The search for reliable marker molecules discriminating between human Th1 and Th2 cells identified a gene encoding a novel member of the G protein-coupled leukocyte chemoattractant receptor family, which is selectively expressed in Th2 but not Th1 lineage cells, thereby named CRTH2 (chemoattractant receptor-homologous molecule expressed on Th2 cells). Studies with anti-CRTH2 mAbs demonstrated that CRTH2 was expressed in a small population (0.4-6.5%) of CD4+ T cells in fresh PBMCs of healthy adults, but no remarkable expression was seen in B cells and NK cells. In some cases, CD8+ T cells ( approximately 3.5%) expressed CRTH2. Phenotypes of CD4+ T cells expressing CRTH2 were CD45RA-, CD45RO+, and CD25+, similar to those of Ag-activated effector/memory T cells. Freshly isolated CRTH2+ CD4+ T cells produced Th2- but little or no Th1-type cytokines upon stimulation with PMA and ionomycin. In addition, an allergen-induced proliferative response in fresh PBMCs was significantly and selectively reduced by subtracting CRTH2+ cells. Together, these results indicate that CRTH2 is selectively expressed in vivo in an activated state of Th2 cells including allergen-responsive Th2 cells, suggesting its pivotal roles in ongoing Th2-type immune reactions.  相似文献

PURPOSE: To test the hypothesis that modification of release pattern of nonsteroidal anti-inflammatory drugs (NSAIDs) formulations shifts gastrointestinal (GI) toxicity of the drugs from the upper GI region to the distal intestine. METHODS: We assessed tiaprofenic acid (TA)-induced upper and lower increased GI permeability (a surrogate marker of toxicity) after administration of 20 mg and 40 mg/kg regular release (powder) and modified release formulations [sustained release (SR) beads and diethyl-beta-cyclodextrin (DCD):TA inclusion complex (INC)]. Urinary excretion of oral doses of GI permeability probes sucrose and 51Cr-EDTA was determined as measures of gastroduodenal and distal intestine, respectively. Pharmacokinetics of TA enantiomers were also studied following administration of a single 20 mg/kg dose of racemic TA as oral SR beads and iv solution. For powder and INC, previously reported pharmacokinetic data were used. RESULTS: Regular powder significantly increased the permeability at the gastroduodenal level. Modified-release formulations, on the other hand, did not cause damage in the gastroduodenum but produced significant increase in the permeability of the lower intestine. Consequently, to assess the pharmacokinetic-pharmacodynamic relationship, a new model was developed in which contribution of toxicity resulted from direct exposure to the drug was considered. CONCLUSIONS: Since the observed site of GI damage corresponds to the site of release and absorption of NSAID from the formulation, the possibility of a shift in the site of damage must be considered for the modified release formulations. A parallel evaluation of upper and lower GI toxicity is essential for a complete assessment of NSAID-induced GI damage.  相似文献

This article reports the effect of cell structure on oil absorption of highly oil-absorptive polyurethane foam (ON-PUR), which is suitable for on-site foaming. We have developed ON-PUR as an oil absorbing polyurethane foam using a very reactive recipe. ON-PUR was synthesized by mixing polyol, water, additives, and polymeric diphenylmethane diisocyanate (P-MDI) using a high-pressure foaming machine. Density, airflow, oil absorption of this foam, and cell structure by microscopy were measured. From these results, it was found that the airflow of this foam increased by crushing, and the oil absorption of this foam increased sharply in a narrow airflow range (from 0.1 to 0.8 scfm). This increase is estimated to be due to the decrease of closed cell structures by crushing treatment. Furthermore, we constituted on-site foaming system in bench scale, which was expected to be applicable to on-site preparation of ON-PUR. © 1997 John Wiley & Sons, Inc. Grounding wires and enclosed ZnO elements have been incorporated generally in 6.6‐kV distribution systems by TEPCO for the reduction of lightning overvoltages. At present, the reliability to lightning surges is tolerably good. However, the facility of grounding wires is not inexpensive and its maintenance is hard due to corrosion and disconnection in some areas. A typical model simulating TEPCO field adopting enclosed ZnO elements has been developed and we have evaluated relative failure risks systematically according to conditions with and without grounding wires against lightning overvoltages. Two kinds of failures discussed in the paper are the flashover of insulation and the overduty of ZnO elements, and two kinds of induced and direct lightning overvoltages are studied in flashover. The greatest problem with no grounding wire is the increase of ZnO elements' duty, but it was demonstrated that a short partial grounding wire around ZnO elements or the selection of heavier ZnO elements provides a solution. The main objectives of this study are to clarify the relative failure risks systematically according to realistic field conditions, the risk of small stroke currents having long duration to ZnO elements' duty, and countermeasures against ZnO elements' overduty. © 1999 Scripta Technica, Electr Eng Jpn, 127(1): 1–10, 1999  相似文献

Because sea cucumbers lack a well-developed immune system and can ingest pathogenic bacteria together with food, some form of active antibacterial substances must be present in the body for defense. In this study, the cDNA of an i-type lysozyme from the sea cucumber Stichopus japonicus (designated SjLys) was cloned by RT-PCR and RACE PCR techniques. The full length cDNA of SjLys was 713 bp with an open reading frame of 438 bp coding for 145 amino acids. Two catalytic residues (Glu34 and Asp47), conserved in i-type lysozymes, and a highly conserved region near the active site, MDVGSLSCG(P Y)(Y F)QIK, were detected in SjLys. In addition, the domain structure analysis of SjLys showed that it is highly similar to the medicinal leech destabilase, which belongs to a new phylogenetic family of invertebrate lysozymes possessing both glycosidase and isopeptidase activities. To gain insight into the in vitro antimicrobial activities of SjLys, the mature peptide coding region was heterologously expressed in Escherichia coli. The recombinant SjLys protein displayed an inhibitive effect on the growth of the tested Gram-positive and Gram-negative bacteria. A remarkable finding is that the recombinant SjLys exhibited more potent activities against all tested bacterial strains after heat-treating at 100 °C for 50 min. These results indicated that the S. japonicus lysozyme is an enzyme with combined enzymatic (glycosidase) and nonenzymatic antibacterial action.  相似文献

Natural photosynthesis, which achieves efficient solar energy conversion through the combined actions of many types of molecules ingeniously arranged in a nanospace, highlights the importance of a technique for site-selective coupling of different materials to realize artificial high-efficiency devices. In view of increasingly serious energy and environmental problems, semiconductor-based artificial photosynthetic systems consisting of isolated photochemical system 1 (PS1), PS2 and the electron-transfer system have recently been developed. However, the direct coupling of the components is crucial for retarding back reactions to increase the reaction efficiency. Here, we report a simple technique for forming an anisotropic CdS-Au-TiO2 nanojunction, in which PS1(CdS), PS2(TiO2) and the electron-transfer system (Au) are spatially fixed. This three-component system exhibits a high photocatalytic activity, far exceeding those of the single- and two-component systems, as a result of vectorial electron transfer driven by the two-step excitation of TiO2 and CdS.  相似文献

An electronic conductance with small fluctuations, which is stipulated in single-molecule junctions, is necessary for the precise control of single-molecule devices. However, the suppression of conductance fluctuations in conventional molecular junctions is intrinsically difficult because the fluctuations are related to the contact fluctuations and molecular motion. In the present study involving experimental and theoretical investigations, it is found that covering a single π-conjugated wire with an α-cyclodextrin molecule is a promising technique for suppressing conductance fluctuations. The conductance histogram of the covered molecular junction measured with the scanning tunneling microscope break-junction technique shows that the conductance peak for the covered junction is sharper than that of the uncovered junction. The covering technique thus has two prominent effects: the suppression of intramolecular motion, and the elimination of intermolecular interactions. Theoretical calculations of electronic conductance clearly support these experimental observations.  相似文献
