Ten-eleven translocation (TET) proteins, a family of Fe2+- and 2-oxoglutarate-dependent dioxygenases, are involved in DNA demethylation. They also help regulate various cellular functions. Three TET paralogs have been identified (TET1, TET2, and TET3) in humans. This study focuses on the evolution of mammalian TET genes. Distinct patterns in TET1 and TET2 vs. TET3 were revealed by codon-based tests of positive selection. Results indicate that TET1 and TET2 genes have experienced positive selection more frequently than TET3 gene, and that the majority of codon sites evolved under strong negative selection. These findings imply that the selective pressure on TET3 may have been relaxed in several lineages during the course of evolution. Our analysis of convergent amino acid substitutions also supports the different evolutionary dynamics among TET gene subfamily members. All of the five amino acid sites that are inferred to have evolved under positive selection in the catalytic domain of TET2 are localized at the protein’s outer surface. The adaptive changes of these positively selected amino acid sites could be associated with dynamic interactions between other TET-interacting proteins, and positive selection thus appears to shift the regulatory scheme of TET enzyme function.  相似文献

Concerns about greenhouse gas emissions have increased research efforts into alternatives in bio-based processes. With regard to transport fuel, bioethanol and biodiesel are still the main biofuels used. It is expected that future production of these biofuels will be based on processes using either non-food competing biomasses, or characterised by low CO2 emissions. Many microorganisms, such as microalgae, yeast, bacteria and fungi, have the ability to accumulate oils under special culture conditions. Microbial oils might become one of the potential feed-stocks for biodiesel production in the near future. The use of these oils is currently under extensive research in order to reduce production costs associated with the fermentation process, which is a crucial factor to increase economic feasibility. An important way to reduce processing costs is the use of wastes as carbon sources. The aim of the present review is to describe the main aspects related to the use of different oleaginous microorganisms for lipid production and their performance when using bio-wastes. The possibilities for combining hydrogen (H2) and lipid production are also explored in an attempt for improving the economic feasibility of the process.  相似文献

Inflammation and haemorrhage are the main characteristics of tissue injury in botropic envenomation. Although some studies have shown that anti-venom prevents systemic reactions, it is not efficient in preventing tissue injury at the site of the bite. Therefore, this work was undertaken to investigate the anti-inflammatory effects of the methanolic extract and fractions from D. elliptica and to evaluate the role of matrix metalloproteinases (MMPs) in this process. Effects of the extract and fractions from D. elliptica were evaluated using a carrageenan-induced paw oedema model in rats, and leukocyte rolling was visualized by intravital. The quantification of MMPs activities (MMP-2 and MMP-9) extracted from the dermis of mice treated with extract and fractions alone or incubated with venom was determined by zymographic analyses. Our results show that intraperitoneal (i.p.) injection of fractions significantly reduced paw oedema after the carrageenan challenge. Treatment with the tannins fraction also resulted in considerable inhibition of the rolling of leukocytes and this fraction was able to decrease the activation of MMP-9. These results confirmed the anti-inflammatory activity of the methanolic extract and tannins fraction of D. elliptica and showed that the dermonecrosis properties of B. jararaca venom might be mediated through the inhibition of MMP-9 activity.  相似文献

Monitoring partial vapor pressure in the freeze-drying chamber is a cheap, global, and non-intrusive way to assess the end of the primary drying stage. Most existing dynamic freeze-drying models which predict this partial pressure describe mass transfer between the product and the condenser via a mass transfer resistance or a mass transfer coefficient. Experimental evidence suggests that such models can be significantly in error for some values of the sublimation flux, leading to physically inconsistent predictions and possibly incorrect assessment of primary drying termination, with potential risk of product damage if moving to secondary drying and increasing shelf temperature while some ice is still present. Assuming a binary gas transport model for vapor and inert gas leads to improved and consistent predictions and explains the apparent variation of the mass transfer resistance with total pressure, shelf temperature, and product sublimation area.  相似文献

To better understand N2 formation in the selective catalytic reduction of NO x by propene under oxidative atmosphere, we studied the catalytic behavior of bimetallics. Surface segregation phenomena may provoke geometric and electronic modifications of the catalytic active sites. To explain our results we invoked: (i) the facile way to oxidize the bimetallic aggregates, (ii) the presence of atoms with low coordination number able to be oxidized first and (iii) an eventual formation of like vic-diNO x species on such bimetallics leading to N2 formation.  相似文献

Interactions between insects and glucosinolate-containing plant species have been investigated for a long time. Although the glucosinolate–myrosinase system is believed to act as a defense mechanism against generalist herbivores and fungi, several specialist insects use these secondary metabolites for host plant finding and acceptance and can handle them physiologically. However, sequestration of glucosinolates in specialist herbivores has been less well studied. Larvae of the turnip sawfly Athalia rosae feed on several glucosinolate-containing plant species. When larvae are disturbed by antagonists, they release one or more small droplets of hemolymph from their integument. This reflex bleeding is used as a defense mechanism. Specific glucosinolate analysis, by conversion to desulfoglucosinolates and analysis of these by high-performance liquid chromatography coupled to diode array UV spectroscopy and mass spectrometry, revealed that larvae incorporate and concentrate the plant's characteristic glucosinolates from their hosts. Extracts of larvae that were reared on Sinapis alba contained sinalbin, even when the larvae were first starved for 22 hr and, thus, had empty guts. Hemolymph was analyzed from larvae that were reared on either S. alba, Brassica nigra, or Barbarea stricta. Leaves were analyzed from the same plants the larvae had fed on. Sinalbin (from S. alba), sinigrin (B. nigra), or glucobarbarin and glucobrassicin (B. stricta) were present in leaves in concentrations less than 1 mol/g fresh weight, while the same glucosinolates could be detected in the larvae's hemolymph in concentrations between 10 and 31 mol/g fresh weight, except that glucobrassicin was present only as a trace. In larval feces, only trace amounts of glucosinolates (sinalbin and sinigrin) could be detected. The glucosinolates were likewise found in freshly emerged adults, showing that the sequestered phytochemicals were transferred through the pupal stage.  相似文献

A microwave protocol to extract lipophilic substances from tissue was modified to extract essential oils (EOs) from plant tissue and insect feculae. The material, in a solvent transparent to microwave radiation, is exposed for a short time to steam in a microwave oven. EO extracts are analyzed directly by GC or GC-MS when plant material is fresh and terpenes contained in glandular structures on leaf surfaces are readily released into the solvent. For dried material or insect feculae, mechanical means are utilized first to break up tissue; however, the complete procedure is carried out inside the same vial to reduce losses. Statistical analysis shows that the reproducibility of the modified method is high. Several samples can be run within an hour with this method.  相似文献

Dibutyroyl derivatives of monoacylglycerols (DBMAG) from conifer seed oil triacylglycerols (TAG) were prepared by partial deacylation of TAG with ethylmagnesium bromide followed by diesterification with n-butyryl chloride. The resulting mixtures were analyzed by gas-liquid chromatography (GCL) with 65% phenylmethyl silicon open tubular fused-silica capillary column operated under optimal conditions and separated according to both their fatty acid structures and their regiospecific distribution. Seed oils of 18 species from 5 conifer families (Pinaceae, Taxaceae, Cupressaceae, Cephalotaxaceae, and Podocarpaceae) were analyzed. The chromatograms showed a satisfactory resolution of DBMAG containing palmitic (16∶0) stearic (18∶0), taxoleic (cis-5, cis-9 18∶2), oleic (cis-9 18∶1), cis-vaccenic (cis-11 18∶1), pinolenic (cis-5, cis-9, cis-12 18∶3), linoleic (cis-9, cis-12 18∶2), α-linolenic (cis-9 cis-12, cis-15 18∶3), and an almost baseline resolution of DBMAG containing gondoic (cis-11 20∶1), cis-5, cis-11 20∶2, sciadonic (cis-5, cis-11, cis-14 20∶3), dihomolinoleic (cis-11 cis-14 20∶2), juniperonic (cis-5, cis-11, cis-14, cis-17 20∶4), and dihomo-α-linolenic (cis-11, cis-14, cis-17 20∶3) acids. We have observed that results for Pinus pinaster and P. koraiensis seed oils obtained with this new simple method compared favorably with literature data established with other usual regiospecific analytical techniques. Δ5-Olefinic acids are esterified mainly at the external positions of the glycerol backbone in all cases, in agreement with data obtained by other methodologies allowing validation of the GLC regiospecific method. To date, 45 gymnosperm species (mostly Coniferophytes) from 21 genera belonging to 9 families have been analyzed, all of them showing a definite enrichment of Δ5-olefinic acids in the external positions of TAG. These fatty acids (FA), with one exception only, represent between-2 and 8% of FA esterified to the internal positions. For some species, i.e., P. koraiensis and P. pinaster, this asymmetrical distribution was established by at least three analytical procedures and confirmed by stereospecific analysis of their seed TAG.  相似文献
