or scrub species differing in quality are studied upstream and downstream of the Canales reservoir, a dam located in the headwater of the River Genil, Sierra Nevada, in southern Spain. This dam, with hypolimnetic release, displays short‐term fluctuations of discharge and nutrient enrichment in the tailwater during the study period. Breakdown rates of the four leaf species studied do not differ between sites, despite the higher dissolved nutrient concentration in the tailwater. This lack of differences is attributed to the potentially high physical breakage of leaves during peak flows that are of higher magnitude at the upstream site. The invertebrate density in leaf bags does not differ between sites, and Chironomidae and Ephemeroptera are the numerically dominant taxa at both sites. With regard to functional feeding groups, the scarcity and lack of significant differences between sites for shredders do not match the trend predicted by the Serial Discontinuity Concept in relation to the effect of a headwater dam. Possibly, the discharge fluctuations at both sites causes excessive instability of the natural substrate (leaf litter) for the shredder guild. However, as expected, the biomass of collectors colonising leaf bags is significantly higher at the tailwater, which might be explained not in terms of quantity, but as a consequence of the higher nutritional quality of the fine particulate organic matter (FPOM) accumulated in leaf bags at this site, owing to the eutrophication caused by the dam. Despite the scarcity of functional shredders at both sites, at the community level, the leaf material is significantly more ingested at the upstream site, suggesting the importance of this source of nutrition for the trophic web at this site in contrast with the tailwater, as predicted by the Serial Discontinuity Concept. This also suggests that caution is needed in using functional feeding groups as trophic guilds to infer system‐level trophic dynamics in streams, given the prevalence of generalist feeders among benthic macroinvertebrates in these environments. The thermal-wave radar method was used to resolve and measure deep hardness case depths (2-3 mm) of AISI9310 and Pyrowear53 steels used in the aerospace industry. Several gears made of these steels were evaluated and correlations between radiometric signal and case depths were obtained using the cross-correlation peak delay time. The foregoing case depth range is significantly deeper than achievable with point-by-point laser frequency scans (800 μm) and swept-sine modulation of the laser beam (1.16 mm).  相似文献

Solving optimization problems using a reduced number of objective function evaluations is an open issue in the design of multi‐objective optimization metaheuristics. The usual approach to analyze the behavior of such techniques is to choose a benchmark of known problems, to perform a predetermined number of function evaluations, and then, apply a set of performance indicators in order to assess the quality of the solutions obtained. However, this sort of methodology does not provide any insights of the efficiency of each algorithm. Here, efficiency is defined as the effort required by a multi‐objective metaheuristic to obtain a set of non‐dominated solutions that is satisfactory to the user, according to some pre‐defined criterion. Indeed, the type of solutions of interest to the user may vary depending on the specific characteristics of the problem being solved. In this paper, the convergence speed of seven state‐of‐the‐art multi‐objective metaheuristics is analyzed, according to three pre‐defined efficiency criteria. Our empirical study shows that SMPSO (based on a particle swarm optimizer) is found to be the best overall algorithm on the test problems adopted when considering the three efficiency criteria. Several studies have suggested that the phenolic fraction plays an important role during storage and therefore in the shelf life of virgin olive oil. This investigation examines the effect of freezing olives (–18 °C) before processing into oil on the transfer of the phenolic compounds into the subsequent oil, and the consequential changes in oxidative stability. Oil samples obtained from frozen olives (24 h at –18 °C), crushed with and without preliminary thawing, were compared to a control sample; both oils were obtained using a two‐phase low‐scale mill. The oxidative stability in different samples was assessed in terms of primary and secondary oxidation products as measured by peroxide values and oxidative stability index times, respectively. The quality of the oil samples was also checked through the percentage of free acidity and the phenolic content. Phenols were determined by both spectrophotometric assays (total phenols and o‐diphenols) and HPLC‐DAD/MSD. The antiradical capacity of the phenolic fraction was determined by DPPH and ABTS spectrophotometric tests. These analyses showed that thawing of olives before oil extraction led to a significant loss of oxidative stability and phenols; in contrast, samples obtained from frozen olives that were not thawed before crushing showed qualitative characteristics similar to control samples.  相似文献
