We report application of the PEGylated lipids (PEG lipopolymers) containing disulphide as supports for sensing films for quartz crystal microbalance (QCM) odor sensors. The materials are binding covalently to the surface of gold QCM sensor electrode creating self-assembled cushion. Additional amounts of lipid or lipid-derived materials can be physisorbed on that chemisorbed coating. Both processes do not require much labor and can be performed with minimum instruments in a simple process. The sensors fabricated with the chemisorbed supports are more sensitive to the tested odorants than their non-supported counterparts. Enhanced sensitivity is derived from higher fluidity of the supported films in comparison to the non-supported ones. Discrimination capability among odorants is also better for the sensors with chemisorbed supports than for the non-supported ones—there are no overlaps between the sample groups and the samples are clustered closely within the groups. Overall, the studied supported sensors introduce interesting properties that can be utilized in the odor sensing systems using QCM sensors.  相似文献

Fructose 1,6-bisphosphatase 2 (Fbp2) is a gluconeogenic enzyme and multifunctional protein modulating mitochondrial function and synaptic plasticity via protein-protein interactions. The ability of Fbp2 to bind to its cellular partners depends on a quaternary arrangement of the protein. NAD+ and AMP stabilize an inactive T-state of Fbp2 and thus, affect these interactions. However, more subtle structural changes evoked by the binding of catalytic cations may also change the affinity of Fbp2 to its cellular partners. In this report, we demonstrate that Fbp2 interacts with Co2+, a cation which in excessive concentrations, causes pathologies of the central nervous system and which has been shown to provoke the octal-like events in hippocampal slices. We describe for the first time the kinetics of Fbp2 in the presence of Co2+, and we provide a line of evidence that Co2+ blocks the AMP-induced transition of Fbp2 to the canonical T-state triggering instead of a new, non-canonical T-state. In such a state, Fbp2 is still partially active and may interact with its binding partners e.g., Ca2+/calmodulin-dependent protein kinase 2α (Camk2α). The Fbp2-Camk2α complex seems to be restricted to mitochondria membrane and it facilitates the Camk2α autoactivation and thus, synaptic plasticity.  相似文献

Ovarian cancer (OC) is the leading cause of death among women with genital tract disorders. Melatonin exhibits oncostatic properties which it may effect through binding to its membrane receptor, MT1. The aim of this study was to determine the expression of MT1 in OC cells and to correlate this with clinical and pathological data. Immunohistochemistry was performed on 84 cases of OC. Normal ovarian epithelial (IOSE 364) and OC (SK-OV-3, OVCAR-3) cell lines were used to examine the MT1 expression at protein level using the western blot and immunofluorescence technique. The expression of MT1 was observed as cytoplasmic-membrane (MT1CM) and membrane (MT1M) reactions. A positive correlation between MT1CM and MT1M was found in all the studied cases. There were no significant differences between the expression of MT1CM, MT1M, and histological type, staging, grading, presence of residual disease, or overall survival time. Immunofluorescence showed both MT1M and MT1CM expression in all the tested cell lines. Western blot illustrated the highest protein level of MT1 in IOSE 364 and the lowest in the OVCAR-3. The results indicate the limited prognostic significance of MT1 in OC cells.  相似文献

Plant polyphenols, such as flavonoids, are ubiquitous components of human diet, and have been recognised as efficient radical scavengers and antioxidants. Their protective effects were especially seen in cell-free assays. Whilst in living cells, depending on experimental conditions, polyphenols may manifest both anti- and pro-oxidative activity. In the present study, the effectiveness of fifteen structurally different flavonoids against hypochlorite-mediated changes in in vitro systems, with and without cells, was tested and compared. Quercetin was found to be the best protector against hypochlorite-mediated fluorescein bleaching and BSA thiol groups oxidation, whilst in living cells its effects on hypochlorite-associated cytotoxicity were moderate. Contrary, epigallocatechin gallate, pelargonidin and catechin, with less impressive hypochlorite scavenging activity in cell-free assays, were the most effective against 3-chlorotyrosine formation after hypochlorite treatment. Taken together, special care should be taken when extrapolating the results on antioxidant propensity of food bioactive compounds in vitro to the cellular milieu.  相似文献

ABSTRACT

The potential of the ozone-enriched atmosphere for the improvement of the shelf life of apples of Gloster variety (Malus domestica) in cold storage was investigated. The storage experiment was conducted for 84 days. During that period, ozone at the concentration of 1 ppm was dosed every 12 hours for 1 minute. However, the exposure to ozone at such a concentration was unsuccessful in terms of inhibition of fungal disease development. Furthermore, the captan level in apples was reduced; therefore, they were more susceptible to fungal disease. On the other hand, the tests of the physical properties showed that utilization of ozone slowed down the ripening of apples; therefore, extending their shelf life provided that they were not infected.  相似文献

Neighbourly contacts organised on-line have become a sign of our times, studied in Canada and the US for decades. This paper is devoted to the so-called ‘e-neighbourhood’ in Poland, and the example chosen is a housing estate in Poznań. The issues addressed include the history of the creation of a portal, changes in the spatial impact of the portal on the estate, and subjects discussed on the forum in the context of the formation of a neighbourhood community. At present, estate discussion forums are a curious novelty rather than a widespread development. They certainly cannot be assessed critically yet. They are not a threat to interpersonal contacts. What is more, they can be seen as a potential link initiating such contacts in rather contact-resistant communities of new estates. Naturally, they can also be an interactive board for notices, complaints and proposals, especially when most, if not all, estate residents have acquired access to the network.  相似文献

The experimental study of a large scale magnesium hydride tank (10 kg) is presented. In order to enhance thermal exchanges and improve storage time, the MgH2 powder has been compacted with 10 wt.% of Expanded Natural Graphite. An efficient heat exchanger has been designed to transfer the heat from the endo/exothermic sorption reactions to an external heat source/sink through a high temperature heat transfer fluid. An improvement of the loading and discharging time along the first hydrogenation cycles is observed owing to material thermal conductivity modifications. The properties of the cycled material have been studied in order to understand this evolution of the tank behavior.  相似文献

The main aim of this study was to compare the cytological difference between ovular mucilage cells in two Asteraceae species—Pilosella officinarum and Taraxacum officinale—in order to determine whether pectic epitopes, arabinogalactan proteins, or extensins are present. The immunocytochemical technique was used. Both the Taracacum and Pilosella genera have been used recently as models for understanding the mechanisms of apomixis. Knowledge of the presence of signal molecules (pectic epitopes, arabinogalactan proteins, and extensins) can help better understand the developmental processes in these plants during seed growth. The results showed that in Pilosella officinarum, there was an accumulation of pectins in the mucilage, including both weakly and highly esterified pectins, which was in contrast to the mucilage of Taraxacum officinale, which had low amounts of these pectins. However, Taraxacum protoplasts of mucilage cells were rich in weakly methyl-esterified pectins. While the mucilage contained arabinogalactan proteins in both of the studied species, the types of arabinogalactan proteins were different. In both of the studied species, extensins were recorded in the transmitting tissues. Arabinogalactan proteins as well as weakly and highly esterified pectins and extensins occurred in close proximity to calcium oxalate crystals in both Taraxacum and Pilosella cells.  相似文献
