Control of interactions between nanomaterials and cells remains a biomedical challenge. A strategy is proposed to modulate the intralysosomal distribution of nanoparticles through the design of 3D suprastructures built by hydrophilic nanocrystals (NCs) coated with alkyl chains. The intracellular fate of two water‐dispersible architectures of self‐assembled hydrophobic magnetic NCs: hollow deformable shells (colloidosomes) or solid fcc particles (supraballs) is compared. These two self‐assemblies display increased cellular uptake by tumor cells compared to dispersions of the water‐soluble NC building blocks. Moreover, the self‐assembly structures increase the NCs density in lysosomes and close to the lysosome membrane. Importantly, the structural organization of NCs in colloidosomes and supraballs are maintained in lysosomes up to 8 days after internalization, whereas initially dispersed hydrophilic NCs are randomly aggregated. Supraballs and colloidosomes are differently sensed by cells due to their different architectures and mechanical properties. Flexible and soft colloidosomes deform and spread along the biological membranes. In contrast, the more rigid supraballs remain spherical. By subjecting the internalized suprastructures to a magnetic field, they both align and form long chains. Overall, it is highlighted that the mechanical and topological properties of the self‐assemblies direct their intracellular fate allowing the control intralysosomal density, ordering, and localization of NCs.  相似文献

This article describes the application of a multiobjective evolutionary algorithm for locating roadside infrastructure for vehicular communication networks over realistic urban areas. A multiobjective formulation of the problem is introduced, considering quality‐of‐service and cost objectives. The experimental analysis is performed over a real map of Málaga, using real traffic information and antennas, and scenarios that model different combinations of traffic patterns and applications (text/audio/video) in the communications. The proposed multiobjective evolutionary algorithm computes accurate trade‐off solutions, significantly improving over state‐of‐the‐art algorithms previously applied to the problem.  相似文献

The acidity and catalytic properties of aluminosilicate mesoporous molecular sieves with the MCM-41 structure and bulk Si/Al ratios in the 10–60 range have been investigated. The incorporation of 4-coordinate aluminium into the structure of MCM-41 generates both BrØnsted and Lewis acid sites in amounts increasing with the degree of incorporation. However, the BrØnsted/Lewis acid population ratio is independent of the content of aluminium. The number and strength of acid sites generated are comparable to those of a pillared acid-activated clay and lower than in zeolite H-Y with Si/Al=3.65. Aluminosilicate MCM-41 is a moderate catalyst for the conversion of cumene which proceeds predominantly via catalytic cracking to propene and benzene. The sample of MCM-41 with the highest content of framework aluminium (Si/Al=10) has the largest number of BrØnsted acid sites and exhibits highest catalytic activity.  相似文献

Background

The biological effects of nanoparticles depend on several characteristics such as size and shape that must be taken into account in any type of assessment. The increased use of titanium dioxide nanoparticles (TiO2NPs) for industrial applications, and specifically as a food additive, demands a deep assessment of their potential risk for humans, including their abilities to cross biological barriers.

Methods

We have investigated the interaction of three differently shaped TiO2NPs (nanospheres, nanorods and nanowires) in an in vitro model of the intestinal barrier, where the coculture of Caco-2/HT29 cells confers inherent intestinal epithelium characteristics to the model (i.e. mucus secretion, brush border, tight junctions, etc.).

Results

Adverse effects in the intestinal epithelium were detected by studying the barrier’s integrity (TEER), permeability (LY) and changes in the gene expression of selected specific markers. Using Laser Scanning Confocal Microscopy, we detected a different behaviour in the bio-adhesion and biodistribution of each of the TiO2NPs. Moreover, we were able to specifically localize each type of TiO2NPs inside the cells. Interestingly, general DNA damage, but not oxidative DNA damage effects, were detected by using the FPG version of the comet assay.

Conclusions

Results indicate different interactions and cellular responses related to differently shaped TiO2NPs, nanowires showing the most harmful effects.

In the present work, D‐isosorbide and 1,3‐propanediol are proposed as alternative plasticizers obtained from renewable resources. Plasticized starch films were prepared by solvent casting method. The influence of using different “green” plasticizers in the final properties of starch‐based films was analyzed. Besides, the characterization of the films was also performed after storage time in order to evaluate the effect of the plasticizer on aging. UV‐spectrophotometry results showed better optical properties for both glycerol and D‐isosorbide films with higher transparency. The thermal and mechanical properties resulted influenced by the nature of the plasticizer. It was demonstrated that water vapor permeability was governed by the starch‐water interactions, whereas the oxygen permeability depended on the plasticizer's nature. The storage time affected the surface, mechanical, and thermal properties of the plasticized starch films. Atomic force microscopy results concluded that the topography of the films changed due to aging. The use of D‐isosorbide as plasticizer reduced the evolution of the mentioned properties and enhanced the reliability of the material. © 2017 Wiley Periodicals, Inc. J. Appl. Polym. The objective of this study was to investigate the potential in vitro biological properties of Oz.Or.Oil 30, a new formulation composed of 30% ozonated sunflower seed oil, which is believed to keep skin smooth and moisturized, supporting repair processes, tissue regeneration and re-epithelialization of wounds. The antibacterial activity, the qualitative and quantitative evaluation of the cytotoxic effect of the formulation on cultures of Vero cells and 3T3 fibroblasts showed that Oz.Or.Oil 30 merits further in vivo study using clinical-laboratory correlations, because it could be suggested as an alternative therapy against bacterial and fungal diseases.  相似文献

The Mycobacterium tuberculosis Ser/Thr kinase PknB is implicated in the regulation of bacterial cell growth and cell division. The intracellular kinase function of PknB is thought to be triggered by peptidoglycan (PGN) fragments that are recognized by the extracytoplasmic domain of PknB. The PGN in the cell wall of M. tuberculosis has several unusual modifications, including the presence of N-glycolyl groups (in addition to N-acetyl groups) in the muramic acid residues and amidation of d -Glu in the peptide chains. Using synthetic PGN fragments incorporating these diverse PGN structures, we analyzed their binding characters through biolayer interferometry (BLI), NMR spectroscopy, and native mass spectrometry (nMS) techniques. The results of BLI showed that muropeptides containing 1,6-anhydro-MurNAc and longer glycan chains exhibited higher binding potency and that the fourth amino acid of the peptide stem, d -Ala, was crucial for protein recognition. Saturation transfer difference (STD) NMR spectroscopy indicated the major involvement of the stem peptide region in the PASTA-PGN fragment binding. nMS suggested that the binding stoichiometry was 1:1. The data provide the first molecular basis for the specific interaction of PGN with PknB and firmly establish PGNs as the effective ligands of PknB.  相似文献

The precise role that nitric oxide (NO) plays in the mechanisms of ischemic brain damage remains to be established. The expression of the inducible isoform (iNOS) of NO synthase (NOS) has been demonstrated not only in blood and glial cells using in vivo models of brain ischemia-reperfusion but also in neurons in rat forebrain slices exposed to oxygen-glucose deprivation (OGD). We have used this experimental model to study the effect of OGD on the neuronal isoform of NOS (nNOS) and iNOS. In OGD-exposed rat forebrain slices, a decrease in the calcium-dependent NOS activity was found 180 min after the OGD period, which was parallel to the increase during this period in calcium-independent NOS activity. Both dexamethasone and cycloheximide, which completely inhibited the induction of the calcium-independent NOS activity, caused a 40-70% recovery in calcium-dependent NOS activity when compared with slices collected immediately after OGD. The NO scavenger oxyhemoglobin produced complete recovery of calcium-dependent NOS activity, suggesting that NO formed after OGD is responsible for this down-regulation. Consistently, exposure to the NO donor (Z)-1-[(2-aminoethyl)-N-(2-ammonioethyl)amino]diazen-1-iu m-1,2-diolate (DETA-NONOate) for 180 min caused a decrease in the calcium-dependent NOS activity present in control rat forebrain slices. Furthermore, OGD and DETA-NONOate caused a decrease in level of both nNOS mRNA and protein. In summary, our results indicate that iNOS expression down-regulates nNOS activity in rat brain slices exposed to OGD. These studies suggest important and complex interactions between NOS isoforms, the elucidation of which may provide further insights into the physiological and pathophysiological events that occur during and after cerebral ischemia.  相似文献

The thermoluminescent behaviour of diamond films subjected to UV irradiation was studied by using an UV lamp of 254 nm wavelength. The UV irradiation was achieved by placing the samples 15 cm away from an UV source for different periods. The thermoluminescent signal was integrated from 0 to 350°C at a linear heating rate of 10°C/s in a N2 atmosphere. The corresponding luminescence spectra show an excitation band centered at 450 nm while the emission band is centered around 500 nm at room temperature. The diamond films were synthesized on molybdenum substrates by the combustion flame technique and characterized by Raman spectroscopy and scanning electron microscopy.
