In this study, we examined the influence of high levels of cytosolic calcium on phosphorylation status and function of GLUT-4 in isolated rat adipocytes. Intracellular calcium was elevated by exposing adipocytes to either extracellular ATP (1.6 mM) or thapsigargin (100 nM). Both agents increased cytosolic calcium 2-3 fold. While basal glucose uptake was unaffected, both ATP and thapsigargin reduced insulin-stimulated glucose transport by 40-70% (p < 0.05). Neither ATP nor thapsigargin affected GLUT-4 content or its translocation from the low density microsomes to the plasma membrane (PM). In contrast, GLUT-4 immunoprecipitated from the PM of adipocytes exposed to either ATP or thapsigargin was phosphorylated to a greater extent than the GLUT-4 isolated from control cells. ATP and thapsigargin also abolished insulin-stimulated dephosphorylation of GLUT-4. At the same time, GLUT-4 intrinsic activity was significantly reduced in adipocytes with high levels of cytosolic calcium (p < 0.05). Preincubation of adipocytes with cAMP antagonist, RpcAMP (10(-4) M), and calcium channel blocker, nitrendipine (30 microM), improved the ability of insulin to dephosphorylate GLUT-4 and restored insulin-stimulated GLUT-4 intrinsic activity. We conclude that elevated levels of cytosolic calcium interfere with insulin's ability to dephosphorylate GLUT-4, thus reducing its intrinsic activity.  相似文献

Sequences corresponding to 855 bp of 5' promoter region and the transit peptide from lambdaGK.1,a genomic clone encoding a 22 kDa alpha-kafirin seed protein from sorghum, were translationally fused to a cloned beta-glucuronidase (GUS) coding sequence from uidA and transferred to tobacco via Agrobacterium tumefaciens-mediated transformation. No GUS expression was detectable at any stage of growth in stems or leaves of these plants. However, GUS expression was detected in both embryo and endosperm tissues of resulting tobacco seeds 10-15 days after flowering. Dissected tissues indicate endosperm expression was localized within the bulk endosperm and not within the parenchyma cell layer underlying the integument. These studies also demonstrate that within dissected tobacco embryos, expression from the kafirin promoter was restricted to the mesocotyl region.  相似文献

Poly(N-isopropylacrylamide-acrylamide-methacrylic acid) [p(NIPa-AAm-Ma)] polymer microgels were prepared by free radical precipitation polymerization method. AgNPs were fabricated in the sieves of polymer network by chemical reduction using AgNO3 salt as a precursor of silver ions. Various techniques like dynamic light scattering (DLS), transmission electron microscopy (TEM), Fourier transform infrared microscopy (FTIR), and UV-Visible spectroscopy were used for characterization of pure and composite microgels. The diameter of AgNPs fabricated in polymeric network was found to be in the range of 10-15 nm. Stimuli responsive behavior of hybrid microgels was same as that of pure microgels. Catalytic efficiency of the hybrid microgels was investigated by reducing 4-Nitroaniline (4-NA) into 4-Aminoaniline (4-AA) using NaBH4 as reducing agent under different conditions of temperature of the medium, concentration of reducing agent, 4-Nitroaniline and hybrid microgels to explore the catalysis process. Kinetic and thermodynamic aspects of reduction of 4-Nitroaniline in the presence of catalyst were also discussed on the basis of values of Arrhenius and Eyring parameters like pre-exponential factor, activation energy, enthalpy of activation and entropy of activation. Catalytic activity of the hybrid microgels was found to be thermally tunable in the temperature range of 25-70 oC. The value of rate constant (k app ) for reduction of 4-NA was minimum at 55 °C, which can be attributed to volume phase transition of the hybrid microgels.  相似文献

The last decade has seen a flurry of research on all-pairs-similarity-search (or similarity joins) for text, DNA and a handful of other datatypes, and these systems have been applied to many diverse data mining problems. However, there has been surprisingly little progress made on similarity joins for time series subsequences. The lack of progress probably stems from the daunting nature of the problem. For even modest sized datasets the obvious nested-loop algorithm can take months, and the typical speed-up techniques in this domain (i.e., indexing, lower-bounding, triangular-inequality pruning and early abandoning) at best produce only one or two orders of magnitude speedup. In this work we introduce a novel scalable algorithm for time series subsequence all-pairs-similarity-search. For exceptionally large datasets, the algorithm can be trivially cast as an anytime algorithm and produce high-quality approximate solutions in reasonable time and/or be accelerated by a trivial porting to a GPU framework. The exact similarity join algorithm computes the answer to the time series motif and time series discord problem as a side-effect, and our algorithm incidentally provides the fastest known algorithm for both these extensively-studied problems. We demonstrate the utility of our ideas for many time series data mining problems, including motif discovery, novelty discovery, shapelet discovery, semantic segmentation, density estimation, and contrast set mining. Moreover, we demonstrate the utility of our ideas on domains as diverse as seismology, music processing, bioinformatics, human activity monitoring, electrical power-demand monitoring and medicine.  相似文献

The widely used ecosystem model DailyDayCent (DDC) was used to predict soil organic carbon (SOC) sequestration and yield under different fertilization treatments on a wheat (Triticum aestivum) monoculture on Broadbalk field, Rothamsted, UK. Over a period of about 170 years, the management practices on the test site, which is separated into plots with different treatments of fertilizer and manure application, are well documented. Four treatments consisting of control (no fertilizer), mineral nitrogen (N) fertilizer (MN), farm yard manure (FYM) and a combination of both (FYMN) have been selected for the study. DDC simulated the greatest increases in SOC (three times higher compared to simulation of the control plot) in the FYM plot. Overall a good agreement between modelled and measured yield was obtained (except in control plots). As observed in the experimental plots, the highest yield predicted by the model is with FYMN plot (more than three times higher relative to the control plot). A sensitivity test for the MN and FYM plots demonstrated that predicted SOC was relatively insensitive (1%) to bulk density, pH and field capacity. Higher sensitivity (4–7%) of predicted SOC changes were observed by changes in external C input. An increase of temperature by 1 °C or precipitation by 10% decreased predicted SOC by 2–4% and vice versa, and simulated a 1–4% variability in yield. DDC is a useful tool for simulating SOC in the long term plots, with a good fit to data, and is useful for examining alternate hypotheses of yield and SOC responses.  相似文献

This article discusses the results for the development of a nanogold‐particle/polyaniline‐modified glassy carbon electrode for the detection of arsenic(III) in water. A thin polyaniline film was electropolymerized onto a glassy carbon electrode. The gold nanoparticle was then deposited onto the polyaniline‐coated glassy carbon electrode via potential step electrolysis from 1.1 to 0 V versus Ag/AgCl/NaCl (saturated) for 45 s from a 0.5M H2SO4 solution containing 0.1 mM NaAuCl4 in the absence and presence of a 0.1 mM KI additive. The surface of the modified electrode was examined with scanning electron microscopy. Cyclic and anodic stripping voltammetry of arsenic(III) was performed on the modified electrode. The thus modified nanogold‐particle/polyaniline‐modified glassy carbon electrode prepared in the presence of the I? (KI) additive showed a high sensitivity in detecting arsenic(III) in water, and with stripping voltammetry, a limit of detection of 0.4 ppb arsenic was obtained, which is much lower than the arsenic guideline limit of the World Health Organization. © 2007 Wiley Periodicals, Inc. The presence of heavy metals in aqueous streams arising from the discharge of industrial effluents into water bodies is one of the most important environmental issues because of their toxic nature and its removal is highly essential. This paper deals with the adsorption studies for the removal of hexavalent chromium ions from aqueous solutions using Schiff based chitosan activated carbonized rice husk composites as adsorbent. The activation and surface properties of the adsorbent were characterized by Scanning Electron Microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), and Brauner Emmet and Teller (BET) analyzer. Central Composite Design (CCD) was used to optimize the process variables such as initial metal ion concentration, adsorbent dosage and pH of the solution on the performance of percentage removal and adsorption capacity. The experimental data were validated with different isotherms and kinetic models to evaluate the solute interaction behavior and nature of adsorption.  相似文献

Butyltin(IV) carboxylate compounds were obtained by reactions of butyltrichlorotin(IV) with potassium pivalate, perfluoroheptanoate, methacrylate, 2,6-pyridinedicarboxylate, and phthalate. The synthesized complexes were fully characterized by nuclear magnetic resonance (1H-, 13C-NMR), Fourier transform infrared (FTIR), mass spectroscopies (MS) and elemental analysis. These tin complexes were used as catalysts for the ring opening polymerization of ε-caplolactone and the conversion of monomers to polymers was completed in just 1 h. The structures of polymers were characterized by a combination of spectroscopic techniques (NMR, FTIR, MS), differential scanning calorimeter (DSC) and gel permeation chromatography. In this study, the ε-caplolactone polymers with different average molecular weights between 5000 and 40,000 Da having a regular structure were obtained.  相似文献
