At atmospheric pressure, inactivation of protease from B. subtilis in raw milk and pasteurized milk (with and without homogenization) was studied in a temperature range of 50–80 °C. Thermal inactivation followed a first order kinetic model in the temperature range tested. Temperature dependence of the first order inactivation rate constants could be accurately described by the Arrhenius equation, allowing Ea values to be calculated. Different milk systems did not show differences in enzyme thermo stability.The combined thermal (40, 50 and 60 °C)-high hydrostatic pressure (300–450 and 600 MPa) effect on protease activity was studied. Protease was very resistant to high pressures. Pressure stability was higher in raw milk than in pasteurized milk; homogenization appeared to have a protective effect on the enzyme. The separate effects of pressure and temperature on enzyme inactivation were related to changes in L?-values and milk appearance.A very pronounced antagonistic effect between high temperature and pressure was observed, i.e. at temperatures where thermal inactivation at atmospheric pressure occurs rapidly, application of pressure up to 600 MPa exerted a protective effect.Industrial relevanceHigh hydrostatic pressure (HHP) is an emerging technology that has been successfully applied as a minimal process for a variety of foods. Although the potential for the use of HHP treatment as an alternative method to heat treatment of milk was proposed almost a century ago, the suitability of this innovative technology to extend the shelf-life of milk hinges not only on its ability to inactivate pathogenic vegetative microorganisms but also on its effectiveness to inactivate indigenous and endogenous enzymes. This work examines the combined effects of temperature, pressure and homogenization on the protease (exogenous enzyme from B. subtilis) activity in milk. People spend a considerable amount of time indoors. As a result, exposure to indoor contaminants is of great concern, notably via settled dust ingestion in particular for infants and toddlers. This paper proposes a critical review on the organic contamination of settled house dust and human exposure over the past 10 years and focused on sources, contaminations and measurement methods (sampling, pretreatment, storage and analysis). As many compounds were identified, arises the question of which ones to consider. Sensitive and selective analytical methods for simultaneous determination of targeted substances should be developed and evaluated. Various methods were described for sampling and sample preparation. Harmonization and standardization are needed to enable comparison of results from similar studies. Finally, an integrated multipollutant and multicompartment (settled dust, suspended particles and air) approach appears essential in order to determine the extent of the threat to public health posed by indoor contaminants. 相似文献

This paper reports on the use of ultrasound (US) as a suitable strategy to control the growth of spoiling yeasts in fruit juices. In a first phase, US technique was tested towards Saccharomyces cerevisiae inoculated in different juices (strawberry, orange, apple, pineapple and red-fruits): the treatment was performed by modulating the level of the power (20–60 %), the duration of the treatment (2–6 min) and the pulse (2–6 s), according to a fractional design. Then, the best treatment was applied against some other spoiling yeasts (Pichia membranifaciens, Wickerhamomyces anomalus, Zygosaccharomyces bailii, Zygosaccharomyces rouxii, Candida norvegica). Finally, a challenge test for a commercial beverage (red-fruit juice), inoculated with Z. bailii and containing a citrus extract, was conducted evaluating the effect of US on the sensory scores of the beverage. The results showed that the effect of US was mainly influenced by the power and the duration of the treatment; on the other hand, the effect of pulse was less significant and slight. The highest reduction of S. cerevisiae was found in the following combination of the design: power 60 %/time 4 min/pulse 2 s and power 60 %/time 6 min/pulse 6 s; this result was confirmed for the other spoiling yeasts. US and citrus extract could be combined to prolong the shelf life of the red-fruit juice and control the growth of Z. bailii. The two hurdles exerted a different role and acted in different times: US reduced the initial contamination, whilst citrus extract controlled the yeast within the storage. 相似文献

The cerebral synthesis of cholesterol is mainly handled by astrocytes, which are also responsible for apoproteins’ synthesis and lipoproteins’ assembly required for the cholesterol transport in the brain parenchyma. In Alzheimer disease (AD), these processes are impaired, likely because of the astrogliosis, a process characterized by morphological and functional changes in astrocytes. Several ATP-binding cassette transporters expressed by brain cells are involved in the formation of nascent discoidal lipoproteins, but the effect of beta-amyloid (Aβ) assemblies on this process is not fully understood. In this study, we investigated how of Aβ1-42-induced astrogliosis affects the metabolism of cholesterol in vitro. We detected an impairment in the cholesterol efflux of reactive astrocytes attributable to reduced levels of ABCA1 transporters that could explain the decreased lipoproteins’ levels detected in AD patients. To approach this issue, we designed biomimetic HDLs and evaluated their performance as cholesterol acceptors. The results demonstrated the ability of apoA-I nanodiscs to cross the blood–brain barrier in vitro and to promote the cholesterol efflux from astrocytes, making them suitable as a potential supportive treatment for AD to compensate the depletion of cerebral HDLs. 相似文献
