Detection of recurrence of medullary thyroid carcinoma (MTC) remains a diagnostic problem. Increased serum tumour marker levels frequently indicate recurrence while conventional imaging techniques (CIT) are non-diagnostic. In this study, we performed indium-111 octreotide scintigraphy and CIT in a series of 20 patients with MTC presenting with elevated serum tumour markers after surgery. 111In-octreotide whole-body studies detected 15 pathological uptake foci in 11 of the 20 patients studied and CIT detected 17 lesions in 11 of the 20 patients. Ten patients underwent reoperation, five of them with positive 111In-octreotide scintigraphy and CIT and two with positive isotopic exploration and negative CIT. Surgical findings demonstrated that the results of isotopic study and CIT had been false-positive for MTC in one case (sarcoidosis). The six patients with true-positive 111In-octreotide studies had significantly higher basal calcitonin (CT) and carcinoembryonic antigen (CEA) levels than the patients with negative isotopic studies. The expression of somatostatin receptor (SSTR) subtypes by PC-PCR could be investigated in four cases with a positive isotopic study. Among the three cases with a true-positive study, SSTR2, the SSTR subtype that preferentially binds to the somatostatin analogue octreotide, was detected in two, SSTR5 was demonstrated in the three, and SSTR3 was detected in one. No subtype of SSTR was detected in the case with a final diagnosis of sarcoidosis. We conclude that 111In-octreotide has limited sensitivity in detecting recurrence in patients with MTC, although its sensitivity may improve with high serum CT levels. This radionuclide imaging technique should be employed when conventional imaging techniques are negative or inconclusive or when the presence of somatostatin receptors may provide the basis for treatment with somatostatin analogues. 相似文献

The microstructure of silicon carbide whiskers synthesized by carbothermal reduction of silicon nitride has been studied using transmission electron microscopy. All of the whiskers examined are single crystals, and grow in the (111) crystallographic direction. Two different forms of stacking faults and microtwins were observed; in one the planar defects are normal to the whisker growth direction, and the other has the defect planes at an angle of about 70° to the growth axis, while both forms of the defects are on the [111] closed-packed planes. Without the addition of catalyst, droplets containing metallic impurities were not found at the tips of the whiskers synthesized by the present process. A core and outer regions were observed in the single-crystal whiskers, which may be evidence that the whiskers were formed by a two-stage mechanism. 相似文献

Two constitutive models of PSA rheology, i.e., Newtonian and Power Law, are considered and experimentally tested for their ability to predict shear holding power. A Power Law model is found to be suitable for this purpose and is used to predict shear holding power with dynamic viscosity and steady shear viscosity measured by means of a transient creep experiment. Dynamic viscosity is shown to fail in predicting shear adhesion of high-viscosity PSA, whereas steady shear produces good agreement. We have recently shown that docosahexaenoic acid (DHA) is necessary for survival and differentiation of rat retinal photoreceptors during development in vitro. In cultures lacking DHA, retinal neurons developed normally for 4 d; then photoreceptors selectively started an apoptotic pathway leading to extensive degeneration of these cells by day 11. DHA protected photoreceptors by delaying the onset of apoptosis; in addition, it advanced photoreceptor differentiation, promoting opsin expression and inducing apical differentiation in these neurons. DHA was the only fatty acid having these effects. Mitochondrial damage accompanied photoreceptor apoptosis and was markedly reduced upon DHA supplementation. This suggests that a possible mechanism of DHA-mediated photoreceptor protection might be the preservation of mitochondrial activity; a critical amount of DHA in mitochondrial phospholipids might be required for proper functioning of these organelles, which in turn might be essential to avoid cell death. Müller cells in culture appeared to be involved in DHA processing: they took up DHA, incorporated in into glial phospholipids, and channeled it to photoreceptors in coculture. Both Müller cells, when cocultured with neuronal cells, and the glial-derived neurotrophic factor (GDNF) protected photoreceptors from cell death. These results suggest that glial cells may play a central role in regulating photoreceptor survival during development through the provision of trophic factors. The multiple effects of DHA on photoreceptors suggest that in addition to its structural role, DHA might be one of the trophic factors required by these cells. 相似文献

Tumor-infiltrating immune cells phenotype is associated with tumor progression. However, little is known about the phenotype of the peripheral blood mononuclear cells (PBMC) from breast cancer patients. We investigated MMP1 and MMP11 expression in PBMC from breast cancer patients and we analyzed gene expression changes upon their interaction with cancer cells and cancer-associated fibroblasts (CAF). We measured the impact of PBMC on proinflammatory gene expression in breast cancer cells, normal fibroblast (NF), and CAF and the impact on proliferation and invasiveness capacity of breast cancer cells. Gene expression of MMP1 and MMP11 in PBMC from breast cancer patients (n = 54) and control (n = 28); expression of IL1A, IL6, IL17, IFNβ, and NFĸB in breast cancer cell lines (MCF-7 and MDA-MB-231); and, additionally, IL10 and MMP11 in CAF and NF were analyzed by qRT-PCR before and after co-culture. Our results show the existence of a subpopulation of breast cancer patients (25.9%) with very high levels of MMP11 gene expression in PBMC. Also, gene expression of MMP1 and MMP11 increases in PBMC after co-culture with breast cancer cell lines, NF or CAF. PBMC from healthy or breast cancer patients induce an increased proliferation rate on MCF-7 and an increased invasiveness capacity of MDA-MB-231. Finally, we show a differential expression profile of inflammatory genes in NF and CAF when co-cultured with control or breast cancer PBMC. We have observed that MMPs’ expression in PBMC is regulated by the microenvironment, while the expression of inflammatory genes in NF or CAF is differentially regulated by PBMC. These findings confirm the importance of the crosstalk between stromal cells and suggest that PBMC would play a role in promoting aggressive tumor behavior. 相似文献

Low dose and non-targeted effect studies continue to attract the attention of a growing number of radiobiologists. Experimental setups based on light ion microbeams constitute a tool of choice for this kind of investigations. However, a careful attention must be given to experimental conditions, as setup-induced stress levels should be well below those induced by the irradiation itself. Here, we present the current status of the single-particle microbeam facility that has been developed these last years at the nuclear microprobe of Saclay. The driving idea was to build a facility in which local irradiation studies are performed in an environment close to cellular biology standards. This facility includes unique features, such as (i) a compact setup that allows easy access and vertical irradiation mode, (ii) a collimated beam that can be mechanically positioned under the desired cells at a very fast speed, avoiding the requirement of a focusing element and (iii) a controlled environment (temperature, CO2, humidity) that allows performing of very long term experiments on cultured cells.Fluorescent techniques are implemented and permit in situ monitoring of cellular responses to irradiations.Several radiobiological studies are already underway and this will be illustrated with recent results regarding DNA damage and reactive oxygen species signaling time courses following targeted irradiations. 相似文献

This paper presents a numerical model for the simulation of a swash-plate axial piston pump, focusing on the characterization of fluid properties.As it is well known, the reduction of flow oscillations (which generates pressure ripples and produces vibration and noise in the entire circuit) and the avoidance of cavitation are the major problems in the design of these pumps. Developing a simulation code can be very useful for component optimisation in order to predict and reduce the undesired phenomena. The paper first gives a quick overview on a previously developed pump model; afterwards four different models of the fluid are presented: they take into account cavitation in different ways. Their aim at characterizing as well as possible the unsteady and erratic cavitation features in a simplified manner, in order to apply the models to the simulation of hydraulic components.In the second part of the paper an application is shown of these models to an axial piston pump: a few results are presented and compared with available test data. Instances of temperature-dependent apparent energies of activation EA, observed for hydrolysis reactions, are briefly reviewed, as are recent calculations according to which the presence of a reactant in two forms which are in mobile equilibrium may lead to a temperature-dependent EA. These calculations are extended to a consideration of the temperature-dependence of the specific heat of activation, and to the possibility of more than two forms being in equilibrium.
