In last few years, special attention has been given to food-induced allergies, in which hazelnut allergy is highlighted. Hazelnut is one of the most commonly consumed tree nuts, being largely used by the food industry in a variety of processed foods. It has been regarded as a food with potential health benefits, but also as a source of allergens capable of inducing mild to severe allergic reactions in sensitized individuals. Considering the great number of reports addressing hazelnut allergens, with an estimated increasing trend, this review intends to assemble all the relevant information available so far on the following main issues: prevalence of tree nut allergy, clinical threshold levels, molecular characterization of hazelnut allergens (Cor a 1, Cor a 2, Cor a 8, Cor a 9, Cor a 10, Cor a 11, Cor a 12, Cor a 14, and Cor a TLP) and their clinical relevance, and methodologies for detection of hazelnut allergens in foods. A comprehensive overview of the current data about the molecular characterization of hazelnut allergens is presented, relating to biochemical classification and biological function with clinical importance. Recent advances in hazelnut allergen detection methodologies are summarized and compared, including all the novel protein-based and DNA-based approaches.  相似文献

Electrophoretic and chromatographic techniques were used to determine water‐soluble peptide profiles aiming to identify the adulteration of buffalo milk mozzarella cheese by the addition of cow's milk. Thus, cheeses were produced with contents of cow's milk varying from 0% to 100%, and the peptides extracted after production and after 20 days of refrigeration. Polyacrylamide gel electrophoresis under denaturing conditions in the presence of sodium dodecyl sulphate (SDS‐PAGE) identified a potential peptide marker of exclusively bovine origin with a size of about 21 kDa for the addition of cow's milk above 30%. Reverse‐phase high‐performance liquid chromatography (RP‐HPLC) indicated the existence of two potential peptides present in higher concentrations in buffalo milk and one exclusive for cow's milk, the latter making it possible to estimate the addition of cow's milk to buffalo milk. Six commercial brands of buffalo mozzarella cheese were evaluated, and indications of adulteration found in four of them.  相似文献

Fermented whey dairy beverages are dairy products obtained by fermentation from a mixture of milk and whey. These beverages have important health benefits, which could be improved with the addition of probiotic cultures. This study assessed the protective effect of the cosupplementation of a probiotic culture (Lactobacillus casei 01) with a fermented whey dairy beverage against infection by Salmonella enterica ssp. enterica serovar Typhimurium in a murine model. Two fermented whey dairy beverages were prepared: conventional (FWB; starter culture) and probiotic (PFWB; starter and probiotic cultures). In the first set of experiments, Balb/C female mice were treated with FWB or PFWB, challenged with Salmonella Typhimurium, and analyzed for clinical signs, weight loss, and mortality for 20 d postinfection. In the second set of experiments, mice were treated with FWB or PFWB, challenged with Salmonella Typhimurium, and killed on d 10 postinfection. The liver, colon, and ileum were used for myeloperoxidase, eosinophil peroxidase, and histological analysis and translocation to the liver. The contents from the small intestine were used for secretory IgA determination. The FWB treatment showed a better effect on animal survival (70%), translocation of the pathogen to the liver (2 out of 10), histopathology (fewer lesions), and inflammation than PFWB, which presented 50% animal survival, translocation in 5 out of 10 animals, and higher lesions. The control group presented 40% animal survival, translocation in 6 out of 10 animals, and severe lesions. Therefore, FWB was deemed to have a greater protective effect against Salmonella Typhimurium infection in the murine model compared with PFWB.  相似文献

Our objective was to evaluate how sampling strategies (i.e., how many cows to sample and which animals to include) used in 4 dairy cattle welfare assessment programs affect the classification of dairy farms relative to thresholds of acceptability for animal-based measures. We predicted that classification performance would improve when more cows were sampled and when selecting from all lactating cows versus when some pens were excluded. On 38 freestall farms, we assessed all 12,375 cows for lameness, injuries on the tarsal (hock) and carpal joints, and body condition score and calculated the farm-level prevalence for each measure. Based on approaches used in the industry, we evaluated 6 sampling strategies generated using formulas with precision (d) of 15, 10, or 5% applied to either a single high-producing pen or all lactating cows; an additional sample was included with d = 10% applied to the entire herd, selecting lactating cows in proportion to their representation in the herd. For each sampling strategy, cow records were selected randomly (in 10,000 replicates) to calculate prevalence. The strategy of assessing all cows in the high-producing pen was also compared. Farms were classified as meeting (below) or failing to meet (above) thresholds of ≤15% moderate lameness; ≤20% moderate carpal or hock injuries; <10, <5, and ≤1% severe lameness; or injuries on the carpus or hock; and <5, <3, <1, or 0% thin cows. For each measure and threshold, we calculated pooled percent agreement, kappa, sensitivity, specificity, and positive and negative predictive value for each sampling strategy using true prevalence as the gold standard for herd classification. Across measures and thresholds, classification performance increased with the number of cows sampled [i.e., when narrower precision values (d = 5 vs. 10 vs. 15%) were used in the sample size calculation]. Because narrower precision values can dramatically increase sample size, assessment programs may need to consider both feasibility and the degree of misclassification they will accept. Applying the formula directly to lactating cows performed better than applying it to the entire herd and then selecting lactating cows in proportion to their representation in the herd. Farm classifications were similar whether cows in the hospital pen were included or excluded from the sample. Selecting all cows from the high-producing pen resulted in classifications similar to when including all lactating cows, suggesting that assessing cows from the high-producing pen may serve as an acceptable proxy for all lactating cows on the farm.  相似文献

Pistachio nut (Pistacia vera) is highly appreciated for its organoleptic characteristics and potential health benefits. However, this tree nut is also responsible for triggering moderate to severe IgE-mediated reactions in allergic individuals. Currently, pistachio nut allergy has gained some special attention, mainly due to its intrinsic relation with cashew nut allergy. Like for other nuts, the prevalence of pistachio nut allergy seems to be increasing at a global scale. Until now, there are five allergenic proteins officially listed for pistachio nut (Pis v 1, Pis v 2, Pis v 3, Pis v 4 and Pis v 5). Relevant data on their biochemical classification has become available, enabling establishing a correlation with the respective clinical symptoms. The establishment of an effective allergen risk assessment is a key issue for the food industry, policy makers and regulatory agencies. Thus, the availability of fast, specific and sensitive methods to detect trace amounts of allergens in processed foods is crucial. In the specific case of pistachio nut, there are some protein- and DNA-based methods for its detection/quantification in foods, which can aid to verify label information. Accordingly, all relevant research advances on this topic were summarised, updated and critically discussed in this review.  相似文献

Dairy cows display a partial preference for being outside, but little is known about what aspects of the outdoor environment are important to cows. The primary aim of this study was to test the preference of lactating dairy cattle for pasture versus an outdoor sand pack during the night. A secondary aim was to determine whether feeding and perching behavior changed when cows were provided outdoor access. A third objective was to investigate how the lying behavior of cows changed when given access to different outdoor areas. Ninety-six lactating pregnant cows were assigned to 8 groups of 12 animals each. After a baseline phase of 2 d in which cows were kept inside the freestall barn, cows were habituated to the outdoor areas by providing them access to each of the 2 options for 24 h. Cows were then given access, in random order by group, to either the pasture (pasture phase) or the sand pack (sand phase). As we tested the 2 outdoor options using space allowances consistent with what would be practical on commercial dairy farms, the space provided on pasture was larger (21,000 m2) than that provided on the sand pack (144 m2). Cows were tested at night (for 2 nights in each condition), from 2000 h until morning milking at approximately 0800 h, as preference to be outdoors is strongest at this time. During the next 3 nights cows were given access to both outside options simultaneously (choice phase). Feeding and perching behaviors were recorded when cows were indoors during the day and night periods. Lying behavior was automatically recorded by HOBO data loggers (Onset, Bourne, MA). Cows spent more time outside in the pasture phase (90.0 ± 5.9%) compared with the sand phase (44.4 ± 6.3%). When provided simultaneous access to both options, cows spent more time on pasture than on the sand pack (90.5 ± 2.6% vs. 0.8 ± 0.5%, respectively). Time spent feeding indoors during the day did not change regardless of what type of outdoor access was provided, but there was a decline in perching during the day when cows were provided access to either outdoor option at night. Lying time in the pasture phase was lower than in the baseline or sand phase. During the nighttime, lying time outside was not different between the sand (55.4 ± 7.9%) and pasture (52.0 ± 7.4%) phases. In summary, cows spent a considerable amount of time outside during the night when given the opportunity and showed a preference for a large pasture versus a small sand pack as an outdoor area.  相似文献

ABSTRACT

Biofortification aims to improve the micronutrient concentration of staple food crops through the best practices of breeding and modern biotechnology. However, increased zinc and iron concentrations in food crops may not always translate into proportional increases in absorbed zinc (Zn) and iron (Fe). Therefore, assessing iron and zinc bioavailability in biofortified crops is imperative to evaluate the efficacy of breeding programs. This review aimed to investigate the advantages and limitations of in vitro and in vivo methods of iron and zinc bioavailability evaluation in the assessment of biofortification program effectiveness. In vitro, animal and isotopic human studies have shown high iron and zinc bioavailability in biofortified staple food crops. Human studies provide direct knowledge regarding the effectiveness of biofortification, however, human studies are time consuming and are more expensive than in vitro and animal studies. Moreover, in vitro studies may be a useful preliminary screening method to identify promising plant cultivars, however, these studies cannot provide data that are directly applicable to humans. None of these methods provides complete information regarding mineral bioavailability, thus, a combination of these methods should be the most appropriate strategy to investigate the effectiveness of zinc and iron biofortification programs.  相似文献
