An established senescence-accelerated model mouse strain, SAMP8, shows the deterioration of learning and memory compared with a normal control strain, SAMR1. D-Serine binds to strychnine-insensitive glycine binding sites of the N-methyl-D-aspartate (NMDA) receptor complex, and enhances glutamate binding to the receptor complex. To investigate the relationship of endogenous brain D-serine and the brain dysfunction caused by aging, the level of brain free D-serine and the D-[3H]serine binding to the brain samples were examined using the SAMP8 and SAMR1 mice. The free D-serine level was highest in the cerebral frontal and occipital cortices in both the SAMP8 and SAMR1; no difference in the D-serine level was shown between the two strains. A receptor autoradiographical analysis showed that the D-[3H]serine binding to the brain section was highest in the hippocampus, and the binding in the SAMP8 brains was lower than that of the SAMR1. The D-[3H]serine binding to the crude cerebral membranes indicated that the value of the total binding sites for the SAMP8 was lower than that for the SAMR1, whereas the value of the dissociation constant Kd for the SAMP8 was similar to that of the SAMR1. These results suggest that the number of D-[3H]serine binding sites was decreased in the SAMP8 compared to the SAMR1, but the affinity of D-[3H]serine to the binding sites was not altered. These results support the view that a decrease of NMDA receptor complex is involved in the age-related neural dysfunction of SAMP8 mice.  相似文献

Experimental and computational investigations were performed in order to better understand the mechanical response of rigid targets with various geometries to the detonation of shallow buried explosives. The motion of the targets was measured by use of high-speed digital video photography. This work involved flat targets, targets that were downwardly convex, and targets that were downwardly concave with explosive charges located at various positions beneath the targets. It was observed that, in general, angled hulls - whether downwardly concave or convex - tended to reduce the amount of momentum imparted to the center of mass of the targets. Computations were performed by use of an arbitrary Langrangian-Eulerian treatment in a nonlinear finite element code. A model based on quasi-static test evaluations of wet concrete sand was used for prediction of the soil behavior. The computational technique provided very good agreement between computation and experiment.  相似文献

Pepstatin A, an inhibitor of acid proteases, was added (7.5, 15 or 30 μmol L-1) to the curds/whey mixture at the start of cooking to inhibit residual coagulant in miniature (20 g) Cheddar-type cheeses. No degradation of s1-casein was observed by urea–polyacryl amide gel electrophoresis (PAGE) in the pepstatin-treated cheeses, indicating that all the concentrations of pepstatin used in this study effectively inhibited residual coagulant throughout ripening. The level of water-soluble N (WSN) as % of total N increased very slowly in the pepstatin-treated cheeses, while there was a steady increase in WSN in the control cheeses; after 4 months of ripening, the level of WSN in the control cheese was nearly three times as high as in the cheese treated with 30 μmol L-1 pepstatin. Urea–PAGE of water-soluble fractions (WSF) showed marked differences between pepstatin-treated cheeses and their respective controls throughout ripening. Reverse-phase HPLC of the WSF of the cheeses showed that the peptides s1-CN f1-9/13, which are formed from the chymosin-produced peptide, s1-CN f1-23, by the action of the cell envelope-associated proteinase of Lactococcus, were not present in pepstatin-treated cheeses. Levels of total free amino acids (as determined by the Cd–ninhydrin method) were higher in controls than in pepstatin-treated cheeses throughout ripening. The results of this study demonstrated that pepstatin is a very effective inhibitor of residual coagulant in cheese.  相似文献

Experiment 1 was conducted to determine NAGase activity in skim, fat, and cell pellet fractions of foremilk and stripping milk from infection-free quarters. Changes in milk NAGase activity during a 12 h in vitro incubation were also determined. Eight cows, two quarters per cow, were used. One quarter of each cow received an intramammary infusion of oyster glycogen. N-Acetyl-beta-D-glucosaminidase activity was highest in stripping milk and in milk from infused quarters. The percentages of NAGase activity in skim, fat, and cell pellet fractions were 62.6, 22.4, and 12.6. The NAGase activity of milk incubated in vitro did not significantly change over time. Experiment 2 was conducted to determine if neutrophils lost NAGase activity during extravasation into milk. Leukocytosis was induced in infection-free quarters of five cows. The NAGase activities of peripheral neutrophils and milk neutrophils were not significantly different. Results from both studies suggest that the major source of milk NAGase is the mammary epithelial cell and that milk somatic cells contribute less than 15% of the total milk NAGase activity.  相似文献

Two acidified sodium chlorite postmilking teat disinfectants were evaluated for efficacy against Staphylococcus aureus and Streptococcus agalactiae by using National Mastitis Council experimental challenge procedures. The effect of these teat dips on teat skin and teat end condition was also determined. Both dips contained 0.32% sodium chlorite, 1.32% lactic, and 2.5% glycerin. Dips differed in the amount of sodium dodecylbenzene sulfonic acid (0.53 or 0.27%) added as a surfactant. Both dips significantly reduced new intramammary infection (IMI) rates compared with undipped controls. The dip containing 0.53% dodecylbenzene sulfonic acid reduced new IMI by Staph. aureus by 72% and Strep. agalactiae by 75%. The dip containing 0.27% dodecylbenzene sulfonic acid reduced new IMI by Staph. aureus by 100% and by Strep. agalactiae by 88%. Changes in teat skin and teat end condition for treatment and control groups varied in parallel over time. Teats treated with either teat dip had higher mean teat skin and teat end scores than control teats at some weeks. However, teat skin and teat end condition did not tend to change from the start to the completion of the trial. Application of the two new postmilking teat dips was effective in reducing new IMI from contagious mastitis pathogens. (Key words: teat dip, contagious mastitis, chlorous acid)  相似文献

A study was conducted on 4 dairy farms in the Cannonsville Reservoir Basin (Delaware County, NY) to identify feeding strategies in commercial dairy herds that will reduce manure phosphorus and mass farm phosphorus balance. Lactating cow diets on all 4 farms were evaluated monthly for 28 mo using the Cornell Net Carbohydrate and Protein System. Milk production and herd reproductive performance were measured monthly. Manure phosphorus content was measured every 6 mo. Reduced phosphorus diets (precision feeding) were implemented in 2 of the herds. Mean herd phosphorus intakes in the 4 herds ranged from 107 to 165% of requirement. Dietary phosphorus intakes in the 2 herds where diets were modified were reduced from 153% of requirement to 111%, an average reduction of 25%. Predicted phosphorus intakes and manure excretions were reduced 11.8 kg/yr per cow. After dietary adjustments in the 2 herds, fecal phosphorus concentrations decreased 33%. Milk production was not adversely affected by reduced phosphorus diets. Whole farm mass phosphorus balances (amount of phosphorus remaining on the farm) on the 2 farms were reduced 49%, with the percentage of imported phosphorus remaining on the farm reduced to less than 45%. Achieving feed phosphorus reductions similar to those of this study on all of the estimated 7000 to 8000 mature dairy cattle in the Cannonsville Basin could reduce feed phosphorus imports and manure phosphorus excretions more than 64,000 kg/yr. This would slow the rate of phosphorus accumulation in agricultural soils in the Cannonsville Basin, which over time could reduce the 50,000 kg/yr average total phosphorus loading of the Cannonsville Reservoir.  相似文献

Silicon carbide nanowires (SCNWs) were grown from anthracite fine surfaces through a simple one-step carbothermal process with silicon powder as the Si precursor. This straightforward and fast formation of SCNWs made it possible to maintain the binding of briquetted waste anthracite fines at very high temperatures as an alternative fuel in foundry cupola furnaces. This SCNW mechanism could thus provide the crucial hot crushing strength in the cupola heat zone and melt zone. Progressive thermal tests exhibited that the formation of the SCNWs started from 1100 ?C, and was favored at 1400 ?C. No extra metal catalyst was needed for the growth of the SCNWs. Characterizations were performed by XRD, SEM, EDS, TEM, and SAED. The SCNWs were 30-60 nm in diameter and were typically grown by stacking the (1 1 1) lattice plane of 3C-SiC along the [1 1 1] direction. Many non-epitaxial branches of the nanowires were also formed through this one-step process as observed by TEM. The results suggest that the SCNWs were most likely grown through the vapor-solid mechanism.  相似文献

Pseudomonas strain AFT 21 produced three heat stable extracellular proteinases in milk and nutrient broth at 7 or 21 degrees C, but the proportions depended on medium and cultivation temperature. The three proteinases were EDTA- and o-phenanthroline-sensitive metalloenzymes and were not inhibited by N-ethylmaleimide or phosphoramidon. Proteinases I and II showed maximum activity at pH 7-7.5 and proteinase III at pH 8.5. All three enzymes showed maximum activity at 45-47.5 degrees C, but had relatively high (19-27% of maximum) activity at 4 degrees C. They were unstable at 55 degrees C in phosphate buffer, pH 6.6, or synthetic milk ultrafiltrate (SMUF) containing 12 mmol Ca2+, but were stabilized by short preheating at 100 degrees C. They were extremely heat stable in both phosphate buffer and SMUF, pH 6.6, at 70-150 degrees C. Their D-values at 140 degrees C were 69, 54 and 80 s respectively. The Z-values for Pseudomonas AFT 21 proteinase III in phosphate buffer and SMUF were 29.7 and 30.3 degrees C respectively; the corresponding activation energies for inactivation were 8.7 x 10(4) J mol-1 and 9.2 X 10(4) J mol-1.  相似文献

Forty-four cows (26 Jerseys and 18 Holsteins) that had at least 1 mammary quarter that was naturally (n = 12) or experimentally (n = 84) infected with Staphylococcus aureus were allotted to three treatment groups of approximately equal number at the end of lactation. Cows were dried off by abrupt cessation of milking, and dry cow therapy was administered as an intramammary infusion of cephapirin benzathine at 10 ml per quarter, an intramammary infusion of tilmicosin (solution containing 300 mg/ml) at 5 ml per quarter, or a subcutaneous injection of tilmicosin at 5 mg/kg of body weight on the day of drying off and another injection 4 d later. Mammary secretions were monitored during the dry period and postpartum for antimicrobial residues, intramammary infection (IMI) status, and somatic cell counts. Results demonstrated the following percentage cures for IMI caused by Staph. aureus at 28 d postcalving based on individual mammary quarters: cephapirin benzathine, 78.1%; tilmicosin infused, 74.2%; and tilmicosin injected, 9.1%. During the first 4 wk after drying off, the mean concentration of tilmicosin in mammary secretions from cows infused with the antibiotic remained approximately 10-fold higher than that in secretions from cows injected with the antibiotic (3.43 vs. 0.32 ppm), and, by the time of calving, concentrations for cows treated with both methods were below the dilution limit of the assay (< 0.1 ppm). Results demonstrated that intramammary infusion of tilmicosin was equally as effective as cephapirin benzathine in curing IMI caused by Staph. aureus at drying off; however, the subcutaneous injection of tilmicosin at the dose used was not effective as a dry cow therapeutic against Staph. aureus.  相似文献
