On the influence of building design,occupants and heat waves on comfort and greenhouse gas emissions in naturally ventilated offices. A study based on the EN 15251 adaptive thermal comfort model in Athens,Greece

According to the Intergovernmental Panel on Climate Change the buildings sector has the largest mitigation potential for CO₂ emissions. Especially in office buildings, where internal heat loads and a relatively high occupant density occur at the same time with solar heat gains, overheating has become a common problem. In Europe the adaptive thermal comfort model according to EN 15251 provides a method to evaluate thermal comfort in naturally ventilated buildings. However, especially in the context of the climate change and the occurrence of heat waves within the last decade, the question arises, how thermal comfort can be maintained without additional cooling, especially in warm climates. In this paper a parametric study for a typical cellular naturally ventilated office room has been conducted, using the building simulation software EnergyPlus. It is based on the Mediterranean climate of Athens, Greece. Adaptive thermal comfort is evaluated according to EN 15251. Variations refer to different building design priorities, and they consider the variability of occupant behaviour and internal heat loads by using an ideal and worst case scenario. The influence of heat waves is considered by comparing measured temperatures for an average and an exceptionally hot year within the last decade. Since the use of building controls for shading affects thermal as well as visual comfort, daylighting and view are evaluated as well. Conclusions are drawn regarding the influence and interaction of building design, occupants and heat waves on comfort and greenhouse gas emissions in naturally ventilated offices, and related optimisation potential.

Effect of dimorphic regulation on heterologous glucose oxidase production by Mucor circinelloides

The production of Aspergillus niger glucose oxidase (GOX) and native amylase by the recombinant M. circinelloides KFA199 strain under conditions of dimorphic growth was investigated. The recombinant KFA199 strain was compared to its parental ATCC 1216b strain and a wild‐type CBS 232.29 strain under similar morphology‐controlled conditions. Cultivation in Vogel's medium supplemented with ergosterol/Tween‐80 and sparged with nitrogen gas was most suitable for yeast‐like biomass production under anaerobic conditions. Anaerobic growth was characterized by high levels of ethanol formation and linear growth rates of 0.24–0.05/h, indicating metabolic stress. Subsequent to anaerobic growth, cultures were shifted to aerobic conditions to induce aerobic mycelial growth. GOX produced by the recombinant KFA199 after the shift to aerobic conditions was poorly secreted and accumulated intracellularly to 0.56 U/ml_culture. Amylase production by the KFA199, ATCC12b and CBS 232.29 strains was determined during growth on starch after the shift to aerobic culture. Growth‐associated amylase production by the ATCC 1216b (0.63 U/ml_culture) and wild‐type CBS 232.29 (0.33 U/ml_culture) strains was substantially higher than by the recombinant KFA199 strain (0.07 U/ml_culture), which may be related to the leucine auxotrophy of the transformation host, or genetic changes induced during transformation of the KFA199 strain. A Predictive Model for the Growth/No Growth Boundary of Zygosaccharomyces bailii at 7?°C and Conditions Mimicking Acidified Sauces

The spoilage potential of Zygosaccharomyces bailii has been widely recognized within the food industry. However, few data are available on the growth characteristics of this yeast at low temperature and in the absence of chemical preservatives. In this study, the growth/no growth boundary of Z. bailii was defined at refrigeration temperature (7?°C) and at conditions relevant to high-sugar, low-pH foods (such as ketchup and salad dressing), i.e. pH?3.0?C5.0 (five levels), a _w 0.93?C0.97 (five levels) and acetic acid concentration 0?C2.5% (v/v; six levels). Yeast growth was followed during 90?days by optical density measurements, and logistic regression models were used to describe the data. Acetic acid had a significant effect on the relation between a _w and NaCl concentration and this interaction had important consequences for the model development. When data were modelled as a function of a _w or NaCl concentration, a stimulatory effect by acetic acid was observed. In contrast, as a function of (toxic) Na⁺ ions, no evidence was found of such a phenomenon. These results indicate that in cases where the relationship between a _w and solute concentration is not straightforward such as in the presence of acid preservatives, one must be critical towards the interpretation of data and correspondingly, the development of predictive models. On a practical note, the developed models, especially the one incorporating Na⁺ ions, may be used (1) to assess the stability of shelf-stable acidified foods stored under chilled conditions after opening or (2) to formulate new additive-free products intended for storage at 7?°C.     

A review of occupant control on natural ventilation

According to IPCC (Intergovernmental Panel on Climate Change), the largest use of energy in commercial buildings is space heating in colder climates and air conditioning in hot climates. In Europe, the Directive on the energy performance of buildings EPBD (European Energy Performance of Buildings Directive) [1] provides a framework for national building performance regulations and calculation procedures. However, there are often large discrepancies between calculated and measured energy performance of buildings. One main reason is the behaviour of occupants, which is often not reflected in calculation models.This paper presents a literature review of various parameters influencing the effectiveness of occupant controlled natural ventilation. Additionally possibilities to implement these influences into calculation procedures/building simulation and adaptive thermal comfort evaluation are discussed.     

Persistence and distribution of intravenously injected DNA in blood and organs of Atlantic salmon (Salmo salar L.)

In a recent study it was reported that DNA could be taken up from the feed and transferred to the blood of Atlantic salmon (Nielsen CR, Berdal KG, Bakke-McKellep AM, Holst-Jensen A, Eur Food Res Tech, 2005). In the present study the transport in blood and accumulation in some organs of intravenously injected DNA was studied. Intravenous injection was used as an alternative to feeding to ensure that the quantity of target DNA would permit a quantitative study of the distribution and degradation of the DNA in the fish. Three DNA fragments of different length were administered by injection to 40 salmon, each in a copy number of 10¹⁰. Blood and tissues were sampled at different time intervals after injection. To ensure the use of a sensitive and reliable detection system, real-time PCR methods with TaqMan chemistry were applied in this study. Target DNA was observed in blood and all investigated tissues, with peak levels observed at 0.5–1 h after injection. Some tissue samples were positive for the target DNA up to 24 h after injection. Results of the present study indicate that dietary DNA taken up in the blood may be transported to organs like liver and kidney, as well as muscle and gonads.     

New advances in automatic disposable rotating cartridge skin staplers

The anti-inflammatory effects of the amide local anaesthetics lidocaine and bupivacaine were evaluated in vitro by examination of the metabolic activation and secretory responses of human polymorphonuclear granulocytes (PMNGs) and mononuclear cells. Pretreatment with lidocaine or bupivacaine had a dose-dependent inhibitory effect on PMNG luminol-amplified chemiluminescence stimulated by bovine serum albumin (BSA)/anti-BSA immune complexes (IC) or by serum-opsonized zymosan (SOZ) particles. Both lidocaine and bupivacaine inhibited the release of the inflammatory mediators leukotriene B4 (LTB4) and interleukin-1 (IL-1) evaluated by radioimmunoassay (RIA). Pretreatment of suspended PMNGs and monocytes with the anaesthetics caused a marked inhibition of LTB4 release when the cells were stimulated with SOZ. In short-term (24 h) cultures of mononuclear cells the addition of lidocaine or bupivacaine reduced, in a dose-dependent manner, the level of IL-1 detected after stimulation with lipopolysaccharide (LPS). In all three assays (chemiluminescence, LTB4 and IL-1 RIA) bupivacaine was found to be more potent than lidocaine. The present results show that amide local anaesthetics have marked suppressive effects on the metabolic activation and secretory functions of leukocytes stimulated by different agonists. Although the detailed mechanisms for these effects are not known, they may explain part of the potent anti-inflammatory actions of local anaesthetics previously described in vivo.     

Severe burn injury from recreational gasoline use

We describe a novel reciprocal translocation, t(14;15)(q11;q24), as the sole cytogenetic aberration in a congenital mesoblastic nephroma. The tumor was predominantly of "classic" morphology, but with small cellular islands of larger cells indicating early transformation to the cellular type. This is the first report of a reciprocal translocation as the sole anomaly in a congenital mesoblastic nephroma.
