Amine-functionalized sol-gels were investigated for the enrichment and purification of phosphopeptides from digested protein mixture solutions. Tetramethylorthosilicate (TMOS) and N'[3-(trimethoxysilyl)-propyl]-diethylenetriamine (TPDA) were used in a 1:1 mole ratio in the production of amine-functionalized sol-gels. The sol-gel network was then used for phosphopeptide enrichment. Phosphopeptide enrichment onto the synthesized amine-functionalized sol-gels was performed using an enolase digested peptide mixture, a β-casein digested peptide mixture, as well as these digested peptide mixtures contaminated 50-fold with bovine serum albumin (BSA). Moreover, phosphopeptide enrichment was successfully performed using nonfat milk as a highly contaminated and complex material. In each phosphopeptide enrichment and purification process, only phosphopeptides were enriched and separated from the other digested peptides. Phosphopeptides were adsorbed onto the amine-functionalized sol-gels at pH 4.0 and eluted at pH 1.0 using trifluoroacetic acid (TFA). For phosphopeptide analysis by MALDI-MS, a 2,5-dihydroxybenzoic acid matrix containing 1.0% phosphoric acid was used to overcome the degradation of phosphopeptides and provide high intensity phosphopeptide protonated molecular ion signal intensities. It was also found that phosphopeptide detection limits were improved to approximately 10 femtomoles. For rapid and specific phosphopeptide enrichment and purification, sol-gel materials were placed in a 10 μL pipet tip with glass wool on either side. Phosphopeptide enrichment from digested peptide mixtures was performed in a very short time (less than 1 min) at subpicomole levels using this novel lab-in-a-pipet-tip approach.  相似文献

The dielectric properties and ac electrical conductivity of Al/polyindole (Al/PIN) Schottky barrier diodes (SBDs) were investigated by using admittance spectroscopy (capacitance–voltage [CV] and conductance–voltage [G/ω‐V]) method. These CV and G/ω‐V characterizations were performed in the frequency range of 1 kHz to 10 MHz by applying a small ac signal of 40 mV amplitude from the external pulse generator, whereas the dc bias voltage was swept from (−10 V) to (+10 V) at room temperature. The values of dielectric constant (ε′), dielectric loss (ε″), dielectric loss tangent (tanδ), real and imaginary part of electrical modulus (M′ and M″), ac electrical conductivity (σac), and series resistance (Rs) of the Al/PIN SBDs were found to be quite sensitive to frequency and applied bias voltage at relatively low frequencies. Although the values of the ε′, ε″, tanδ, and Rs of the device were observed to decrease with increasing frequencies, the electric modulus and σac increased with increasing frequency for the high forward bias voltages. These results revealed that the interfacial polarization can more easily occur at low frequencies and that the majority of interface states (Nss) between Al and PIN, consequently, contribute to deviation of dielectric properties of the Al/PIN SBDs. Furthermore, the voltage‐dependent profile of both Rs and Nss were obtained from the CV and G/ω‐V characteristics of the Al/PIN SBDs at room temperature. © 2010 Wiley Periodicals, Inc. Bond strength, physical, and mechanical properties of lightweight PC were investigated with inclusion of pumice lightweight aggregate in maximum size of 12 mm. As binder material, epoxy resin‐based polymer was used with its hardener. The binder to aggregate ratio was 30% by weight. In addition, steel fibers were added to lightweight PC mixtures in ratio of 0, 0.5, and 1%. After lightweight PC mixture was prepared, it was poured in the molds with different type of steel‐bars in size of 100 × 100 × 100 mm3. The steel‐bars centered in the cubic molds, and they were in size of Ø12, Ø14, and Ø16. The specimens were cured at 60°C for 2 h. On the hardened polymer lightweight concrete (PLC), pull‐out test for bond strength and compressive strength tests were performed. Moreover, ultrasonic pulse velocity, water absorption by weight, specific porosity, and density experiments were carried out. The relation between physical and mechanical properties showed that PLCs become more durable when using ratio of steel fibers. POLYM. In practice, ferrous based materials are not used because they show similar properties to shaft material and are known to cause adhesive wear in journal bearings. With boronizing process, adhesive wear can significantly be decreased. In this study, boronizing has been applied to some ferrous based (SAE 1020, TS-DDK 40) journal bearings. Wear experiments were done on SAE 1050 steel shaft as counter abrader with 100 N loads, and 1500 rpm for 5 h by using a radial journal bearing test rig. Boronizing treatment increased wear resistance of SAE 1020, TS-DDK 40 about 10–25, and 5–7 times respectively.  相似文献

New π‐conjugated polymers containing dithieno(3,2‐b:2′,3′‐d)pyrrole (DTP) were successfully synthesized via electropolymerization. The effect of structural differences on the electrochemical and optoelectronic properties of the 4‐[4H‐dithieno(3,2‐b:2′,3′‐d)pyrrol‐4‐yl]aniline (DTP–aryl–NH2), 10‐[4H‐dithiyeno(3,2‐b:2′,3′‐d)pirol‐4‐il]dekan‐1‐amine (DTP–alkyl–NH2), and 1,10‐bis[4H‐dithieno(3,2‐b:2′,3′‐d)pyrrol‐4‐yl] decane (DTP–alkyl–DTP) were investigated. The corresponding polymers were characterized by cyclic voltammetry, NMR (1H‐NMR and 13C‐NMR), and ultraviolet–visible spectroscopy. Changes in the electronic nature of the functional groups led to variations in the electrochemical properties of the π‐conjugated systems. The electroactive polymer films revealed redox couples and exhibited electrochromic behavior. The replacement of the DTP–alkyl–DTP unit with DTP–aryl–NH2 and DTP–alkyl–NH2 resulted in a lower oxidation potential. Both the poly(10‐(4H‐Dithiyeno[3,2‐b:2′,3′‐d]pirol‐4‐il)dekan‐1‐amin) (poly(DTP–alkyl–NH2)) and poly(1,10‐bis(4H‐dithieno[3,2‐b:2′,3′‐d]pyrrol‐4‐yl) decane) (poly(DTP–alkyl–DTP)) films showed multicolor electrochromism and also fast switching times (<1 s) in the visible and near infrared regions. © 2014 Wiley Periodicals, Inc. In this paper, bees algorithm (BA) has been used for determine the optimal number of material handling equipment (MHE) used on the production centers. The unmet demands become zero at the end of the planning horizon, i.e., the part demands are totally satisfied through the horizon. The newly developed model provides network information, such as unmet demands and number of loaded and empty of MHE at any given time and centers. Consequently, the model provides a tool for helping managers with planning and decision-making in manufacturing systems. Computational tests showed that small-sized instances can be solved by the exact approach in a fair amount of central processing unit time, but it is not feasible for medium and large-sized instances. To tackle this problem, a bees algorithm is proposed to solve the model. The algorithm is a search procedure inspired by the way honeybees forage for food. The results obtained show the robust performance of the bees algorithm.  相似文献

In the present study, laser cutting of 7050 aluminum alloy sheets reinforced with Al2O3 and B4C particles are carried out. The cut geometry is examined using scanning electron microscope and the optical microscope. The lump parameter analysis technique is used to formulate and determine the kerf width size. The predictions for kerf width are compared with experimental data. The percentage kerf width size variation along the cut edges is determined and the influence of the laser power level and duty cycle of the laser pulses on the percentage kerf width size variation is examined. It is found that 7050 aluminum alloy reinforced with 20% Al2O3 composite results in relatively large kerf width size as compared to its counter parts that corresponding to 7050 aluminum alloy reinforced with 20% B4C composite. The kerf width size predicted agreed with the experimental data for both composites.  相似文献

The laser cutting of metallic substrates results in the development of thermal stresses around the cut edges. Depending on the cutting speed, laser power intensity, and material properties, stress levels reaching and exceeding the yielding limit of the substrate material can result. In the present study, the laser cutting situation is simulated and temperature as well as thermal stress fields are computed for steel, Inconel 625, and Ti-6Al-4V alloy. The cutting speed of the laser is considered to be constant and a constant temperature heat source with a focused spot diameter is assumed along the kerf surface at the cut edge, resembling the laser heat source. The equations for energy and thermal stresses are solved numerically using the finite element method (FEM). It is found that the temperature decays sharply in the vicinity of the cut edges and that the equivalent stress attains high values in this region. Inconel 625 results in the highest thermal stress levels in the vicinity of the cut edges and is then followed by steel and titanium alloy.  相似文献

In this study, the main objective was to asses the distribution of Cu, Cr, and As in soils adjacent to CCA-treated utility poles in Eastern Blacksea Region of Turkey (Trabzon, Rize and Artvin ) and determine the influence of soil composition.

Surface (0–5 cm) and subsurface soil samples (30–40 cm) were collected near CCA-treated utility poles and control soil samples away from CCA-treated utility poles were also collected. Water holding capacity, pH and mechanical properties of soil samples were determined for both depth levels.

Results showed that Cu, Cr and As concentration in soil samples taken from all three cities in 0–5 cm depth was higher than soil samples taken from 30–40 cm depth. Cu, Cr and As concentrations were much higher in soil samples taken from city of Rize  相似文献
