MPPA光动力作用诱导人鼻咽癌细胞凋亡的实验研究 总被引:3,自引:0,他引:3
为观察MPPa光动力作用对鼻咽癌细胞凋亡的影响,应用AnnexinV—PI双染结合流式细胞仪分析MPPa光动力作用后人鼻咽癌细胞株CNE2细胞发生凋亡和继发性坏死的比率。结果显示MPPa光动力作用实验组人鼻咽癌细胞株CNE2细胞发生凋亡和继发性坏死的比率分别增加到16.43 %和4.64 % ,且均显著高于单纯光照射组、单纯MMPa光敏剂处理组和假照射组(P <0 .0 1) ,而三对照组间无明显差异(P >0 .0 5 )。表明MPPa光动力作用能有效诱导低分化人鼻咽癌细胞株CNE2细胞凋亡的发生。这也可能是MPPa光动力作用杀伤鼻咽癌的重要机制之一。 相似文献

Effect of freezing/thawing conditions and long‐term frozen storage on the quality of mashed potatoes

The effects of freezing temperature (−80, −40 or −24 °C) and thawing mode (microwave or overnight at 4 °C) on quality parameters of mashed potatoes made from tubers (cv Kennebec) and from potato flakes were examined, as was the effect of long‐term frozen storage on the quality of mashed potatoes. Mashed potatoes were tested for texture profile analysis (TPA) and cone penetration, oscillatory and steady rheometry, colour, dry matter, Brix and sensory analyses. In natural mashed potatoes, TPA hardness and oscillatory parameters showed that processing resulted in a softer product than the fresh control. The parameters were lower in the samples thawed at 4 °C than in those thawed by microwave at all the freezing temperatures used, which may be ascribed to gelatinisation of the starch released from damaged cells. Differences from the freshly prepared product decreased when the samples were frozen at −80 °C and thawed by microwave. No difference was found in sensory acceptability between samples frozen at −80 and −40 °C, which probably reflects the panellists' mixed preferences for air‐thawed versus microwave‐thawed samples. Increasing the time in frozen storage led to a natural mash with a firmer texture, higher L*/b* value and Brix; nonetheless, panellists found the samples at 0, 3 and 12 months of frozen storage equally acceptable. In commercial mash, penetration and oscillatory parameters showed that processing made for a firmer product than the fresh control, probably owing to retrogradation of gelatinised starch. Thawing mode had a significant effect on parameters, which were lower in the samples thawed at 4 °C. The structure and quality of commercial mash was more detrimentally affected by freezing and, therefore, we would not recommend either freezing or frozen storage of this mashed potato in the used conditions. Natural mash made from Kennebec potatoes should be frozen quickly and thawed by microwave in the conditions described to obtain a product more similar to that freshly made. This research investigated the intakes of six intense sweeteners: acesulfame-K (E950), aspartame (E951), cyclamate (E952), saccharin (E954), sucralose (E955), and steviol glycosides (E960) in the diets of Irish adults, using data from the National Adult Nutrition Survey. A food label survey that included products currently available on the Irish market supplemented the analysis. Sweetener intakes were investigated using three different exposure scenarios; beginning with a crude assessment which assumed that all foods permitted to contain the additives of interest always did contain them, and at their maximum permitted level (Tier 1). Refined assessments estimated intakes of the six sweeteners using food consumption data up to brand level with additive occurrence data from a survey of products currently available on the Irish market (Tier 2) and sweetener concentration data (Tier 3). Results of all exposure assessment scenarios demonstrate that intakes of each of the sweeteners of interest by the total population were below the relevant ADI level (mg kg?1 bodyweight?1), even by high consumers (P99). The three sweeteners consumed in highest amounts were acesulfame-k, aspartame, and sucralose. The main sources of these sweeteners in the diet were ‘cider and perry’, ‘energy reduced and no added sugar (ER and NAS) carbonated flavoured drinks’, ‘table-top sweeteners’, ‘dairy products’, ‘solid food supplements’, and ‘sauces’. Intakes of the six intense sweeteners are currently not a concern among Irish adults. Electrokinetic extraction has been demonstrated to be very effective in removing heavy metals from Georgia kaolinite. The relatively high removal efficiency depends on the extremely acidic soil environment generated by the electrokinetic process. However, the efficiency observed in Georgia kaolinite cannot be achieved in soils of high acid/base buffer capacity without enhancement. In this study, the effect of ethylenediaminetetraacetic acid (EDTA) to enhance electrokinetic extraction of cadmium from Milwhite kaolinite was examined. The influence of electro-osmotic flow direction on the migration of cadmium, EDTA, and their complexes were also investigated. It was observed that injection of EDTA from the cathode reservoir by a reverse electro-osmotic flow could mobilize the cadmium in the specimen effectively. A less significant mobilization of cadmium was observed when the electro-osmotic flow was directed toward the cathode. However, accumulation of cadmium near the anode was observed regardless of the electro-osmotic flow direction. 相似文献

This article describes the use of structural equation modeling with latent variables to examine group differences and test competing models about cause–effect relationships in passive longitudinal designs. This approach is compared with several other statistical methods including analysis of cross-lagged panel correlations, regression analysis, and path analysis. The mechanics and advantages of structural equation modeling are illustrated using an example based on a 3-wave longitudinal study of adolescents' alcohol use. Within this example, the generalizability of the measurement model and structural model are assessed across gender and time, and competing models about the causes and consequences of adolescents' alcohol use are tested. The article concludes with a discussion of some of the strengths and limitations of using structural equation modeling with longitudinal data. People with Alzheimer's disease living in a defined community in New York City were identified and assessed on two occasions to determine the number of hours of activities of daily living (ADL) care they received. Nearly half received all care hours from informal sources; however, a quarter received all ADL care hours from formal sources. The mean number of daily ADL hours in the sample was 9.7, of which 4.2 came from formal sources. This reflects the extensive use of Medicaid-based home care services in New York City. As dementia worsened, substitution of formal for informal care was rare, but formal care assumed a greater proportion of total care hours. 相似文献

The enzyme nitric oxide synthase catalyzes the oxidation of the amino acid L-arginine to L-citrulline and nitric oxide in an NADPH-dependent reaction. Nitric oxide plays a critical role in signal transduction pathways in the cardiovascular and nervous systems and is a key component of the cytostatic/cytotoxic function of the immune system. Characterization of nitric oxide synthase substrates and cofactors has outlined the broad details of the overall reaction and suggested possibilities for chemical steps in the reaction; however, the molecular details of the reaction mechanism are still poorly understood. Bioremediation of oil spills on tidally influenced beaches commonly involves the addition of a nutrient solution to the contaminated region of the beach at low tide to stimulate the growth of indigenous oil-degrading bacteria. Maximizing the residence time of nutrients in the beach and subsequently their contact time with microorganisms is a main goal for successful bioremediation. Therefore, understanding the effects of the tide on water flow and solute transport in a beach is an essential task for designing a nutrient application strategy. We investigated these effects by conducting a tracer study in a laboratory beach simulating nutrient application on natural beaches. The study consisted of applying, at low tide, a conservative tracer solution onto the beach surface near the high-tide line and monitoring its movement in the beach subsurface. The tidal motion caused the applied plume to move downward and seaward. The downward movement occurred during rising tides, while the seaward movement occurred mainly during falling tides. The results indicate that nutrients should be applied at the high-tide line during low tides. Guidelines for scaling up the results to natural beaches are provided along with an example. 相似文献
