Gas-phase compost biofilters are extensively used in concentrated animal feeding operations to remove odors and, in some cases, ammonia from air sources. The expected biochemical pathway for these predominantly aerobic systems is nitrification. However, non-uniform media with low oxygen levels can shift biofilter microbial pathways to denitrification, a source of greenhouse gases. Several factors contribute to the formation of anoxic/anaerobic zones: media aging, media and particle structure, air velocity distribution, compaction, biofilm thickness, and moisture content (MC) distribution. The present work studies the effects of media moisture conditions on ammonia (NH3) removal and greenhouse gas generation (nitrous oxide, N2O and methane, CH4) for gas-phase compost biofilters subject to a 100-day controlled drying process. Continuous recordings were made for the three gases and water vapor (2.21-h sampling cycle, each cycle consisted of three gas species, and water vapor, for a total of 10,050 data points). Media moisture conditions were classified into three corresponding media drying rate (DR) stages: Constant DR (wetter media), falling DR, and stable-dry system. The first-half of the constant DR period (0-750 h; MC = 65-52%, w.b.) facilitated high NH3 removal rates, but higher N2O generation and no CH4 generation. At the drier stages of the constant DR (750-950 h; MC = 52-48%, w.b.) NH3 removal remained high but N2O net generation decreased to near zero. In the falling DR stage (1200-1480 h; MC = 44-13%) N2O generation decreased, CH4 increased, and NH3 was no longer removed. No ammonia removal or greenhouse gas generation was observed in the stable-dry system (1500-2500 h; MC = 13%). These results indicate that media should remain toward the drier region of the constant DR (in close proximity to the falling DR stage; MC = 50%, approx.), to maintain high levels of NH3 removal, reduced levels of N2O generation, and nullify levels of CH4 generation.  相似文献

Quantitative methods such as model-based predictive control are known to facilitate the design of strategies to manipulate biological systems. This study develops a sparse-grid-based adaptive model predictive control (MPC) strategy to direct HL60 cellular differentiation. Sparse-grid sampling and interpolation support a computationally efficient adaptive MPC scheme in which multiple data-consistent regions of the model parameter space are identified and used to calculate a control compromise. The algorithm is evaluated in silico with structural model mismatch. Simulations demonstrate how the multiscenario control strategy more effectively manages the mismatch compared to a single scenario approach. Furthermore, the controller is evaluated in vitro to differentiate HL60 cells in both normal and perturbed environments. The controller-derived input sequence successfully achieves and sustains the specified target level of granulocytes when implemented in the laboratory. The results and analysis given here imply that adoption of this experiment planning technique to direct cell differentiation within more complex tissue engineered constructs will require the use of a reasonably accurate mathematical model and an extension of this algorithm to multiobjective controller design.  相似文献

The present study aimed to evaluate in situ the effect of CO2 laser irradiation to control the progression of enamel erosive lesions. Fifty‐six slabs of bovine incisors enamel (5 × 3 × 2.5 mm3) were divided in four distinct areas: (1) sound (reference area), (2) initial erosion, (3) treatment (irradiated or nonirradiated with CO2 laser), (4) final erosion (after in situ phase). The initial erosive challenge was performed with 1% citric acid (pH = 2.3), for 5 min, 2×/day, for 2 days. The slabs were divided in two groups according to surface treatment: irradiated with CO2 laser (λ = 10.6 µm; 0.5 W) and nonirradiate. After a 2‐day lead‐in period, 14 volunteers wore an intraoral palatal appliance containing two slabs (irradiated and nonirradiated), in two intraoral phases of 5 days each. Following a cross‐over design during the first intraoral phase, half of the volunteers immersed the appliance in 100 mL of citric acid for 5 min, 3×/day, while other half of the volunteers used deionized water (control). The volunteers were crossed over in the second phase. Enamel wear was determined by an optical 3D profilometer. Three‐way ANOVA for repeated measures revealed that there was no significant interaction between erosive challenge and CO2 laser irradiation (P = 0.419). Erosive challenge significantly increased enamel wear (P = 0.001), regardless whether or not CO2 laser irradiation was performed. There was no difference in enamel wear between specimens CO2‐laser irradiated and non‐irradiated (P = 0.513). Oxygen can influence the composition and quality of apple must and cider. In this study, the effect of unit operations on oxygen dissolution during apple must processing was investigated and the technological implications discussed. Oxygen dissolution after different apple must processing unit operations, and also the effect of enzymatic browning reaction on dissolved oxygen uptake were analysed. During pressing, the highest and first oxygen dissolution occurred in the must at 6.8 mg/L, on average. The enzymatic browning reaction after 10 min was responsible for an oxygen uptake above 80%. The addition of sulphur dioxide or maceration of crushed apple results in a must with more oxygen content, however the beverage colour is negatively affected. The incorporation of oxygen into the must by bubbling oxygen or the movement of liquid may not be effective owing to consumption by the reaction of residual polyphenol oxidase. Moreover, this can affect the alcoholic fermentation because of the oxidation of nutrients such as vitamins. If the apple must is depectinized, racked and filtered, there will be enough oxygen to obtain a quality cider. This paper presents ProFlex, a distributed data storage protocol for large-scale Heterogeneous Wireless Sensor Networks (HWSNs) with mobile sinks. ProFlex guarantees robustness in data collection by intelligently managing data replication among selected storage nodes in the network. Contrarily to related protocols in the literature, ProFlex considers the resource constraints of sensor nodes and constructs multiple data replication structures, which are managed by more powerful nodes. Additionally, ProFlex takes advantage of the higher communication range of such powerful nodes and uses the long-range links to improve data distribution by storage nodes. When compared with related protocols, we show through simulation that Proflex has an acceptable performance under message loss scenarios, decreases the overhead of transmitted messages, and decreases the occurrence of the energy hole problem. Moreover, we propose an improvement that allows the protocol to leverage the inherent data correlation and redundancy of wireless sensor networks in order to decrease even further the protocol’s overhead without affecting the quality of the data distribution by storage nodes.  相似文献

The aim of this study was to assess differences between apple juices and fermented apple beverages elaborated with fruits from different varieties and at different ripening stages in the aroma profile by using chemometrics. Ripening influenced the aroma composition of the apple juice and fermented apple. For all varieties, senescent fruits provided more aromatic fermented apple beverages. However, no significant difference was noticed in samples made of senescent or ripe fruits of the Lisgala variety. Regarding the juices, ripe Gala apple had the highest total aroma concentration. Ethanal was the major compound identified in all the samples, with values between 11.83 mg/L (unripe Lisgala juice) and 81.05 mg/L (ripe Gala juice). 3-Methyl-1-butanol was the major compound identified in the fermented juices. Principal component analysis (PCA) and hierarchical cluster analysis (HCA) were applied and classified the juices and fermented juices based on physicochemical and aroma profile, demonstrating their applicability as tools to monitor the quality of apple-based products.  相似文献

The use of sonicated melon juice as substrate for Lactobacillus casei growth in cantaloupe melon juice and its effect on product quality were investigated. The survival of L. casei in the fermented juice, the product color, the post-acidification and other quality parameters were evaluated in the refrigerated product (4 °C) for a period of 42 days. Storage pH, color, sugar content, lactic acid concentration, biomass and viable cells count were determined. The caloric value of the product was also evaluated. During the refrigerated storage period, acidification and a slight reduction in the number of viable cells were observed. However, a sufficient number of viable cells were observed to guarantee the beneficial properties from probiotic consumption (>8 log CFU?ml?1) throughout the storage period, for both stocked samples (with and without sugar addition). The product color was also preserved during storage. The developed product resulted in a low caloric product. The caloric value was reduced during the storage period due to sugar consumption by the remaining microorganisms.  相似文献

Leishmania survival inside macrophages depends on factors that lead to the immune response evasion during the infection. In this context, the metabolic scenario of the host cell–parasite relationship can be crucial to understanding how this parasite can survive inside host cells due to the host’s metabolic pathways reprogramming. In this work, we aimed to analyze metabolic networks of bone marrow-derived macrophages from C57BL/6 mice infected with Leishmania amazonensis wild type (La-WT) or arginase knocked out (La-arg), using the untargeted Capillary Electrophoresis-Mass Spectrometry (CE-MS) approach to assess metabolomic profile. Macrophages showed specific changes in metabolite abundance upon Leishmania infection, as well as in the absence of parasite-arginase. The absence of L. amazonensis-arginase promoted the regulation of both host and parasite urea cycle, glycine and serine metabolism, ammonia recycling, metabolism of arginine, proline, aspartate, glutamate, spermidine, spermine, methylhistidine, and glutathione metabolism. The increased L-arginine, L-citrulline, L-glutamine, oxidized glutathione, S-adenosylmethionine, N-acetylspermidine, trypanothione disulfide, and trypanothione levels were observed in La-WT-infected C57BL/6-macrophage compared to uninfected. The absence of parasite arginase increased L-arginine, argininic acid, and citrulline levels and reduced ornithine, putrescine, S-adenosylmethionine, glutamic acid, proline, N-glutamyl-alanine, glutamyl-arginine, trypanothione disulfide, and trypanothione when compared to La-WT infected macrophage. Moreover, the absence of parasite arginase leads to an increase in NO production levels and a higher infectivity rate at 4 h of infection. The data presented here show a host-dependent regulation of metabolomic profiles of C57BL/6 macrophages compared to the previously observed BALB/c macrophages infected with L. amazonensis, an important fact due to the dual and contrasting macrophage phenotypes of those mice. In addition, the Leishmania-arginase showed interference with the urea cycle, glycine, and glutathione metabolism during host–pathogen interactions.  相似文献

This paper presents the characterization of single-mode waveguides for 980 and 1550 nm wavelengths. High quality planar waveguide structure was fabricated from Y1 − xErxAl3(BO3)4 multilayer thin films with x = 0.02, 0.05, 0.1, 0.3, and 0.5, prepared through the polymeric precursor and sol-gel methods using spin-coating. The propagation losses of the planar waveguides varying from 0.63 to 0.88 dB/cm were measured at 632.8 and 1550 nm. The photoluminescence spectra and radiative lifetimes of the Er3+ 4I13/2 energy level were measured in waveguiding geometry. For most samples the photoluminescence decay was single exponential with lifetimes in between 640 μs and 200 μs, depending on the erbium concentration and synthesis method. These results indicate that Er doped YAl3(BO3)4 compounds are promising for low loss waveguides.  相似文献
