Hybrid MalE–GVP is a bifunctional protein in vitro sinceit binds maltose as protein MalE of Escherichia coli and sinceit is dimeric and specifically binds single-stranded DNA asprotein GVP of phage M13. The oxidation rate of a unique cysteineresidue was used to compare the stabilities of GVP in its freeand hybrid forms, under conditions where MalE was either foldedor unfolded by a denaturing agent. The results showed that boththe covalent link and tertiary non-covalent interactions betweenMalE and GVP destabilized GVP in MalE–GVP. To test whetherGVP had identical structures in its free and hybrid forms, mutationswere used as local conformational probes. The effects of thesemutations on the capabilities of MalE–GVP to dimerizeand to bind single-stranded DNA were assayed in vitro. Theywere compatible with the effects of the same mutations on theglobal activity of free GVP in vivo and with the effects thatcould be predicted from the known data on free GVP, in particularits crystal structure. Thus, one partner of a hybrid proteincan be destabilized by the other partner while maintaining itsstructural and functional characteristics.  相似文献

The Fabry?Perot (FP) method of semiconductor laser gain measurement, first proposed by Hakki and Paoli (1975), is widely used. It is based on the measurement of the FP resonances excited by spontaneous emission. Its validity rests on the assumption that a single mode is significant. We show, using a simplified laser model, that this assumption is valid only when the power mirror reflectivity is very small, or near the laser oscillating frequency. For example, the error is in the order of 20% when the power facet reflectivities are equal to 37% and the modal gain is unity. These results apply to both index-guided and gain-guided lasers.  相似文献

This paper presents a compact model for MOS transistor mismatch. The mismatch model uses the carrier number fluctuation theory to account for the effects of local doping fluctuations along with an accurate and compact dc MOSFET model. The resulting matching model is valid for any operation condition, from weak to strong inversion, from the linear to the saturation region, and allows the assessment of mismatch from process and geometric parameters. Experimental results from a set of transistors integrated on a 0.35 /spl mu/m technology confirm the accuracy of our mismatch model under various bias conditions.  相似文献

The damage growth in a softening interface connected to an elastic block is analysed. The elastic block, assumed to be infinite, is modelled as a two-dimensional continuum and the interface is one-dimensional with a constitutive response which follows a scalar damage model. The solution technique is based on the equilibrium of the interfacial forces resulting from the deformation of the elastic block and from the interface constitutive response. The interface failure process is compared to that of a hierarchical model which was obtained analytically (Delaplace et al., 1998). The two are found to be similar, without an internal length scaling the distribution of damage at the inception of macro-cracking. Finally, scale effects on the occurrence of bifurcation and instability are considered. It is shown that bifurcation may occur prior to or after the limit point under displacement control, depending on the elastic block height or stiffness. Self-routing of 100-Mb/s data through a photonic switch is demonstrated. Individual data bits are subencoded into one of 124 destination addresses using a pulse-interval technique with 80-ps pulses. At the switch, optical processing is used to read the address of an incoming bit and to route that bit to the correct switch output without the aid of an external clock. Although the pulse-interval technique is presented in a bit-switching context, it can readily be extended to a packet-switching environment  相似文献

Soils are important components of ecosystem and their functioning is of great importance for human well-being. Describing, understanding, foreseeing, and controlling biological activities and functions in soil in the context of global change are major objectives for scientists. Modelling soil bioturbation by soil engineers is of great importance although it is faced with the difficulty to describe the soil physical environment. Creating a model of a soil leads to complexity problems: a soil is a multi-scale heterogeneous, three-dimensional and dynamic environment that cannot be modelled and simulated without defining a suitable and optimized representation and taking assumptions about the studied system. An approach based on fractal theory (often used in soil sciences) was chosen to model such a real complex environment; it was integrated into a multi-agent system (MAS). MAS allows to simulate situated agents (earthworms) in an virtual world. The originality of this present MAS is that it is based on a dynamic environment which builds itself, on demand, according to an abstract canvas tree and agent movements. The aim of this paper is to present this approach and its originality, and to describe the model and the simulator. A theoretical view of the approach is given and applied to a case study: the impact of earthworms on soil structure and organic matter dynamics.  相似文献

The current classification of cutaneous melanoma was developed in 1972 and revised in 1982. Since that time new concepts and terminology have evolved that require consideration of a further revision. This paper reviews some of the concepts that will form part of that process. Regional meetings of interested parties have been held to review the classification and there will be an open meeting on the topic at the 1997, 4th World Conference on Melanoma in Sydney, Australia. A questionnaire is included that will allow the interested reader to provide comments on the topic.  相似文献

In situ X-ray diffraction was used to study the oxide formation on AISI 316L stainless steel (SS) specimens during isothermal oxidation at 900 °C in air. Results were compared with those obtained on AISI 304 SS to determine the role of molybdenum on the oxidation process for the AISI 316L SS specimens. Our results show that molybdenum plays a major protective role during steel oxidation. This element is found in a NiMoO4 phase at the internal oxide–metal interface. The high molybdenum content of the alloy hinders the outward diffusion of iron and leads to a lower growth rate and better scale adherence. The oxide scale is then composed of Cr2O3 with a small amount of Mn1.5Cr1.5O4 at the external interface. The improved scale adherence appears to be due to a keying effect at the scale/alloy interface promoted by molybdenum.  相似文献

Coplanar waveguide (CPW) and thin film microstrip (TFMS) lines integrating porous ultra low-k as inter-metal dielectric layers (k = 2.5) and copper as metal, are for the first time experimentally measured up to 110 GHz and under different temperature conditions, up to 200 °C. The extracted attenuation and propagation coefficients of those transmission lines are compared to simulations performed with MAGWEL software, a frequency domain 3-D Maxwell solver. Based on the characterization results some guidelines related to interconnect design are presented for future applications.  相似文献
