The effect of starch conversion on the susceptibility of potato granules to α‐amylase was studied by direct sampling at different pasting times corresponding to different points on the RVA profile of a 6.4% (w/w) suspension of starch in distilled water. Native granules showed high resistance to α‐amylase with 8.6 ± 0.4% digestibility for a 6 h incubation period with the enzyme. When the suspension was heated to 60 °C, the digestibility increased to 53.5 ± 0.7% although, at this temperature, there was still no noticeable increase in the measured viscosity (≤0.040 Pa · s). The material sampled after a pasting time corresponding to the RVA peak viscosity showed a digestibility of 88.4 ± 0.5%. This suggested, owing to the expected retrogradation of amylose on cooling, the quasi‐total susceptibility of amylopectin to enzymatic digestion at this pasting stage. The effect of ions on the swelling of potato starch was used to assess whether the decrease of the swelling of the granules in the presence of NaCl was paralleled by an increase in resistance to α‐amylase. A small (∼6.1%) but significant decrease in the digestibility of pasted starch was observed in the presence of salt. Finally, the effect of the retrogradation of the amylopectin fraction on its digestibility was assessed in extruded potato starch ribbons containing 35% (w/w) water and stored at different temperatures. After 14 days of storage, the digestibility decreased from 77.0 ± 0.9% in the freshly extruded samples to between 28.0 ± 1.7% and 42.1 ± 0.3%, depending on the storage temperature. This suggested a measurable difference in the α‐amylase susceptibility between the A and B polymorphs of retrograded amylopectin.  相似文献

This study was aimed to evaluate the performance of near-infrared reflectance spectroscopy (NIRS) in the analysis of the oil composition for fatty acids like oleic (C18:1), linoleic (C18:2) and linolenic (C18:3) in zero-erucic acid rapeseed (Brassica napus L.). Intact-seed samples of 1094 lines from a breeding programme for high-oleic acid rapeseed were analyzed by both NIRS and gas chromatography (GC). Previously developed calibration equations were initially used for NIRS analyses. The accuracy of NIRS was considerably improved by including some samples of the actual breeding population into the original calibration set and developing new calibration equations. The inclusion of twenty randomly selected samples led to a reduction of the standard error of performance (SEP) from 2.6% to 1.9% for oleic, from 3.8% to 2.0% for linoleic, and from 1.1% to 0.9% for linolenic acid. The application of the new equations to the remaining population of 1074 samples resulted in coefficients of correlation between NIRS and GC values of 0.95 for oleic, 0.92 for linoleic, and 0.90 for linolenic acid. Furthermore, the effectiveness of a selection for high oleic, high linoleic, or low linolenic acid content based on NIRS data was demonstrated. The results of this study will help potential users to choose the optimal selection strategy in routine analysis of C18 unsaturated fatty acids by NIRS within a breeding programme.  相似文献

Hypoxia-regulated protein carbonic anhydrase IX (CA IX) is up-regulated in different tumor entities and correlated with poor prognosis in breast cancer patients. Due to the radio- and chemotherapy resistance of solid hypoxic tumors, derivatives of betulinic acid (BA), a natural compound with anticancer properties, seem to be promising to benefit these cancer patients. We synthesized new betulin sulfonamides and determined their cytotoxicity in different breast cancer cell lines. Additionally, we investigated their effects on clonogenic survival, cell death, extracellular pH, HIF-1α, CA IX and CA XII protein levels and radiosensitivity. Our study revealed that cytotoxicity increased after treatment with the betulin sulfonamides compared to BA or their precursors, especially in triple-negative breast cancer (TNBC) cells. CA IX activity as well as CA IX and CA XII protein levels were reduced by the betulin sulfonamides. We observed elevated inhibitory efficiency against protumorigenic processes such as proliferation and clonogenic survival and the promotion of cell death and radiosensitivity compared to the precursor derivatives. In particular, TNBC cells showed benefit from the addition of sulfonamides onto BA and revealed that betulin sulfonamides are promising compounds to treat more aggressive breast cancers, or are at the same level against less aggressive breast cancer cells.  相似文献

The interplay of structural and magnetic properties of magnetic shape memory alloys is closely related to their composition. In this study the influence of the valence electron concentration on the tetragonal transformation in Ni2Mn1 + xZ1 ? x (Z = Ga, In, Sn, Sb) and Co2Ni1 + xGa1 ? x is investigated by means of ab initio calculations. While the type of magnetic interaction is different for the two series, the trends of the total energy changes under a tetragonal transformation are very similar. We find that tetragonal structures become energetically preferred with respect to the cubic one as the valence electron concentration e/a is increased regardless of the system under consideration. In particular, the energy difference between the austenite and martensite structures increases linearly with e/a, which is in part responsible for the linear increase of the matensite transformation temperature. The substitution of nickel by platinum increases even further the transformation temperature.  相似文献

There are dozens of studies made and ongoing related to wind integration. However, the results are not easy to compare. IEA WIND R&D Task 25 on ‘Design and Operation of Power Systems with Large Amounts of Wind Power’ collects and shares information on wind generation impacts on power systems, with analyses and guidelines on methodologies. In the state‐of‐the‐art report (October, 2007), and the final report of the 3 years period (July, 2009) the most relevant wind power grid integration studies have been analysed especially regarding methodologies and input data. Several issues that impact on the amount of wind power that can be integrated have been identified. Large balancing areas and aggregation benefits of wide areas help in reducing the variability and forecast errors of wind power as well as help in pooling more cost effective balancing resources. System operation and functioning electricity markets at less than day‐ahead time scales help reduce forecast errors of wind power. Transmission is the key to aggregation benefits, electricity markets and larger balancing areas. Best practices in wind integration studies are described. There is also benefit when adding wind power to power systems: it reduces the total operating costs and emissions as wind replaces fossil fuels and this should be highlighted more in future studies. A better and regular control of the production chain of fresh fruits and vegetables is necessary, because a contamination of the product by human- and phyto-pathogenic microorganisms may result in high losses during storage and poses a threat to human health. Therefore, detailed knowledge about the occurrence and the diversity of microorganisms within single processing steps is required to allow target-oriented produce safety control. Recently, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) was successfully used to identify bacterial colonies. Extraction is a downstream process option in biobased processes. Because knowledge of phase‐separation behavior is essential for designing efficient separation processes, this study investigates the settling and coalescence behavior of biobased extraction systems by using a standard laboratory‐scale settling cell. The influence of different buffer media and Escherichia coli cells on coalescence was determined for the reactive extraction of hexane‐1,6‐diamine with isostearic acid and di(2‐ethylhexyl)phosphoric acid by using kerosene and oleyl alcohol as diluents. As a result, an increasing pH value of the buffer significantly increases the settling time. The presence of E. coli cells hinders phase separation of the investigated systems, in particular, with dispersed organic phases.  相似文献
