Baseline concentration levels of As, B, Ca, Cd, Cr, Cu, Co, Fe, Mg, Mn, Mo, Ni, P, Pb, Se, V, and Zn were determined for Porphyra columbina and Ulva sp. collected from three locations along San Jorge Gulf, in Patagonia Argentina. Elements were quantified by inductively coupled plasma-optical emission spectrometry, with the exception of lead and cadmium in some samples which were determined by electrothermal atomic absorption spectrometry. Three stations with different exposure degree to human activities, Bahía Solano, the mouth of Arroyo La Mata stream and Punta Maqueda, were selected as sampling points. The results showed a wide range of metal retention capacity between the two studied species. Regarding the levels of pollutants found in the researched sites, Punta Maqueda seemed to be less influenced by anthropogenic activities than the other two sites except for Cd. Taking into account their toxicities seasonal variations in Pb and Cd levels were studied in both algae in Punta Maqueda. Maximum concentrations of Cd (9.8 microg g(-1) dry wt.) were observed in P. columbina during winter, and maximum levels of Pb (0.82 microg g(-1) dry wt.) were detected in Ulva sp. during summer. Legislative and health safety aspects were evaluated for Cd and Pb.  相似文献

Wood ash, a by-product generated in power plants, can be used to fertilize forest plantations to replenish nutrients lost during harvesting. Although wood ash generally contains low levels of trace metals, release of some of these may occur soon after ash application in acid soils. The risk of heavy metal contamination associated with application of mixed wood ash was assessed in six Pinus radiata D. Don plantations, on two types of mineral soil differing in texture, drainage and CECe. Four of the stands received a single application of 4500 kg ha(-1) (March 2003), and in the other two stands the same treatment was applied over three consecutive years (2003-2005). Trace metal (Cd, Cr, Cu, Mn, Ni, Pb, Zn) concentrations were monitored throughout the 3 years in different components of the forest ecosystem--soil solid fraction, soil solution, tree needles, ground vegetation and different mushroom species. Repeated applications of wood ash led to moderate increases in soil extractable Mn and Zn, and Mn in all mushrooms species. However, the maximum concentrations did not reach levels potentially harmful to organisms. Concentrations of Zn, Cu and Cd decreased in some mushroom species, probably because of increased soil pH caused by the treatment. Heavy metal concentrations in tree needles and ground vegetation were not altered. Although the risk of heavy metal contamination appears to be low, the long-term effects of wood ash application must be assessed.  相似文献

Nisin is a bacteriocin used against food spoilage bacteria. Sulphur dioxide is a potent antioxidant as well as an antimicrobial agent widely used in the wine industry. In this study we describe the effect of these important antibacterial agents on the growth of a collection of 64 lactic acid bacteria (23 Oenococcus, 29 Lactobacillus, 3 Leuconostoc and 9 Pediococcus), 23 acetic acid bacteria and 20 yeast isolates, most of them recovered from wine. Minimal inhibitory concentrations (MIC) and minimal bactericide concentrations of nisin, potassium metabisulphite and ethanol were determined. Nisin MIC(50) values for the tested isolates were as follows: 0.024, 12.5, 200 and > or micro for oenococci, lactobacilli-pediococci-leuconostoc, acetic acid bacteria and yeasts, respectively. Synergistic effects on bacterial growth inhibition were observed, and potassium metabisulphite MIC(50) values decreased from one to three orders of dilution when it was combined with subinhibitory concentrations of nisin in the growth media. This effect was observed in all lactic acid bacteria species of our study. Significant differences in nisin sensitivity were observed between Gram-positive and Gram-negative bacteria, and between Oenococcus oeni and other species of lactic acid bacteria. It is concluded that appropriate combinations of nisin and metabisulphite could control the growth of spoilage bacteria in wine and therefore allow a decrease in the levels of sulphur dioxide currently used by the wine industry.  相似文献

Despite the important role of total soluble phenolic compounds (TSPC) in plant and human health, a large number of studies that have evaluated the TSPC in food matrices did not consider possible interfering compounds such as soluble sugars, ascorbic acid (AA), and other reducing compounds in the Folin-Ciocalteu (F-C) quantification method. Thus, the present study describes steps for the optimization of a methodology to extract the TSPC using banana leaves as an example. In addition, the method was tested using ascorbate oxidase (AOX) as a tool to determine and emphasize the importance of the evaluation of AA interference in food products using orange juice as an example. The results showed that two extraction cycles with 80 % acetone and a posterior hexane extraction cycle to remove the excess of chlorophylls were able to obtain a good TSPC extraction yield from banana leaves without extracting compounds that may interfere with the F-C method. The methodology has proven to be accurate, precise, simple, rapid, and inexpensive. Additionally, an overestimation of the TSPC levels in AA-rich matrices was proven using orange juice as an example. Finally, it was demonstrated that the use of AOX could be a useful and simple tool to verify and reduce the AA interference. This work proves and emphasizes the importance of evaluating the yield of the extraction and interferences in the quantification of the TSPC in vegetal-derived foods, which are complex matrices that should be cautiously evaluated.  相似文献

The effect of high-intensity pulsed electric field (HIPEF) treatments on polygalacturonase (PG) activity in an aqueous solution of a commercial enzyme preparation was evaluated. HIPEF treatments reduced PG activity notably. Exponentially decaying pulses of 40 and 160 µs generated by a laboratory scale device were applied in a batch processing and bipolar mode. Electric fields ranged from 5.18 to 19.39 kV/cm. The number of pulses ranged up to 400. Temperature was always below 25 °C. Maximum inactivation of PG activity (98%) required 32.4 ms HIPEF treatment at 10.28 kV/cm. PG activity depleted exponentially because of HIPEF treatments. The first order rate constants ranged from 32 to 590 µs -1 and increased exponentially with electric field intensity. PG activity decreased exponentially with input electric energy density.  相似文献

Results are presented of the effect of pre-treatments before freezing followed by long and short-term frozen storage (12 months at –18 °C and 24 days suffering temperature fluctuations between –18 °C and –12 °C) on quality parameters of raspberry. Pre-treatments were carried out with calcium, low methoxyl pectin, a combined solution, and results compared with untreated control fruits. Kramer shear, back extrusion, compression, and multiple penetration tests were used to measure rheological behavior. One-hundred mM CaCl2 reduced the long and short-term frozen storage induced loss of firmness. For long-term storage at –18 °C, a softening of the tissue became evident between 3 and 12 months and at each date test the stored fruits were firmer than those without storage. For short-term storage with fluctuations, the loss of firmness was evident between 0 and 24 days, and at all the testing dates the stored fruits were softer than those without storage. Results evidenced a higher cell damage in the short-term frozen storage. Coefficients of softening per day suffering fluctuation were determined, the highest values being given by Kramer shear energy and back extrusion maximum force (>1%). Short-term frozen storage affected physical and physico-chemical characteristics, increasing the saturation (r) and the anthocyanins and decreasing the ascorbic acid of the raspberries. In both storage conditions, pre-treatments reduced the drip loss, which correlated best with the Kramer shear energy. Panelists detected mainly time effect on the sensory firmness. For long-term, sensory firmness and juiciness gave the highest correlations with back extrusion maximum force, while for the short-term, sensory firmness and drip loss gave the highest correlations with the Kramer shear energy. SEM revealed different degrees of mechanical damage to structure, which accounted for rheological behavior of the fruits.  相似文献

Activated carbon (AC) has been used to catalyze the ozonation of pyruvic acid in water. Pyruvic acid conversions were found to be 9 and 37% after 90 min of single ozonation and single adsorption with 40 gL?1 AC, respectively, while 82% was reached at the same conditions during the AC catalytic ozonation. Also, for similar conditions, mineralization reached values of 67% in the AC catalytic ozonation against hardly 5% in the non-catalytic experiment. The process likely develops through both adsorption of ozone and pyruvic acid on the AC surface and generation of hydroxyl radicals that eventually is the responsible oxidizing species. Rate constants for both non-catalytic ozonation and AC-Ozone catalytic surface reaction, at 20°C and pH 7.5, were found to be 0.025 min?1 and 87.9 Lg?1s?1, respectively. For AC concentrations higher than 2.5 gL?1 gas-liquid mass transfer of ozone constituted the limiting step. At lower concentrations, internal diffusion plus surface reaction controlled the process rate.  相似文献

We report two experiments that investigated the regulation of memory accuracy with a new regulatory mechanism: the plurality option. This mechanism is closely related to the grain-size option but involves control over the number of alternatives contained in an answer rather than the quantitative boundaries of a single answer. Participants were presented with a slideshow depicting a robbery (Experiment 1) or a murder (Experiment 2), and their memory was tested with five-alternative multiple-choice questions. For each question, participants were asked to generate two answers: a single answer consisting of one alternative and a plural answer consisting of the single answer and two other alternatives. Each answer was rated for confidence (Experiment 1) or for the likelihood of being correct (Experiment 2), and one of the answers was selected for reporting. Results showed that participants used the plurality option to regulate accuracy, selecting single answers when their accuracy and confidence were high, but opting for plural answers when they were low. Although accuracy was higher for selected plural than for selected single answers, the opposite pattern was evident for confidence or likelihood ratings. This dissociation between confidence and accuracy for selected answers was the result of marked overconfidence in single answers coupled with underconfidence in plural answers. We hypothesize that these results can be attributed to overly dichotomous metacognitive beliefs about personal knowledge states that cause subjective confidence to be extreme. Since the fall of the Wall, there has been heavy international investment in building in East Berlin. On the whole, efforts have remained marginal. Only the western part of East Berlin with its baroque grid has been developed. The central and eastern parts remain as they were. The planning for City East follows four main criteria: to redefine the historic core by structural means without resort to architectural pastiche; to redesign the major roads which occupy much of the historic centre; to restructure the modernism of the GDR capital without demolishing the (mainly prefabricated) buildings; to form an idea of the third town which would be developed by merging existing features with our new plan.  相似文献
