Thermomonas hydrothermalis is a Gram‐negative thermophilic bacterium that is able to live at 50 °C. This ability is attributed to chemical modifications, involving those to bacterial cell‐wall components, such as proteins and (glyco)lipids. As the main component of the outer membrane of Gram‐negative bacteria, lipopolysaccharides (LPSs) are exposed to the environment, thus they can undergo structural chemical changes to allow thermophilic bacteria to live at their optimal growth temperature. Furthermore, as one of the major target of the eukaryotic innate immune system, LPS elicits host immune response in a structure‐dependent mode; thus the uncommon chemical features of thermophilic bacterial LPSs might exert a different biological action on the innate immune system—an antagonistic effect, as shown in studies of LPS structure–activity relationship in the ongoing research into antagonist LPS candidates. Here, we report the complete structural and biological activity analysis of the lipo‐oligosaccharide isolated from Thermomonas hydrothermalis, achieved by a multidisciplinary approach (chemical analysis, NMR, MALDI MS and cellular immunology). We demonstrate a tricky and interesting structure combined with a very interesting effect on human innate immunity.  相似文献

Smouldering combustion in burning piles was experimentally investigated by studying temperature changes in six piles of 2 m of diameter of cutting debris of hazel (Corylus avellana) for three days after extinction. The piles were monitored using an IR camera and K-type thermocouples. The experiment was designed in order to study how the maximum temperature of the charcoal might be influenced by the individual and interaction effects of both the quality of extinction and the elapsed time until the start of extinction of the piles. The piles that were properly extinguished (i.e. using a high-pressure, homogeneously distributed water flow of 50 l/min for 4 min) had a uniform temperature profile and did not have significant hot spots. The temperature reached equilibrium with the environment in less than 10 h after extinction. In contrast, a smouldering front moved throughout the poorly extinguished piles, which had a wide temperature distribution and hot spots of up to 700 °C. A simulation of windy conditions after three days of experiments on a poorly extinguished pile showed that the reactivation of charcoal combustion was possible. It gave a high-risk scenario to cause a wildfire, with hot smouldering embers being transported by wind flow. The results are of interest to improve pile burning protocols so that the number of wildfires caused by such practices may be reduced.  相似文献

Solving optimization problems using a reduced number of objective function evaluations is an open issue in the design of multi‐objective optimization metaheuristics. The usual approach to analyze the behavior of such techniques is to choose a benchmark of known problems, to perform a predetermined number of function evaluations, and then, apply a set of performance indicators in order to assess the quality of the solutions obtained. However, this sort of methodology does not provide any insights of the efficiency of each algorithm. Here, efficiency is defined as the effort required by a multi‐objective metaheuristic to obtain a set of non‐dominated solutions that is satisfactory to the user, according to some pre‐defined criterion. Indeed, the type of solutions of interest to the user may vary depending on the specific characteristics of the problem being solved. In this paper, the convergence speed of seven state‐of‐the‐art multi‐objective metaheuristics is analyzed, according to three pre‐defined efficiency criteria. Our empirical study shows that SMPSO (based on a particle swarm optimizer) is found to be the best overall algorithm on the test problems adopted when considering the three efficiency criteria. Carbon dioxide (CO2) adsorption is an important adsorbent characterization method and a significant industrial process. In separation and recovery technology, the adsorption of the CO2 is important to reduce the concentration of this gas considered as one of the greenhouse gases. Natural zeolites, particularly clinoptilolite, are widely applied as adsorbents. In this regard, in the present research, the structure, composition and morphology of modified with hexafluorosilicate (HFSi) and orthophosphoric acid (H3PO4) clinoptilolites were investigated by characterizations and measurements made with, X-ray diffraction (XRD), thermogravimetric analysis (TGA), scanning electron microscopy-energy dispersive X-ray analysis (SEM-EDAX) and gravimetric adsorption. Additionally, the surface Chemistry of the modified clinoptilolites was analyzed by applying diffuse reflectance fourier transform infrared spectrometry (DRIFTS). Further, the interaction of CO2 within the adsorption space of these modified clinoptilolites and a synthetic ZSM-5 zeolite was studied with the help of adsorption measurements. After all, an appropriate theoretical methodology for the analysis of the XRD and adsorption data was applied. The calculated cell parameters of the tested are similar to those reported for a typical clinoptilolite of: a = 17.662 A, b = 17.911A, c = 7.407 ~ and fl = 116.40 The resolution of the TGA derivative profiles indicated the presence of two steps for water release, one of them represents the loss of majority of the water present in the micropores. This was evidenced as a broad peak centered at about 50℃ for the CSW-HFSi-0.1, but at 100 ℃ for the samples CSW-HFSi-0.4. The SEM micrographs corresponding to the modified clinoptilolites, was evidenced that the CSW zeolite shows secondary particles exhibiting diameters from 3 to 40 μm, formed by primary clinoptilolite crystallites showing a crystallite size, φ = 40 nm. The EDAX elemental analysis it can be demonstrated that the exchange process replaced about 85% of the charge compensating ions. The DRIFT spectra of the modified clinoptilolites, specifically, CSW-HFSi-0.1, show a narrow band at about: 3,740 cm-1 corresponding to terminal silanol groups (Si-OH) and a band 3,600-3,650 cm1 resulting from extra-framework AI-OH. With the precision of the measured micropore volumes related to the excellent fitting of the adsorption data by the D-R isotherm equation, it can be affirm that carbon adsorption took only place in the micropore region. The isosteric heat of adsorption calculated for the modified clinoptilolites was greater than those values reported of ZSM-5 zeolite, particle packing silica, dealuminated Y zeolite (DAY) Cd, Zn and Ni-nitroprussides and Cu-nitroprusside and a Ni-MOF. With the obtained result it can be concluded that the modified clinoptilolites with HFSi showed a quality as adsorbent comparable to commercial synthetic zeolites.  相似文献

Poly (2-ethyl-2-oxazoline) (PEOZ) is one of the commercial members of a family of materials that have shown significant application potential in a large number of technological contexts, most of them related with biomedical problems where water-soluble polymer systems are highly desirable. Polymeric fibers with diameters in the 200–800 nm range of PEOZ were prepared by electrospinning of its water solutions. Processing and solution parameters effects on the morphology and the diameter of the fibers were investigated. SEM results showed that the polymer concentration and the applied voltage might be used as variables to control the morphology and the diameter of the electrospun fibers. Solutions of the same polymer in other two organic solvents (N,N-dimethylformamide (DMF) and tetrahydrofurane (THF)) were also processed by the same technique but without the promising results of the water solutions.  相似文献

The ubiquitous presence of alkylphenol ethoxylates in the environment as well as concern for endocrine disruption effects in biota caused by their degradation products (such as octyl- and nonylphenol) has raised interest in the environmental fate of these compounds. As part of an effort to model their behavior in a subestuary of the Chesapeake Bay, a quantitative method for the analysis of octyl- and nonylphenol, and their ethoxylates (1-5) in water and sediment was developed. Extraction procedures are based on solid-phase extraction techniques. Identification and quantitation of the analytes is done by liquid chromatography coupled to tandem mass spectrometry. Instrument detection limits for the compounds ranged from 0.1 to 9 pg injected on column, which allowed method detection limits of 0.04-3 ng/L in water and 0.2-13 ng/g of dry weight in sediment. The method was used to analyze water and sediment from the Back River, MD, where concentrations for the individual compounds ranged from <8 to 200 ng/L in water and <9 to 6700 ng/g of dry weight in sediment. Additionally, structural information obtained in the mass spectrometer is presented that supports previous observations that nonylphenol and its ethoxylates are composed mainly of isomers with a tertiary alpha-carbon.  相似文献

The fluorogenic dye 2-methoxy-2,4-diphenyl-3(2H)-furanone (MDPF) has been used for the detection of total protein patterns on polyvinylidene difluoride (PVDF) membranes. Fluorescent staining of protein bands on membranes with this covalent dye is completed in 20 min. Wet membranes are translucent, allowing protein visualization by transillumination with ultraviolet light. The resulting images can be recorded using Polaroid film or a charge-coupled device camera. Electrophoretic bands containing 5-10 ng of protein can be detected on the MDPF-stained Western blot. When proteins are directly transferred to the membrane using a slot blotting device, as little as 0.5 ng of protein can be detected. Previous visualization of protein bands on sodium dodecyl sulfate-polyacrylamide gels with the noncovalent fluorescent dye Nile red (Alba et al., BioTechniques, 1996, 21, 625-626) does not interfere with further MDPF staining and fluorescent detection of these bands transferred to PVDF membranes. Thus, Nile red and MDPF staining can be performed sequentially, allowing the rapid monitoring of total protein patterns on both the electrophoretic gel and Western blot. Using the conditions described in this study, MDPF staining does not preclude further N-terminal microsequencing and immunodetection of specific bands with polyclonal antibodies.  相似文献

Photosynthetic microorganisms are expected to be useful to maintain an oxygenic atmosphere and to provide biomass for astronauts in the International Space Station as well as in future long-term space flights. However, fluxes of complex ionizing radiation of various intensities and energies make space an extreme environment for the microorganisms, affecting their photosynthetic efficiency. To automatically monitor the photosynthetic Photosystem II (PSII) activity of microorganisms under space conditions an optical biosensor, which utilizes chlorophyll fluorescence as biological transduction system, was built; the PSII activity was monitored by the biosensor during balloon flights at stratospheric altitudes of about 40 km. The effect of space stress on quantum yield of PSII varied among the tested species depending on the growth light conditions at which they were exposed during the flights.  相似文献

Exploratory Analytics is the process of analyzing data for the purpose of forming hypotheses. Patent data sets, because they are relatively large and diverse and because they consist of a mixture of structured and unstructured information present a formidable challenge and a great opportunity in applying exploratory analytics techniques. In this paper we describe methods we have implemented for effective exploratory analytics on patent data sets using an interactive approach and a web based software tool called SIMPLE. We use real-world case studies to demonstrate the effectiveness of our exploratory analytics approach in the discovery of useful information from the patent corpus.  相似文献
