Genomic instability is prevented by the DNA damage response (DDR). Micronutrients, like zinc (Zn), are cofactors of DDR proteins, and micronutrient deficiencies have been related to increased cancer risk. Acute myeloid leukemia (AML) patients commonly present Zn deficiency. Moreover, reports point to DDR defects in AML. We studied the effects of Zn in DDR modulation in AML. Cell lines of AML (HEL) and normal human lymphocytes (IMC) were cultured in standard culture, Zn depletion, and supplementation (40 μM ZnSO4) conditions and exposed to hydrogen peroxide (H2O2) or ultraviolet (UV) radiation. Chromosomal damage, cell death, and nuclear division indexes (NDI) were assessed through cytokinesis-block micronucleus assay. The phosphorylated histone H2AX (yH2AX) expression was monitored at 0 h, 1 h, and 24 h after exposure. Expression of DDR genes was evaluated by quantitative real time polymerase chain reaction (qPCR). Zn supplementation increased the genotoxicity of H2O2 and UV radiation in AML cells, induced cytotoxic and antiproliferative effects, and led to persistent yH2AX activation. In contrast, in normal lymphocytes, supplementation decreased damage rates, while Zn depletion favored damage accumulation and impaired repair kinetics. Gene expression was not affected by Zn depletion or supplementation. Zn presented a dual role in the modulation of genome damage, preventing damage accumulation in normal cells and increasing genotoxicity and cytotoxicity in AML cells. 相似文献

This study was designed to assess the stability of Wechsler Adult Intelligence Scale—Revised (WAIS—R) factor scores across time. Information provided in the manual (D. Wechsler, 1981) on 2 subsets of the standardization data was used to estimate sample stability in Verbal Comprehension, Perceptual Organization, and Memory/Freedom from Distractibility scores. Injection molded cones have been successfully produced using a high content of recycled wire and cable fluff. The cone meets all general specifications. Dichlorocarbene modification of natural rubber (NR) carried out by alkaline hydrolysis of chloroform in presence of cetyl trimethyl ammonium bromide as phase‐transfer catalyst was investigated. Extent of chemical reaction was characterized by estimation of chlorine content and FTIR studies. Rate of dichlorocarbene addition depends on the time and temperature of reaction. Reaction carried out at 60°C for 2 h yielded a material with a chlorine content of 15%. Chemical modification of NR was accompanied by introduction of chlorine through cyclopropyl ring to the main chain of NR as revealed from FTIR studies. As level of chlorination increased, the physical nature of NR changed from a soft flexible state to a hard nontacky form. Blends of NR with hydrogenated nitrile rubber (HNBR) containing three to seven parts of dichlorocarbene‐modified NR (DCNR) of chlorine content 15% could be prepared by conventional mill mixing. Incorporation of DCNR into blends of NR and HNBR promoted polar interaction between the chlorine segments and acrylonitrile segments of the blend as shown from the shift in characteristic IR absorption peaks and shift in Tg from DSC studies. As a consequence, DCNR acted as an interface modifier in blends of NR and HNBR. Blends of NR and HNBR containing DCNR showed a considerable improvement in cure behavior, physical properties, and ageing characteristics in oil, ozone, and high temperature compared to pure blends of NR and HNBR. © 2006 Wiley Periodicals, Inc. Buildings with symmetrical façades are ubiquitous in urban landscapes and detailed models of these buildings enhance the visual realism of digital urban scenes. However, a vast majority of the existing urban building models in web‐based 3D maps such as Google earth are either less detailed or heavily rely on texturing to render the details. We present a new framework for enhancing the details of such coarse models, using the geometry and symmetry inferred from the light detection and ranging (LiDAR) scans and 2D templates. The user‐defined 2D templates, referred to as coded planar meshes (CPMs), encodes the geometry of the smallest repeating 3D structures of the façades via face codes. Our encoding scheme, take into account the directions, type as well as the offset distance of the sculpting to be applied at the respective locations on the coarse model. In our approach, LiDAR scan is registered with the coarse models taken from Google earth 3D or Bing maps 3D and decomposed into dominant planar segments (each representing the frontal or lateral walls of the building). The façade segments are then split into horizontal and vertical tiles using a weighted point count function defined over the window or door boundaries. This is followed by an automatic identification of CPM locations with the help of a template fitting algorithm that respects the alignment regularity as well as the inter‐element spacing on the façade layout. Finally, 3D boolean sculpting operations are applied over the boxes induced by CPMs and the coarse model, and a detailed 3D model is generated. The proposed framework is capable of modelling details even with occluded scans and enhances not only the frontal façades (facing to the streets) but also the lateral façades of the buildings. We demonstrate the potentials of the proposed framework by providing several examples of enhanced Google earth models and highlight the advantages of our method when designing photo‐realistic urban façades. 相似文献

We present an incremental Voronoi vertex labelling algorithm for approximating contours, medial axes and dominant points (high curvature points) from 2D point sets. Though there exist many number of algorithms for reconstructing curves, medial axes or dominant points, a unified framework capable of approximating all the three in one place from points is missing in the literature. Our algorithm estimates the normals at each sample point through poles (farthest Voronoi vertices of a sample point) and uses the estimated normals and the corresponding tangents to determine the spatial locations (inner or outer) of the Voronoi vertices with respect to the original curve. The vertex classification helps to construct a piece‐wise linear approximation to the object boundary. We provide a theoretical analysis of the algorithm for points non‐uniformly (ε‐sampling) sampled from simple, closed, concave and smooth curves. The proposed framework has been thoroughly evaluated for its usefulness using various test data. Results indicate that even sparsely and non‐uniformly sampled curves with outliers or collection of curves are faithfully reconstructed by the proposed algorithm. 相似文献

The present study evaluates a hypothesis that diet-related hypercholesterolemia increases oxidative stress-related burden to cardiovascular tissue, resulting in progressively increased mortality, along with deterioration of electrophysiological and enzymatic function in rabbit myocardium. New Zealand white rabbits were divided into four groups, defined as follows: GROUP I, cholesterol-free rabbit chow for 12 weeks; GROUP II, cholesterol-free chow, 40 weeks; GROUP III, chow supplemented with 2% cholesterol, 12 weeks; GROUP IV, chow supplemented with 2% cholesterol, 40 weeks. At the 12 and 40 weeks time points, animals in each of the aforementioned cohorts were subjected to echocardiographic measurements, followed by sacrifice. Significant deterioration in major outcome variables measured in the present study were observed only in animals maintained for 40 weeks on 2% cholesterol-supplemented chow, with much lesser adverse effects noted in animals fed high cholesterol diets for only 12 weeks. It was observed that rabbits receiving high cholesterol diets for 40 weeks exhibited significantly increased mortality, worsened ejection fraction and general deterioration of cardiac functions, along with increased atherosclerotic plaque formation and infarct size. Additionally, myocardium of GROUP IV animals was observed to contain lower levels of heme oxygenase-1 (HO-1) and cytochrome c oxidase III (COX III) protein relative to the controls. 相似文献
