On the efficient implementation of conditional critical regions and the construction of monitors

Summary A method is presented that allows the efficient implementation of conditional critical regions combined with scheduling of the waiting processes. It is based on the knowledge of static and invariant relations that exist among the conditional critical regions of a process system. Mathematical methods are applied in order to show the nature of these relations, and to determine them for practical applications.By collecting the conditional critical regions in a program module, a monitor is obtained. Its rather abstract user level allows clear and well structured programming style. It also shows some advantages in comparison to other monitor proposals on the implementation level.Work done mainly at the Fakultät für Informatik, Universität Karlsruhe, Karlsruhe, and also at the Department of Computer Science, University of Toronto, Toronto.     

Flip the tip: an automated,high quality,cost-effective patch clamp screen

The race for creating an automated patch clamp has begun. Here, we present a novel technology to produce true gigaseals and whole cell preparations at a high rate. Suspended cells are flushed toward the tip of glass micropipettes. Seal, whole-cell break-in, and pipette/liquid handling are fully automated. Extremely stable seals and access resistance guarantee high recording quality. Data obtained from different cell types sealed inside pipettes show long-term stability, voltage clamp and seal quality, as well as block by compounds in the pM range. A flexible array of independent electrode positions minimizes consumables consumption at maximal throughput. Pulled micropipettes guarantee a proven gigaseal substrate with ultra clean and smooth surface at low cost.     

Electrical, optical and morphological properties of nanoparticle indium-tin-oxide layers

Porous layers were prepared from DEGUSSA's ITO (In₂O₃:Sn) nanoparticle dispersion by doctor blading followed by annealing in air. We investigated the influence of various annealing parameters on electrical, optical and morphological thin film properties.Conductance rises with increasing annealing temperature and time by more than three orders of magnitude up to 44 Ω^− 1cm^− 1. Besides this we found an abrupt decrease in free charge carrier concentration above a critical annealing temperature of 250 °C, which leads to a step in conductance curve. In spite of particle growing during annealing no decrease in porosity was observed and in opposite to compact material, nanoparticle layers do not exhibit an appreciable shrinkage below recrystallisation temperature. These both indicate a densification hindering particle pinning effect, which is believed to be currently the main obstruction to achieve higher electrical conductivities.     

Using situation calculus for e-business agents

As the Internet grows, it is becoming less feasible for customers and merchants to manually visit each web site, analyze the information there, and make sound business decisions regarding the trading of goods or services. To cope with this evolution, software agents can be designed that are capable of automating the more routine, tedious, and time-consuming tasks involved in current trading processes. At a higher level agents may also be able to negotiate and make autonomous decisions and commitments on behalf of their owners.

This paper describes an agent implementation using the situation calculus, which offers a possibly unifying paradigm for dynamic agents. Interesting applications are currently being developed. Our contribution is a situation calculus agent system developed for e-business. Ongoing work is focused on implementing this system in an open marketplace environment.     

Driving Automotive User Interface Research

Cars offer an interesting but challenging microcosm for pervasive computing research and, in particular, for interaction with pervasive computing systems. Increasingly, researchers are looking at interactive applications in the car and investigating human-car interaction from a computer science—rather than an ergonomics or mechanical engineering—perspective. This article reports on the International Conference on Automotive User Interfaces and Interactive Vehicular Applications, wherein participants shared presentations on topics such as aesthetics, user interaction and distraction, safety, and driver monitoring.     

Molecular Subtyping of Invasive Breast Cancer Using a PAM50-Based Multigene Expression Test-Comparison with Molecular-Like Subtyping by Tumor Grade/Immunohistochemistry and Influence on Oncologist’s Decision on Systemic Therapy in a Real-World Setting

In intermediate risk hormone receptor (HR) positive, HER2 negative breast cancer (BC), the decision regarding adjuvant chemotherapy might be facilitated by multigene expression tests. In all, 142 intermediate risk BCs were investigated using the PAM50-based multigene expression test Prosigna® in a prospective multicentric study. In 119/142 cases, Prosigna® molecular subtyping was compared with local and two central (C1 and C6) molecular-like subtypes relying on both immunohistochemistry (IHC; HRs, HER2, Ki-67) and IHC + tumor grade (IHC+G) subtyping. According to local IHC, 35.4% were Luminal A-like and 64.6% Luminal B-like subtypes (local IHC+G subtype: 31.9% Luminal A-like; 68.1% Luminal B-like). In contrast to local and C1 subtyping, C6 classified >2/3 of cases as Luminal A-like. Pairwise agreement between Prosigna® subtyping and molecular-like subtypes was fair to moderate depending on molecular-like subtyping method and center. The best agreement was observed between Prosigna® (53.8% Luminal A; 44.5% Luminal B) and C1 surrogate subtyping (Cohen’s kappa = 0.455). Adjuvant chemotherapy was suggested to 44.2% and 88.6% of Prosigna® Luminal A and Luminal B cases, respectively. Out of all Luminal A-like cases (locally IHC/IHC+G subtyping), adjuvant chemotherapy was recommended if Prosigna® testing classified as Prosigna® Luminal A at high / intermediate risk or upgraded to Prosigna® Luminal B.     

The Chinese Room: Visualization and Interaction to Understand and Correct Ambiguous Machine Translation

We present The Chinese Room , a visualization interface that allows users to explore and interact with a multitude of linguistic resources in order to decode and correct poor machine translations. The target users of The Chinese Room are not bilingual and are not familiar with machine translation technologies. We investigate the ability of our system to assist such users in decoding and correcting faulty machine translations. We found that by collaborating with our application, end-users can overcome many difficult translation errors and disambiguate translated passages that were otherwise baffling. We also examine the utility of our system to machine translation researchers. Anecdotal evidence suggests that The Chinese Room can help such researchers develop better machine translation systems.     

Malignant meningioma in Gorlin's syndrome: cytogenetic and p53 gene analysis. Case report

Gorlin's syndrome, also known as multiple basal cell carcinoma syndrome, is a familial tumor condition with autosomal-dominant inheritance. Patients develop multiple basal cell carcinomas beginning in childhood. They also have a typical dysmorphic facies, skeletal malformations, and a particular type of epithelial cyst of the jaws. Recent evidence localizes a Gorlin's syndrome locus on chromosome 9 at band q31. Both tumors and malformations of the central nervous system occur with Gorlin's syndrome. Medulloblastoma is the primary brain tumor most frequently associated with this syndrome; over 40 such cases have been reported. However, only seven cases of meningioma associated with Gorlin's syndrome have been described. The authors report the case of a woman with Gorlin's syndrome whose mother and maternal grandfather also had the condition. The patient was found to have a medulloblastoma at 4 years of age and presented with a large bifrontal meningioma at 19 years of age. The meningioma was histologically malignant and had a complex karyotype with multiple translocations including a t(5;9) with the breakpoint on chromosome 9 located at 9q32. The constitutional karyotype of the mother was normal. No mutations of exons 5 to 9 of the p53 gene were detected using single-stranded conformational polymorphism analysis.     

Responses of Human Cells to PAH-Induced DNA Damage

Benzo[ a ]pyrene (B[ a ]P) and dibenzo[ a,l ]pyrene (DB[ a,l ]P) induce cytochrome P450 (CYP) CYP1A1 and CYP1B1, which metabolize these polycyclic aromatic hydrocarbons (PAHs) into DNA-binding species. In order to detail roles of CYP1A1 and CYP1B1 in activation of DB[ a,l ]P to the diol epoxide, we here report the inhibition of CYP1A1 in human MCF-7 cells with phosphorodiamidate morpholino antisense oligomers (morpholinos). PAH-DNA adduct formation was also determined after treatment with morpholinos and B[ a ]P or DB[ a,l ]P. p53 is involved in DNA repair, cell cycle arrest, and apoptosis. Cells with normal p53 protein arrest in the G1 phase of the cell cycle on exposure to DNA-damaging agents (presumably allowing the cell sufficient time to repair damaged DNA prior to replication). Previous studies in human MCF-7 cells indicate that cells with PAH-DNA adducts escape cell cycle arrest and accumulate in the S phase. In the present study the effect of PAH-DNA adducts on the cell cycle were observed in human diploid fibroblasts (HDF). We found that treatment of HDF with the diol epoxide of DB[ a,l ]P causes cell cycle arrest in G 1 . An increase in DNA adduct formation with increase in concentration of dibenzo[ a,l ]pyrene diol epoxide {( m )- anti -DB[ a,l ]PDE} was also observed.
