Photobleaching of the fluorophore fluorescein in an aqueous solution is dependent on the oxygen concentration. Therefore, the time‐dependent bleaching behavior can be used to measure of dissolved oxygen concentrations. The method can be combined with epi‐fluorescence microscopy. The molecular states of the fluorophore can be expressed by a three‐state energy model. This leads to a set of differential equations which describe the photobleaching behavior of fluorescein. The numerical solution of these equations shows that in a conventional wide‐field fluorescence microscope, the fluorescence of fluorescein will fade out faster at low than at high oxygen concentration. Further simulation showed that a simple ratio function of different time‐points during a fluorescence decay recorded during photobleaching could be used to describe oxygen concentrations in an aqueous solution. By careful choice of dye concentration and excitation light intensity the sensitivity in the oxygen concentration range of interest can be optimized. In the simulations, the estimation of oxygen concentration by the ratio function was very little affected by the pH value in the range of pH 6.5–8.5. Filming the fluorescence decay by a charge‐coupled‐device (ccd) camera mounted on a fluorescence microscope allowed a pixelwise estimation of the ratio function in a microscopic image. Use of a microsensor and oxygen‐consuming bacteria in a sample chamber enabled the calibration of the system for quantification of absolute oxygen concentrations. The method was demonstrated on nitrifying biofilms growing on snail and mussel shells, showing clear effects of metabolic activity on oxygen concentrations. Microsc. Res. Advanced biomaterials and scaffolds for tissue engineering place high demands on materials and exceed the passive biocompatibility requirements previously considered acceptable for biomedical implants. Together with degradability, the activation of specific cell–material interactions and a three-dimensional environment that mimics the extracellular matrix are core challenges and prerequisites for the organization of living cells to functional tissue. Moreover, although bioactive signalling combined with minimization of non-specific protein adsorption is an advanced modification technique for flat surfaces, it is usually not accomplished for three-dimensional fibrous scaffolds used in tissue engineering. Here, we present a one-step preparation of fully synthetic, bioactive and degradable extracellular matrix-mimetic scaffolds by electrospinning, using poly(D,L-lactide-co-glycolide) as the matrix polymer. Addition of a functional, amphiphilic macromolecule based on star-shaped poly(ethylene oxide) transforms current biomedically used degradable polyesters into hydrophilic fibres, which causes the suppression of non-specific protein adsorption on the fibres’ surface. The subsequent covalent attachment of cell-adhesion-mediating peptides to the hydrophilic fibres promotes specific bioactivation and enables adhesion of cells through exclusive recognition of the immobilized binding motifs. This approach permits synthetic materials to directly control cell behaviour, for example, resembling the binding of cells to fibronectin immobilized on collagen fibres in the extracellular matrix of connective tissue.  相似文献

A range of physico‐chemical properties of 21 Californian red wines from 1 to 28 years old were correlated with wine age. The level of monomeric phenolics and low molecular‐weight phenolic oligomers decreased with age, while the level of high molecular‐weight phenolics increased. On the other hand, total phenols given by the Folin–Ciocalteau method, antioxidant capacity determined using a controlled chain reaction of methyl linoleate and the intensity of the anodic peak determined by cyclic voltammetry (CV) did not show any significant correlation with wine age. Total phenols and the shapes of CV traces were more dependent on the grape variety and the particular viticultural and wine‐making practices rather than on wine age. At the same time, young and aged wines showed a pronounced difference in the shape of [O2] traces recorded during the oxidation of methyl linoleate in a micellar solution inhibited by wine addition. In vitro inhibition of microsomal lipid peroxidation (MLP) by the major South African red (Cabernet Sauvignon, Ruby Cabernet, Pinotage, Shiraz, Merlot) and white (Sauvignon blanc, Chenin blanc, Chardonnay, Colombard) commercial cultivar wines is presented for the first time. Of the red wines, Merlot was the most effective MLP inhibitor, with Ruby Cabernet and Pinotage being the least effective. Of the white wines, Chenin blanc and Chardonnay were the least and most effective MLP inhibitor, respectively. The mean antioxidant potencies (AP) of the red and white wine total phenols were 14.25 and 4.19, respectively. Ascorbic acid, present in some white wines, counteracted their ability to inhibit MLP. Inhibition of MLP significantly (P<0.001) correlated with the total phenol content of red (r=0.90) and white (r=0.73) wines, as well as the flavanol content (r=0.88) of red wines and the flavanol (r=0.79) and tartaric acid ester (r=0.73) contents of white wines. The MLP inhibitory activities of selected flavonoids were in the order: Quercetin > procyanidin B3 > malvidin > cyanidin ≈ (−)-epicatechin > (+)-catechin ≈ delphinidin.  相似文献

A novel substructuring method is developed for the coupling of boundary element and finite element subdomains in order to model three‐dimensional multi‐region elastodynamic problems in the time domain. The proposed procedure is based on the interface stiffness matrix approach for static multi‐region problems using variational principles together with the concept of Duhamel integrals. Unit impulses are applied at the boundary of each region in order to evaluate the impulse response matrices of the Duhamel (convolution) integrals. Although the method is not restricted to a special discretization technique, the regions are discretized using the boundary element method combined with the convolution quadrature method. This results in a time‐domain methodology with the advantages of performing computations in the Laplace domain, which produces very accurate and stable results as verified on test examples. In addition, the assembly of the boundary element regions and the coupling to finite elements are greatly simplified and more efficient. The aim of this work was to obtain further information about the origin of fluoride profiles in cementum. Fluoride was administered to rats at varying doses (0.50, 100 ppm F in drinking water) and for different durations (4, 13 and 25 weeks). Fluoride distribution across the full thickness of molar cementum in rats was measured by means of an abrasive micro-sampling technique. The average fluoride concentrations in cementum increased significantly with increasing dose and duration of fluoride administration. The relative reduction of the average fluoride concentrations after cessation of fluoride administration was 94.2-36.5% at 50 ppm F and 62.2-49.2% at 100 ppm F in the outer layers (1-60 microns) and 91.5-24.1% at 50 ppm F and 74.1-7.6% at 100 ppm F in the middle (61-120 microns) layers of the cementum, respectively. The reduction rates were more closely related to the time intervals following cessation rather than fluoride concentrations in drinking water or specificity within the cementum. Two factors which may influence this are new cementum formation after withdrawal of fluoride and some fluoride release from cementum surfaces when the fluoride supply stopped. It was concluded that the cessation of fluoride administration reduced the fluoride concentration on the outer layers of cementum differing from bone where reduction occurs across the entire thickness.  相似文献

II–VI polycrystalline semiconducting materials have come under increased scrutiny because of their wide use in the cost reduction of devices for photovoltaic applications. Cd1−xZnxSe is an important semiconducting alloy because of the tunability of its physical parameters such as band gap and lattice parameters by controlling its stoichiometry. Many more material characteristics of it would be altered and excellently controlled by controlling system composition x.Polycrystalline thin films of Cd1−xZnxSe with variable composition (0x1) have been deposited onto ultra-clean glass substrates by sintering process. The optical, structural and electrical transport properties of Cd1−xZnxSe thin films have been examined. The optical band gap and optical constants of these films were determined by using double beam spectrophotometer. The DC conductivity and activation energy of the films were measured in vacuum by two-probe technique. The Schottky junction of Cd1−xZnxSe with indium was made and the barrier height and ideality factor were determined using current–voltage characteristics. The nature of sample, crystal structure and lattice parameters were determined from X-ray diffraction patterns. The films were polycrystalline in nature having cubic zinc-blende structure over the whole range studied.Sintering is very simple and viable compared to other cost intensive methods. The results of the present investigation will be useful in characterizing the material, Cd1−xZnxSe, for its applications in photovoltaics.  相似文献
