The objective of this study was to integrate data on the effect of water activity (aw; 0.995–0.93) and temperature (20–35°C) on activation of the biosynthetic FUM genes, growth and the mycotoxins fumonisin (FB1, FB2) by Fusarium verticillioides in vitro. The relative expression of nine biosynthetic cluster genes (FUM1, FUM7, FUM10, FUM11, FUM12, FUM13, FUM14, FUM16 and FUM19) in relation to the environmental factors was determined using a microarray analysis. The expression was related to growth and phenotypic FB1 and FB2 production. These data were used to develop a mixed-growth-associated product formation model and link this to a linear combination of the expression data for the nine genes. The model was then validated by examining datasets outside the model fitting conditions used (35°C). The relationship between the key gene (FUM1) and other genes in the cluster (FUM11, FUM13, FUM9, FUM14) were examined in relation to aw, temperature, FB1 and FB2 production by developing ternary diagrams of relative expression. This model is important in developing an integrated systems approach to develop prevention strategies to control fumonisin biosynthesis in staple food commodities and could also be used to predict the potential impact that climate change factors may have on toxin production.  相似文献

The work–family interface is a highly relevant, yet underexposed issue in project management. When projects require family-separation, work demands are likely to conflict with family life and the consequences may be neither beneficial for (project) organizations nor for families. The aim of this study was twofold: a) to assess within-person changes over time in employees' work–family conflict experiences, relationship satisfaction, and turnover intentions and b) to examine the interrelations between these variables, over the course of a project that requires family-separation. Quantitative data were collected among military personnel before and after their four to six months assignments abroad. The results revealed that relationship satisfaction among military personnel had decreased significantly and turnover intentions had increased significantly over time. Moreover, employees who experienced higher levels of work–family conflict reported lower levels of relationship satisfaction and higher levels of turnover intentions. Understanding the interrelations may help managing projects in a way that benefits project organizations as well as employees and their families, who are critical to its performance and success.  相似文献

A nectarine purée was manufactured with different pretreatments (thermal blanching or ascorbic acid – AA – addition), and then, the purée was processed by high‐pressure treatment to evaluate the effect of the initial manufacture conditions in the stability of the processed purées. A thermal treatment was also carried out to compare the effect with the high‐pressure processing (HPP). All applied processes were effective to ensure the microbiological safety of the purées. However, the pretreatment (thermal blanching or AA addition) applied during the manufacture affected the final quality of the processed purées. Initially, the AA addition had a protective effect on colour degradation during the manufacture of the purées; however, when these purées were treated by HPP showed less colour stability during storage, lower bioactive compounds content, and antioxidant activity. In contrast, purées with an initial thermal blanching maintained better the quality after HPP and during storage.  相似文献

We fabricate freely suspended nanosheets of molybdenum disulphide (MoS2) which are characterized by quantitative optical microscopy and high-resolution friction force microscopy. We study the elastic deformation of freely suspended nanosheets of MoS2 using an atomic force microscope. The Young''s modulus and the initial pre-tension of the nanosheets are determined by performing a nanoscopic version of a bending test experiment. MoS2 sheets show high elasticity and an extremely high Young''s modulus (0.30 TPa, 50% larger than steel). A pilot study was conducted to determine whether membrane treatment on a side stream of recirculating cooling-tower water could reduce overall water usage and discharge. The treated permeate was returned to the cooling tower while the concentrate was discharged to the sanitary sewer. Flow rates, pressures and water chemistry were monitored. The pilot demonstrated potential substantial water savings. Maximum make-up water and discharge reduction were 16% and 49%, respectively. As high as possible permeate recovery is needed to maximize water conservation. Silica scaling on the membranes limited water savings in this pilot. Development of membranes with a solute-rejection capacity less than the 92% average of the membranes used in the pilot would assist in optimizing water savings. Decreased water outlays compensated for the additional energy used by membrane treatment. Scaling control is critical for economic operation.  相似文献

Global demand for mobility is increasing and the environmental impact of transport has become an important issue in transportation network planning and decision-making, as well as in the operational management phase. Suitable methods are required to assess emissions and fuel consumption reduction strategies that seek to improve energy efficiency and furthering decarbonization. This study describes the development and application of an improved modeling framework – the HERA (Highway EneRgy Assessment) methodology – that enables to assess the energy and carbon footprint of different highways and traffic flow scenarios and their comparison. HERA incorporates an average speed consumption model adjusted with a correction factor which takes into account the road gradient. It provides a more comprehensive method for estimating the footprint of particular highway segments under specific traffic conditions. It includes the application of the methodology to the Spanish highway network to validate it. Finally, a case study shows the benefits from using this methodology and how to integrate the objective of carbon footprint reductions into highway design, operation and scenario comparison.  相似文献
