The Coronavirus Disease (COVID-19) pandemic is demanding the rapid action of the authorities and scientific community in order to find new antimicrobial solutions that could inactivate the pathogen SARS-CoV-2 that causes this disease. Gram-positive bacteria contribute to severe pneumonia associated with COVID-19, and their resistance to antibiotics is exponentially increasing. In this regard, non-woven fabrics are currently used for the fabrication of infection prevention clothing such as face masks, caps, scrubs, shirts, trousers, disposable gowns, overalls, hoods, aprons and shoe covers as protective tools against viral and bacterial infections. However, these non-woven fabrics are made of materials that do not exhibit intrinsic antimicrobial activity. Thus, we have here developed non-woven fabrics with antimicrobial coatings of cranberry extracts capable of inactivating enveloped viruses such as SARS-CoV-2 and the bacteriophage phi 6 (about 99% of viral inactivation in 1 min of viral contact), and two multidrug-resistant bacteria: the methicillin-resistant Staphylococcus aureus and the methicillin-resistant Staphylococcus epidermidis. The morphology, thermal and mechanical properties of the produced filters were characterized by optical and electron microscopy, differential scanning calorimetry, thermogravimetry and dynamic mechanical thermal analysis. The non-toxicity of these advanced technologies was ensured using a Caenorhabditis elegans in vivo model. These results open up a new prevention path using natural and biodegradable compounds for the fabrication of infection prevention clothing in the current COVID-19 pandemic and microbial resistant era. 相似文献

The resistance of malaria parasites to available drugs continues to grow, and this makes the need for new antimalarial therapies pressing. Aminoacyl‐tRNA synthetases (ARSs) are essential enzymes and well‐established antibacterial targets and so constitute a promising set of targets for the development of new antimalarials. Despite their potential as drug targets, apicoplastic ARSs remain unexplored. We have characterized the lysylation system of Plasmodium falciparum, and designed, synthesized, and tested a set of inhibitors based on the structure of the natural substrate intermediate: lysyl‐adenylate. Here we demonstrate that selective inhibition of apicoplastic ARSs is feasible and describe new compounds that that specifically inhibit Plasmodium apicoplastic lysyl‐tRNA synthetase and show antimalarial activities in the micromolar range. 相似文献

Marine bacteria, which are often described as chemical gold, are considered an exceptional source of new therapeutics. Considerable research interest has been given to lipopolysaccharides (LPSs), the main components of the Gram-negative outer membrane. LPS and its lipid A portion from marine bacteria are known to exhibit a tricky chemistry that has been often associated with intriguing properties such as behaving as immune adjuvants or anti-sepsis molecules. In this scenario, we report the structural determination of the lipid A from three marine bacteria within the Cellulophaga genus, which showed to produce an extremely heterogenous blend of tetra- to hexa-acylated lipid A species, mostly carrying one phosphate and one D-mannose on the glucosamine disaccharide backbone. The ability of the three LPSs in activating TLR4 signaling revealed a weaker immunopotential by C. baltica NNO 15840T and C. tyrosinoxydans EM41T, while C. algicola ACAM 630T behaved as a more potent TLR4 activator. 相似文献

The present study evaluated the antiulcerogenic activity and mechanisms of the aqueous (AqF 100 mg/kg) and ethyl acetate (AcF 50 mg/kg) fractions from Indigofera truxillensis leaves. This dose was selected to assess its activity on ulcer healing and its action on gastric acid and mucus secretion, prostaglandin production and antioxidant enzyme activity (superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and glutathione reductase (GSH-Rd)). Gastric ulcer was induced by absolute ethanol. Antisecretory action, mucus and prostaglandin production, healing and antioxidant enzyme activities were evaluated for both fractions. AqF and AcF significantly inhibited the gastric mucosal damage caused by ethanol. This effect was statistically significant at 100 and 50 mg/kg compared with the vehicle. Neither fraction interfered with gastric secretion. AcF increased the PGE2 production, and both fractions increased mucus production. l-NAME did not alter the gastroprotection exerted by the fractions, but N-ethylmaleimide attenuated only AcF. In the ischemia/reperfusion model both fractions inhibited the mucosal damage. AcF increased SOD, GSH-Px and GSH-Rd activity, but AqF increased only SOD and GSH-Px. In the acetic acid-induced ulcer model AcF only accelerated ulcer healing. These results showed that Indigofera truxillensis acted as a gastroprotective agent, stimulating protective factors and antioxidants enzymes. 相似文献

Carbon fibre-reinforced silicon carbide composites (C-SiC) were fabricated combining, for the first time, a liquid infiltration process (LI) of a mesophase pitch doped with silicon carbide nanoparticles followed by reactive liquid silicon infiltration using Spark Plasma Sintering (SPS) technique. A graphitization step was applied in order to improve the effectiveness of the processing. Up to three different morphologies of SiC particles were identified with a noticeable influence on the preliminary oxidation tests carried out. The presence of SiC nanoparticles added to the carbon matrix affects the morphology of the SiC obtained by in situ reaction of silicon and carbon during the LI process by SPS and it leads to an improvement of the material oxidation resistance. The results show that SPS is a promising method to develop C-SiC composites in a short time and with a high efficiency in the liquid silicon infiltration process. 相似文献

A modified ferrous oxidation‐xylenol orange (FOX) method was adapted to measure lipid hydroperoxides (LHP) in lipid extracts from snack foods fried in vegetable oils. First, a methanol‐based FOX reaction medium was assayed, but this became turbid upon addition of the lipid extracts dissolved in ethanol. To avoid the precipitation of lipids, the polarity of the reaction medium was reduced by lowering its water content and by replacing the methanol as the basis of the medium for less polar solvents. Some of the solvents used instead of methanol yielded a lower FOX reaction response. Of the reaction media assayed, the one based on dichloromethane/ethanol (3:2, vol/vol) was not turbid at high lipid extract concentrations (assayed at up to 25 mg of lipid extract/mL reaction medium) and provided the same response level as the methanol‐based medium. Thus, this FOX method shows high sensitivity and is particularly useful for lipid extracts with low LHP content. This method was also successfully applied to edible oils. Solvents such as 2‐propanol, ethyl acetate and butanol were discarded, because they easily produce hydroperoxides, which interfere in the FOX reaction. Xylenol orange preparations from a number of suppliers were tested, and some differences affecting the sensitivity of the reaction were observed. 相似文献

Treatment of wastewater containing high phenol concentrations (up to 4,000 mg/l, 1,600 kg/ha.d) in laboratory-scale stabilisation ponds enriched with activated sludge was studied. Phenol was biodegraded efficiently, even when fed as the sole carbon source. At influent concentrations of 1,000, 1,300, 1,600, 1,900, 2,500, 3,000 and 4,000 mg/l of phenol (loading rates of 400, 520, 640, 760, 1,000, 1,200 and 1,600 kg phenol/ha.d), the phenol removal efficiencies were 92, 89, 81, 81, 76, 65 and 22%, respectively. At 4,000 mg/l of phenol, the enriched ponds were significantly inhibited. The maximum phenol removal rate observed was 780 kg/ha.d, which is 7.7 times higher than the maximum value reported for attached-growth waste stabilisation ponds. All along the experiments, the enriched ponds showed removal rates 1.8-20.5 times higher than the values observed in control pond (not enriched). The results suggest that enrichment is an effective method to increase xenobiotic removal rates of stabilisation ponds. 相似文献

A three‐dimensional model of a complex between HIV‐1 integrase (IN), viral DNA, and metal ions that we recently built was used as a target for a docking method (induced‐fit docking, IFD) that accurately predicts ligand binding modes and concomitant structural changes in the receptor. Six different well‐known integrase strand transfer inhibitors (INSTIs): L‐708,906, L‐731,988, S‐1360, L‐870,810, raltegravir, and elvitegravir were thus used as ligands for our docking simulations. The obtained IFD results are consistent with the mechanism of action proposed for this class of IN inhibitors, that is, metal chelating/binding agents. This study affords new insight into the possible mechanism of inhibition and binding conformations for INSTIs. The impact on our hypothesis of specific mutations associated with IN inhibitor resistance was also evaluated. All these findings might have implications for integrase‐directed HIV‐1 drug discovery efforts. 相似文献

This paper introduces a systematic approach for dealing with potential adverse environmental impacts at the pre-construction stage. The proposed methodology serves as an assessment tool for construction projects to measure the environmental performance of their construction activities. It also provides a consistent basis for comparisons and for future eco-labelling and environmental benchmarking among construction companies and construction sites. Within the methodological framework, nine categories of environmental aspects are proposed: atmospheric emissions; water emissions; waste generation; soil alteration; resource consumption; local issues; transport issues; effects on biodiversity; and incidents, accidents and potential emergency situations. The methodology includes 20 performance indicators developed with the help of a panel of experts. In order to avoid a typical shortcoming in environmental assessments methods, these environmental indicators, both direct and indirect, are always based on quantitative data available in the project documents. Significance limits for environmental aspects are also developed based on a statistical analysis of 55 new-start construction projects. Four case studies are provided to illustrate the practical use of the proposed methodology. 相似文献

The effectiveness of long-term retardants used in forest fire fighting has been studied at three different scales. In this paper, retardant effectiveness experiments conducted since 1965 are examined and reviewed. These experiments include studies carried out with analytical instrumentation as well as studies involving laboratory and field flame-spread tests. Special attention is paid to the work performed with diammonium phosphate (DAP) and ammonium sulfate (AS). The experimental methodologies and the results obtained are reviewed in detail. A lack of effort in the modeling of fire behavior under the effect of these products is evidenced and recommendations for further work are made. 相似文献
