A survey of the occurrence of Campylobacter, Salmonella, Listeria and Shiga toxin-producing E. coli was performed on 60 flocks of free-range chicken from 34 farms in the Basque Country (Northern Spain). Campylobacter was the most prevalent of the four pathogens, isolated in 70.6% of the farms, followed by L. monocytogenes (26.5%), and Salmonella (2.9%). No E. coli O157 or other STEC were isolated. In total 48 flocks from 26 farms were positive for at least one pathogen: 31 of them for a single pathogen (64.6%), and 17 for more than one species (35.4%). C. coli was more prevalent than C. jejuni (15 vs. 13 farms), and both species of Campylobacter were found in 3 farms. L. monocytogenes isolates were identified as serotype 4b complex, and the only Salmonella isolated was serovar Enteritidis. flaA PCR-RFLP performed on 91 Campylobacter isolates (36 C. jejuni and 55 C. coli) yielded 26 patterns, with higher diversity among the C. jejuni isolates. More than one pattern was found in 11 farms, and in 8 of them several patterns were found within the same flock. The findings of the present study suggest that the free-range rearing conditions described herein might have an advantageous effect on diminishing Salmonella but not on Campylobacter or L. monocytogenes flock contamination. 相似文献

An accurate and sensitive method for determination of 18 polycyclic aromatic hydrocarbons (PAHs) (16 PAHs considered by USEPA as priority pollutants, dibenzo[a,l]pyrene and benzo[j]fluoranthene) in fish samples was validated. Analysis was performed by microwave-assisted extraction and liquid chromatography with photodiode array and fluorescence detection. Response surface methodology was used to find the optimal extraction parameters. Validation of the overall methodology was performed by spiking assays at four levels and using SRM 2977. Quantification limits ranging from 0.15–27.16 ng/g wet weight were obtained. The established method was applied in edible tissues of three commonly consumed and commercially valuable fish species (sardine, chub mackerel and horse mackerel) originated from Atlantic Ocean. Variable levels of naphthalene (1.03–2.95 ng/g wet weight), fluorene (0.34–1.09 ng/g wet weight) and phenanthrene (0.34–3.54 ng/g wet weight) were detected in the analysed samples. None of the samples contained detectable amounts of benzo[a]pyrene, the marker used for evaluating the occurrence and carcinogenic effects of PAHs in food. 相似文献

Background: Pyroptosis is a catabolic process relevant to periodontal disorders for which interleukin-1β (IL-1β) inflammation is central to the pathophysiology of the disease. Despite platelet-rich fibrin (PRF) anti-inflammatory properties and its application to support periodontal regeneration, the capacity of PRF to modulate pyroptosis, specifically the production and release of IL-1β, remains unknown. The question arises whether PRF could regulate IL-1β release from macrophages in vitro. Methods: To answer this question, RAW 264.7 macrophages and primary macrophages obtained from murine bone marrow were primed with PRF before being challenged by lipopolysaccharide (LPS). Cells were then analysed for the pyroptosis signalling components by gene expression analyses and IL-1β secretion at the protein level. The release of mitochondrial reactive oxygen species (ROS) was also detected. Results: PRF lowered the LPS-induced expression of IL-1β and NLRP3 inflammasome, caspase-11 and IL-18 in primary macrophages, and IL-1β and caspase-11 in RAW 264.7 cells. Additionally, PRF diminished the secretion of IL-1β at the protein level in LPS-induced RAW 264.7 cells. This was shown through immunoassays performed with the supernatant and further confirmed by analysing the lysates of permeabilised cells. Furthermore, PRF reduced the ROS release provoked by LPS in RAW 264.7 cells. Finally, to enhance IL-1β release from the LPS-primed macrophages, we introduced a second signal with adenosine triphosphate (ATP). In this setting, PRF significantly reduced IL-1β release in RAW 264.7 cells and a trend to diminish IL-1β release in primary macrophages. Conclusion: These findings suggest that PRF can reduce IL-1β release and, at least in part, inhibit pyroptosis-related factors in LPS-challenged macrophages. 相似文献

The chemical modification of commodity polymers such as polyethylene (PE) is a versatile synthetic approach for preparing materials that cannot be manufactured cost-effectively using conventional polymerization techniques. Aiming to improve PE character low contents of dicumyl peroxide (DCP), from 0% to 1.5% was added as crosslinker to an industrial batch (PEs mixture and additives). From tensile testing crosslinking provided higher elastic modulus most due to the restrained microstructure where XPEs macromolecular chains are interconnected also providing lower strain at break. Crosslinking effects on the nonisothermal melt crystallization rate (Cmax) and degree of crystallinity (Xc) were evaluated; Cmax increased with the cooling rates, whereas Xc increased upon DCP addition. The melt crystallization kinetics were thoroughly investigated applying Pseudo-Avrami, Ozawa, and Mo models. Ozawa failed to describe the crystallization most due to ignore the secondary crystallization and spherulites impingement at the end of crystallization while Pseudo-Avrami and Mo provided quite good fits. The activation energy was computed using Arrhenius' approach, crosslinked compounds presented higher energy consumption, whereas exception was verified for 0.5XPE which displayed the lowest energy and overall the best mechanical performance this is the most proper compound for industrial applications, such as packaging, and disposables as well as general goods. 相似文献

The objective of this research was to investigate the effects of temperature (40–70 °C), frequency (37 and 80 kHz) and ultrasonic operation mode (normal, pulse and sweep), and their interactions on the recovery of phenolic compounds and antioxidants present in pomegranate peel using ultrasound-assisted extraction. The content of individual phenolic compounds, total phenolic compounds (TPC), antioxidant capacity (AC) and total extract yield (X0) were analysed. Significant effects were observed between some sources of variation in recovery of individual compounds at TPC and X0, and no significant effect was observed for AC. The process and conditions studied shown to be interesting to extract antioxidants, since a high antioxidant capacity was verified in the extracts. Temperatures around 50–60 °C, using 37 kHz frequency, and normal or pulse modes were the conditions that provided better yields of phenolic compounds and X0. Regarding AC, the best condition observed was at 70 °C, 80 kHz frequency and continuous mode. 相似文献

Pre-impregnated carbon fiber/epoxy resin (CF/epoxy prepreg) gained its popularity for significant stress applications, especially in the aerospace industry, owing to its excellent resistance and low specific mass. However, these CF/epoxy prepregs have a tendency to crack propagation. A solution for the prepregs fragility is the addition of carbon nanotubes (CNTs), especially those functionalized with amino groups, reinforcing the material due to its exceptional mechanical properties. In this work, the influence of the carbon chain length of two different amino-functionalized CNTs from diverse backgrounds (commercial and laboratory growth CNTs) is studied. The nanofillers were added in CF/epoxy prepregs by dry spraying without solvent aid. CNTs' samples were characterized by X-ray photoelectron spectroscopy, Raman spectroscopy, transmission electron microscopy, and thermogravimetric analysis (TGA), while the composites were analyzed by TGA, dynamic-mechanical analysis, and field emission scanning electron microscopy. The various surface treatment occurred at different levels according to the CNTs background, and all samples exhibited a distinct behavior. These differences were also observed in the composites' thermomechanical performance: CNTs functionalized with larger carbon chain amine presented the best results, with an increase of almost 100% in the storage moduli (E'), confirming the efficiency of amino-functionalized CNTs in the reinforcement of CF/epoxy prepregs. 相似文献

This study was aimed to evaluate the efficiency of the organic chlorine and acetic acid solutions on the inactivation of adhered cells of Escherichia coli, Cronobacter sakazakii and Klebsiella pneumoniae to lettuce. Besides, the growth and inactivation of K. pneumoniae adhered to lettuce was modeled. According to the findings, the use of chlorine solution (170 mg/ml of total residual chlorine) caused reductions of 1.8, 1.9, and 1.9 log for E. coli, C. sakazakii, and K pneumoniae, respectively, were recorded. In this regard, the organic chloramine was more effective in controlling the adhered microorganisms while compared with 1.5% acetic acid solution, while the addition of 0.5% sodium chloride to 1.5% acetic acid solution increased microbial inactivation. K. pneumoniae RC-34 inactivation was characterized by the presence of two sub-populations with different resistances against the proposed sanitizers. Moreover, the growth kinetic parameters of K. pneumoniae RC-34 adhered to lettuce leaves were very similar to that reported in the literature for nonadhered microorganisms. The predictive data generated can be valuable to assess the growth and inactivation of produce adhered microorganisms in leafy produce. 相似文献
