As synchrotron radiation becomes more intense, detectors become faster and structure‐solving software becomes more elaborate, obtaining single crystals suitable for data collection is now the bottleneck in macromolecular crystallography. Hence, there is a need for novel and advanced crystallisation agents with the ability to crystallise proteins that are otherwise challenging. Here, an Anderson–Evans‐type polyoxometalate (POM), specifically Na6[TeW6O24] ? 22 H2O (TEW), is employed as a crystallisation additive. Its effects on protein crystallisation are demonstrated with hen egg‐white lysozyme (HEWL), which co‐crystallises with TEW in the vicinity (or within) the liquid–liquid phase separation (LLPS) region. The X‐ray structure (PDB ID: 4PHI) determination revealed that TEW molecules are part of the crystal lattice, thus demonstrating specific binding to HEWL with electrostatic interactions and hydrogen bonds. The negatively charged TEW polyoxotungstate binds to sites with a positive electrostatic potential located between two (or more) symmetry‐related protein chains. Thus, TEW facilitates the formation of protein–protein interfaces of otherwise repulsive surfaces, and thereby the realisation of a stable crystal lattice. In addition to retaining the isomorphicity of the protein structure, the anomalous scattering of the POMs was used for macromolecular phasing. The results suggest that hexatungstotellurate(VI) has great potential as a crystallisation additive to promote both protein crystallisation and structure elucidation.  相似文献

Slow chemical reactions are a big challenge in the modern pharmaceutical industry. Their accelerations together with the introduction of continuous manufacturing modes are major drivers for future development. One example reaction is hydroamination, a reaction between unsaturated hydrocarbons and amines. Such a reaction type is the main focus of this work. More precisely, the anti‐Markovnikov hydroamination reaction between cis/trans 9H‐thioxanthene‐2‐chloro‐9‐(2‐propenylidene)‐(9CI) and 1‐(2‐hydroxyethyl)piperazine (HEP) is investigated in detail. It has been traditionally performed in toluene with a huge excess of HEP and reaction times of up to 24 h. Acceleration of the reaction from 24 down to 4 h is achieved by switching from batch operation mode in toluene to either solvent‐free batch mode or microwave‐assisted hydroamination with tetrahydrofuran as a solvent.  相似文献

Producing energy from renewable sources in Serbia is in its initial phase, and therefore this paper points towards the basic assumptions, potentials and institutional framework for the development of this activity in Serbia. Until recently, production and consumption of energy in Serbia was a social category, but the shift towards market economy together with the fact that Serbia has adopted Kyoto protocol, production of energy from renewable sources became a competitive and obligatory activity. Research shows that the current potentials for producing energy from renewable sources in Serbia are favorable. Besides this, the paper provides an overview of the existing institutional structure in the energy sector in Serbia, but also the short overview of the adequate legal acts. Researches that were done so far have proven that energy potentials of Serbia are insufficiently known and therefore authors will give an overview of the foreign investments in this sector and reveal the possibilities for further investing. Considering the existing know-how base and potentials for the production of equipment, geothermal energy and energy coming from biomass have been identified as priority renewable sources of energy. Producing energy from other renewable sources is also possible, but would require substantial foreign investments. As a final conclusion, the paper states that completely unused potentials for production of energy from renewable sources, together with adequately set institutional framework, would create great possibilities for foreign investments.  相似文献

Researchers at the National Institute for Occupational Safety and Health (NIOSH) are developing methods for characterizing diesel particulate matter in mines. Introduction of novel engine and exhaust aftertreatment technologies in underground mines is changing the nature of diesel emissions, and metrics alternative to the traditional mass-based measurements are being investigated with respect to their ability to capture changes in the properties of diesel aerosols. The emphasis is given to metrics based on measurement of number and surface area concentrations, but analysis of collected particles using electron microscopy (EM) is also employed for detailed particle characterization. To collect samples for EM analysis at remote workplaces, including mining and manufacturing facilities, NIOSH is developing portable particle samplers capable of collecting airborne nano-scale particles. This paper describes the design, construction, and testing of a prototype thermophoretic precipitator (TP) particle sampler optimized for collection of particles in the size range of 1–300 nm. The device comprises heated and cooled metal plates separated by a 0.8 mm channel through which aerosol is drawn by a pump. It weighs about 2 kg, has a total footprint of 27 × 22 cm, and the collection plate size is approximately 4 × 8 cm. Low power consumption and enhanced portability were achieved by using moderate flow rates (50–150 cm3/min) and temperature gradients (10–50 K/mm with ΔT between 8 K and 40 K). The collection efficiency of the prototype, measured with a condensation particle counter using laboratory-generated polydisperse submicrometer NaCl aerosols, ranged from 14–99%, depending on temperature gradient and flow rate. Analysis of transmission electron microscopy images of samples collected with the TP confirmed that the size distributions of collected particles determined using EM are in good agreement with those determined using a Fast Mobility Particle Sizer.

We look at the effect of modeling branch-outage contingencies on locational marginal prices. To model contingencies in the day-ahead auction, we formulate a two-stage stochastic program. Rather than follow the current practice of including a list of possible contingencies that must be satisfied, we incorporate a larger set of contingencies in the model and allow contingencies to result in load reductions/outages at a cost. The model can be used and interpreted in two ways. One is to look at the tradeoff between reliability and outage costs. Another is to consider the load losses resulting from a contingency to be consumer offers of load reductions in response to line outages as part of the day-ahead auction. In analyzing the model structure, we find that the prices in the model closer in definition to those currently used in the day-ahead auction do not maximize expected surplus because the day-ahead auction produces prices that assume shortages will never occur. This raises issues with the design of auctions with important stochastic elements in the market. We present results for a 68-node grid with 86 branches (lines and transformers) to illustrate how prices and expected values change as the costs of outages are varied.  相似文献

The aim of this study was to investigate the effect of changing spray-drying parameters on the production of a naratriptan/maltodextrin/lactose composite, as well as to evaluate the application of design of experiments and multilayer perceptron (MLP) neural network in studying the spray-drying process. The system was spray dried as an aqueous solution using a Büchi 290-Mini Spray Dryer (Büchi Laboratoriums-Technik AG, Switzerland). A 24?1 factorial design study was undertaken to select the spray-drying processing variables that significantly affect the production yield, outlet air temperature, and residual moisture content. The process parameters studied were inlet air temperature, pump speed, aspirator setting (drying air flow rate), and feed concentration. After performing this screening study, three process parameters, pump speed, inlet air temperature, and feed concentration, were selected for further analysis, applying a central composite design and artificial neural network. Overall, the parameter that had the greatest influence on each investigated response was pump speed. It significantly influenced yield, moisture content, and particle size. Interaction between inlet air temperature and feed concentration was the only statistically significant interaction that influenced the moisture content. Particle size was mostly influenced by feed concentration as well as by a pump speed. A multilayer perceptron was the type of artificial neural network applied. The selected MLP structure had three layers: the first layer had three input units, the second layer had three hidden units, and the third layer had four output units. The selected MLP was trained through 10,000 epochs. Design of experiments using response surface methodology allows insight into interactions between variables, and an artificial neural network provides better prediction potential, enabling simultaneously determination of several outputs. Design of experiments and artificial neural network proved to be useful tool for optimization of the spray-drying process, where a design space for achieving the best process yields and optimum particle characteristics was established. There is a possibility that this conclusion can be drawn for other maltodextrin/lactose systems with other drug substances or for other similar spray-dried systems (the feedstock type is a carbohydrate-based aqueous solution).
