The aim of the study was to demonstrate the biostimulating effect of exogenous melatonin (MEL) applied to seeds via hydroconditioning. It was indicated that only well-chosen application technique and MEL dose guarantees success concerning seed germination and young seedlings growth under stress conditions. For maize seed, 50 μM of MEL appeared to be the optimal dose. It improved seed germination and embryonic axes growth especially during chilling stress (5 °C/14 days) and during regeneration after its subsided. Unfortunately, MEL overdosing lowered IAA level in dry seeds and could disrupt the ROS-dependent signal transduction pathways. Very effective antioxidant MEL action was confirmed by low level of protein oxidative damage and smaller quantity of lipid oxidation products in embryonic axes isolated from seeds pre-treated with MEL and then exposed to cold. The stimulatory effects of MEL on antioxidant enzymes: SOD, APX and GSH-PX and on GST-a detoxifying enzyme, was also demonstrated. It was indicated for the first time, that MEL induced defence strategies against stress at the cytological level, as appearing endoreplication in embryonic axes cells even in the seeds germinating under optimal conditions (preventive action), but very intensively in those germinating under chilling stress conditions (intervention action), and after stress removal, to improve regeneration.  相似文献

The impact of seminal plasma components on the fertilization outcomes in humans is still under question. The increasing number of couples facing problems with conception raises the need for predictive biomarkers. Detailed understanding of the molecular mechanisms accompanying fertilization remains another challenge. Carbohydrate–protein recognition may be of key importance in this complex field. In this study, we analyzed the unique glycosylation pattern of seminal plasma proteins, the display of high-mannose and hybrid-type oligosaccharides, by means of their reactivity with mannose-specific Galanthus nivalis lectin. Normozoospermic infertile subjects presented decreased amounts of lectin-reactive glycoepitopes compared to fertile donors and infertile patients with abnormal semen parameters. Glycoproteins containing unveiled mannose were isolated in affinity chromatography, and 17 glycoproteins were identified in liquid chromatography-tandem mass spectrometry with electrospray ionization. The N-glycome of the isolated glycoproteins was examined in matrix-assisted laser desorption ionization mass spectrometry. Eleven out of 27 identified oligosaccharides expressed terminal mannose residues, responsible for lectin binding. We suggest that lowered content of high-mannose and hybrid type glycans in normozoospermic infertile patients may be associated with impaired sperm protection from preterm capacitation and should be considered in the search for new infertility markers.  相似文献

Two series of aromatic polyimides containing various linkage groups based on 2,7‐bis(4‐aminophenoxy)naphthalene or 3,3′‐dimethyl‐4,4′‐diaminodiphenylmethane and different aromatic dianhydrides, namely 4,4′‐(4,4′‐isopropylidenediphenoxy)bis(phthalic anhydride), 4,4′‐(hexafluoroisopropylidene)bis(phthalic anhydride), 3,3′,4,4′ benzophenonetetracarboxylic dianhydride, 9,9‐bis[4‐(3,4‐dicarboxyphenoxy)phenyl]fluorene dianhydride and 4,4′‐(4,4′‐hexafluoroisopropylidenediphenoxy)bis(phthalic anhydride), were synthesized and compared with regard to their thermal, mechanical and gas permeation properties. All these polymers showed high thermal stability with initial decomposition temperature in the range 475–525 °C and glass transition temperature between 208 and 286 °C. Also, the polymer films presented good mechanical characteristics with tensile strength in the range 60–91 MPa and storage modulus in the range 1700–2375 MPa. The macromolecular chain packing induced by dianhydride and diamine segments was investigated by examining gas permeation through the polymer films. The relationships between chain mobility and interchain distance and the obtained values for gas permeability are discussed. © 2014 Society of Chemical Industry  相似文献

The aim of this research was to observe the relationship between zeta potential, morphology, surface area, porosity, chemical composition, and ecotoxicity of nanocomposite powders such as Au/TiO2, Ag2O/TiO2, PdO/TiO2, Ag/TiO2/SiO2, Ag/N(C)TiO2, and SiO2/TiO2 from which Ag2O/TiO2, Ag/N(C)/TiO2, and Ag/TiO2/SiO2 were exhibiting good antimicrobial properties. It was observed, that nanomaterials characterized by similar morphology and zeta potential revealed the similar toxic behavior. The samples of higher agglomeration and higher zeta potential, especially Ag/TiO2/SiO2 xerogel and TiO2/SiO2 aerogel were generally less ecotoxic to water organisms and plants. They were also not genotoxic in concentrations up to 500 and 250 mg/L, respectively.  相似文献

Excess adiposity is associated with chronic inflammation, which takes part in the development of obesity-related complications. The aim of this study was to establish whether subcutaneous (SAT) or visceral (VAT) adipose tissue plays a major role in synthesis of pro-inflammatory cytokines. Concentrations of interleukins (IL): 1β, 6, 8 and 15 were measured at the protein level by an ELISA-based method and on the mRNA level by real-time PCR in VAT and SAT samples obtained from 49 obese (BMI > 40 kg/m2) and 16 normal-weight (BMI 20–24.9 kg/m2) controls. IL-6 and IL-15 protein concentrations were higher in SAT than in VAT for both obese (p = 0.003 and p < 0.0001, respectively) and control individuals (p = 0.004 and p = 0.001, respectively), while for IL-1β this was observed only in obese subjects (p = 0.047). What characterized obese individuals was the higher expression of IL-6 and IL-15 at the protein level in VAT compared to normal-weight controls (p = 0.047 and p = 0.016, respectively). Additionally, obese individuals with metabolic syndrome had higher IL-1β levels in VAT than did obese individuals without this syndrome (p = 0.003). In conclusion, concentrations of some pro-inflammatory cytokines were higher in SAT than in VAT, but it was the increased pro-inflammatory activity of VAT that was associated with obesity and metabolic syndrome.  相似文献

Aspects of calibration of intensities of SIMS secondary ions vs concentration as well as sputtering time vs depth are considered for Cr-doped CoO. Advantages and limitations of the SIMS method in quantitative analysis of segregation-induced concentration profiles in oxide crystals are discussed. The studies indicate a substantial effect due to charging the surface during sputtering. The depth calibration was performed by using the Ta2O5/Ta system as a standard. Good depth resolution was revealed. The calibration dependence of Cr intensities on concentration is characterized by a wide scatter of data caused by charging the surface. Very good shape reproducibility of the intensity ratio vs depth profiles was revealed. Therefore, normalized intensity ratios can be used for calibration.  相似文献

Polyethylene wastes (low-density polyethylene, high-density polyethylene and their binary blends) were subjected to high-energy radiation, using a 60Co gamma radiation source. The crosslinked materials thus obtained were processed to heatshrinkable films. Tensile strength could be sharply improved by increasing the dose up to 20 Mrad, simultaneously increasing the elongation at break of the most degraded PE waste. An increase of the degree of compatibility of LDPE and HDPE waste was also observed. All samples examined exhibit a “memory effect” after drawing at 130°C and cooling under tension followed by further heating under relaxed conditions. The value of shrinkage depended on the degree of degradation of the PE waste and on the irradiation dose.  相似文献

The effect of a Lewis acid addition to a coking coal on the porosity and reactivity towards steam of the resulting iron enriched coal chars are studied. GIC (FeCl3 graphite intercalation compound) or free FeCl3 are used as iron containing additives. Coal iron enrichment was performed using either directly FeCl3 in vapour phase, or by mixing of coal and additives in decaline or by common grinding of coal and additives under argon. Iron enriched coals were carbonized at 750°C (heating RATE = 5°C min) and activation made with pure steam at 800°C to a burn-off off of 50 wt%. The pore structures of coal chars before and after activation were evaluated on the basis of CO2 and C6H6 sorption at 25°C. A significant development of the microporosity is observed in the iron enriched char before activation and its steam reactivity is also increased. After activation, BET surface area values are increased in presence of iron, and porosity is mainly microporous.  相似文献

Micro-Raman light scattering experiments on PbZrO3 (PZO) single crystal doped with Nb5+ have been investigated. Special attention was paid to the paraelectric (PE) phase in which nominally forbidden first-order Raman spectra were detected at temperatures far above the phase transition TC. Complex Raman spectra were observed in the vicinity of three structural phase transitions. These results mainly from the coexistence of phases with different symmetries in a wide temperature range below TC. The Raman measurements have been compared with dielectric and optical observations and proved that polar nanoregions in a centrosymmetric lattice appear well above TC. It was shown that doping ABO3 perovskites with heterovalent ions like Nb5+ unbalances charge neutrality of the lattice and strongly extends the temperature range of polar regions. The investigations performed point out that in the PE matrix the interaction between electrons and lattice vibrations, as recently suggested for pure PZO, plays an important role.  相似文献
