The effect of topcoat properties on the tendency of painted materials to blistering was studied. Six topcoats were applied on identical panels of hot-dip galvanized steel painted with a polyester primer. The tendency to blistering was assessed under the conditions of permanent condensation in a Q-panel condensation test at 60 °C. Internal tensile stress and stress development in organic coatings during temperature and relative humidity cycling were investigated by the cantilever curvature method. Although blisters originated from the metal/polymer interface, the extent of blistering was strongly influenced by the topcoat. Available data suggest that it may increase with the coating thickness, glass transition temperature (Tg) and thermal expansion properties. Connection was found between the internal tensile stress formed in topcoats during the paint film preparation and the extent of blistering. A hypothesis that stress-assisted interfacial bond hydrolysis was responsible for blister initiation is proposed. Other experiments suggested that local paint buckling over non-adherent sites can be caused by plastic deformation of the paint due to relief of compressive stress generated at elevated temperature or by ingress of water.  相似文献

This article presents a modified robust disturbance observer-RIC for a mechanicalpositioning system. The proposed RIC structure ensures a higher level of stability margin and offers transparent selection of the controller’s structure and feedback dynamic. The modified RIC controller design is divided into two main stages, where the first stage provides a design for the internal robust controller and the second a design for the external performance controller. The controller structure ensures a robust stability and performances property and good capability of low-frequency input and output disturbances suppression. The controller synthesis based on a pole-placement technique using optimization of the robust criteria based on even polynomials, and positive conditions. The solution to the problem is based on a multi-criterion optimization algorithm with a fixed order controller structure.  相似文献

Replacement of the Lactobacillus dominated vaginal microbiome by a mixed bacterial population including Prevotella bivia is associated with bacterial vaginosis (BV). To understand the impact of P. bivia on this microbiome, its growth requirements and mode of energy production were studied. Anoxic growth with glucose depended on CO2 and resulted in succinate formation, indicating phosphoenolpyruvate carboxylation and fumarate reduction as critical steps. The reductive branch of fermentation relied on two highly active, membrane-bound enzymes, namely the quinol:fumarate reductase (QFR) and Na+-translocating NADH:quinone oxidoreductase (NQR). Both enzymes were characterized by activity measurements, in-gel fluorography, and VIS difference spectroscopy, and the Na+-dependent build-up of a transmembrane voltage was demonstrated. NQR is a potential drug target for BV treatment since it is neither found in humans nor in Lactobacillus. In P. bivia, the highly active enzymes L-asparaginase and aspartate ammonia lyase catalyze the conversion of asparagine to the electron acceptor fumarate. However, the by-product ammonium is highly toxic. It has been proposed that P. bivia depends on ammonium-utilizing Gardnerella vaginalis, another typical pathogen associated with BV, and provides key nutrients to it. The product pattern of P. bivia growing on glucose in the presence of mixed amino acids substantiates this notion.  相似文献

Activated endothelial, immune, and cancer cells prefer glycolysis to obtain energy for their proliferation and migration. Therefore, the blocking of glycolysis can be a promising strategy against cancer and autoimmune disease progression. Inactivation of the glycolytic enzyme PFKFB3 (6-phosphofructo-2-kinase/fructose-2,6-biphosphatase) suppresses glycolysis level and contributes to decreased proliferation and migration of cancer (tumorigenesis) and endothelial (angiogenesis) cells. Recently, several glycolysis inhibitors have been developed, among them (E)-1-(pyridin-4-yl)-3-(quinolin-2-yl)prop-2-en-1-one (PFK15) that is considered as one of the most promising. It is known that PFK15 decreases glucose uptake into the endothelial cells and efficiently blocks pathological angiogenesis. However, no study has described sufficiently PFK15 synthesis enabling its general availability. In this paper we provide all necessary details for PFK15 preparation and its advanced characterization. On the other hand, there are known tyrosine kinase inhibitors (e.g., sunitinib), that affect additional molecular targets and efficiently block angiogenesis. From a biological point of view, we have studied and proved the synergistic inhibitory effect by simultaneous administration of glycolysis inhibitor PFK15 and multikinase inhibitor sunitinib on the proliferation and migration of HUVEC. Our results suggest that suppressing the glycolytic activity of endothelial cells in combination with growth factor receptor blocking can be a promising antiangiogenic treatment.  相似文献

In addition to their LDL-cholesterol-lowering effect, statins have pleiotropic beneficial effects on the cardiovascular system. However, long-term treatment with statins may be associated with serious side effects. With the aim to make statin therapy more effective, we studied the effects of simvastatin- and coenzyme-Q10-loaded polymeric nanoparticles on the lipid profile and nitric oxide (NO)/reactive oxygen species (ROS) balance in the heart and aorta of adult male obese Zucker rats. The rats were divided into an untreated group, a group treated with empty nanoparticles, and groups treated with simvastatin-, coenzyme Q10 (CoQ10)-, or a combination of simvastatin- and CoQ10-loaded nanoparticles (SIMV+CoQ10). After 6 weeks, the lipid profile in the plasma and the concentration of conjugated dienes in the liver were determined. Nitric oxide synthase (NOS) activity, Akt, endothelial NOS (eNOS), phosphorylated eNOS (p-eNOS), nicotinamide adenine dinucleotide phosphate (NADPH) oxidase, and nuclear factor kappaB (NF-kappaB) protein expressions were measured in the heart and aorta. All simvastatin, CoQ10, and SIMV+CoQ10 treatments decreased plasma LDL levels, but only the combined SIMV+CoQ10 treatment increased NOS activity and the expression of Akt, eNOS, and p-eNOS in both the heart and the aorta. Interestingly, NADPH oxidase in the heart and NF-kappaB protein expression in the aorta were decreased by all treatments, including nanoparticles alone. In conclusion, only combined therapy with SIMV- and CoQ10-loaded nanoparticles increased NOS activity and upregulated the Akt-eNOS pathway in obese Zucker rats, which may represent a promising tool for the treatment of cardiometabolic diseases.  相似文献

The aim of the present study is to determine how electroretinographic (ERG) responses reflect age-related disease progression in the Stargardt disease (STGD1). The prospective comparative cohort study included 8 patients harboring two null ABCA4 variants (Group 1) and 34 patients with other ABCA4 genotypes (Group 2). Age at exam, age at onset, visual acuity (VA) and ERG responses were evaluated. The correlation between ERG responses and age in each patient group was determined using linear regression. A Mann-Whitney U Test was used to compare the median values between the groups. Age of onset was significantly earlier in Group 1 than in Group 2 (8 vs. 18), while disease duration was similar (13 vs. 12 years, i.e., advanced stage). Group 1 had significantly worse VA and lower ERG responses. ERG responses that significantly correlated with age in Group 1 were DA 0.01 and 3.0 ERG, which represented a retinal rod system response. The only ERG response that significantly correlated with age in Group 2 was the S-cone ERG. The observed difference was likely due to early cone loss occurring in double-null patients and slower photoreceptor loss in patients with other genotypes. The results suggest that specific ERG responses may be used to detect double-null patients at an early stage and monitor STGD1 disease progression in patients with specific genotypes.  相似文献

A liquid–liquid phase transfer catalyzed (PTC) esterification reaction of 4-t-butylphenol in aqueous phase (1 M sodium hydroxide solution) and 4-methoxybenzoyl chloride in organic phase (dichloromethane) in a microchannel under parallel laminar flow conditions was studied in this work. Tetrabutylammonium bromide was used as the PTC. Stable liquid–liquid hydrodynamic flow and a defined specific interfacial area in a microreactor offer considerable benefits over conventional batch reactors and are crucial to study interactions between kinetics and mass transfer effects. Mentioned features were used to develop a 3D mathematical model considering convection in the flow direction, diffusion in all spatial directions, and reactions in organic and aqueous phases. Results have shown a much higher mass transfer rate of the PTC between both phases as the one predicted by the 3D mathematical model. It may be assumed that the instability of parallel flow, along with the mass transfer of catalyst between both phases, causes rippling and erratic pulsation at the interface which then leads to interfacial convection and increased mass transfer rates. With a proposed correlation for mass transfer enhancement due to interfacial convection, all the experimental data were successfully predicted by the model.  相似文献
