A multiresidue analytical procedure for determination of seven fluoroquinolones (marbofloxacin, norfloxacin as internal standard, ciprofloxacin, danofloxacin, enrofloxacin, sarafloxacin and difloxacin), and three quinolones (oxolinic acid, nalidixic acid and flumequine) in eggs is presented. The procedure is based on dispersive solid-phase extraction technique with acetonitrile as extractant. Norfloxacin and ciprofloxacin - d8 were used as internal standards to quantify the (fluoro)quinolones. Analyses were realised by LC-FLD for screening and LC-MS/MS for confirmatory purposes. The whole procedure was evaluated according to the Commission Decision 2002/657/EC. Specificity, decision limit (CCα), detection capacity (CCβ), recovery (absolute and relative), precision (repeatability and reproducibility) were determined during validation process. Recoveries (relative) for the LC-FLD screening determination ranged from 85% to 93%, repeatability and reproducibility were in the range of 5-9% to 9-16%, respectively. CCα and CCβ were 13-37 and 17-43 μg/kg pending on analite. For the LC-MS/MS confirmatory method, the relative recoveries were satisfactory (92-99%) with repeatability and reproducibility in the range of 4-7% to 6-12%, respectively. CCα and CCβ were 3-7 and 7-11 μg/kg depending on the analite. The results of both prepared methods showed these analytical procedures simple, rapid, sensitive and suitable for routine control of eggs.  相似文献

In recent years, there has been a particular focus on the application of antimicrobial compounds produced by lactic acid bacteria (LAB) as natural preservatives to control the growth of spoilage and pathogenic bacteria in food. Bacteriocins are antimicrobial peptides which can be added to foods in concentrated forms as food preservatives, e.g. additives, or they can be produced in situ by starters or protective cultures. In this study, twenty Lactococcus lactis bacteriocin producers previously isolated from Italian fermented foods were subjected to a variety of physical and biochemical tests in order to identify those with the greatest potential as starter cultures in cheese production. Of these, four strains isolated from cheese (one nisin Z producer, one nisin A producer and two lacticin 481 producers) which fulfilled the desired technological criteria were assessed for their ability to control Listeria monocytogenes. The subsequent application of these bacteriocinogenic strains as starter cultures in Cottage cheese established that the nisin A producing Lact. lactis 40FEL3, and to a lesser extent the lacticin 481 producers 32FL1 and 32FL3, successfully controlled the growth of the pathogen. This is the first study to directly compare the ability of nisin A, nisin Z and lacticin 481 producing strains to control listerial growth during the manufacture and storage of Cottage cheese.  相似文献

Motor vehicles are a major source of polycyclic aromatic hydrocarbon (PAH) emissions in urban areas. Motor vehicle emission control strategies have included improvements in engine design, exhaust emission control, and fuel reformulation. Therefore, an updated assessment of the effects of the shifts in fuels and vehicle technologies on PAH vehicular emission factors (EFs) is needed. We have evaluated the effects of ambient temperature on the size-resolved EFs of nine US EPA Priority Pollutant PAH, down to 10 nm diameter, from on-road California gasoline light-duty vehicles with spark ignition (SI) and heavy-duty diesels with compression ignition (CI) in summer 2004 and winter 2005. During the winter, for the target PAH with the lowest subcooled equilibrium vapor pressure --benzo[a]pyrene, benzo[ghi]perylene, and indeno[1,2,3-cd]pyrene-- the mass in the nucleation mode, defined here as particles with dp <32 nm, ranged between 14 and 38% for SI vehicles and 29 and 64% for CI vehicles. Our observations of the effect of temperature on the mass of PAH in the nucleation mode are similar to the observed effect of temperature on the number concentration of diesel exhaust particles in the nucleation mode in a previous report.  相似文献

Sertoli cell proliferation occurs in two major waves after birth, one neonatally and another prepubertally, each contributing to final testicular size and sperm production. However, little is known about the regulation of either wave. We have previously shown that letrozole, an inhibitor of estrogen synthesis, increases Sertoli cell number and testicular size at sexual maturity in boars. These studies were conducted to determine whether letrozole affects the first or second proliferative wave. Boars were treated with letrozole during the first wave (treatment at 1, 3, and 5 weeks), less frequently (1 week of age only, or 1 and 5 weeks), on postnatal day 1, or during the second wave (weeks 11-16). Sertoli cells were enumerated in testes and estrogen concentrations were evaluated in serum and testes. Compared with vehicle controls, letrozole reduced estrogen in boars treated at weeks 1 and 5 or 1, 3, and 5, on postnatal day 1, or prepubertally. However, Sertoli cell numbers were increased only in boars treated at 1, 3, and 5 weeks of age. Neither perinatal (1 day old) nor prepubertal letrozole treatment affected Sertoli cell numbers. Hence, Sertoli cell proliferation was sensitive to letrozole only if letrozole was administered throughout the first wave, even though estrogen synthesis was effectively inhibited at all ages. These data indicate that the neonatal but not the prepubertal window of Sertoli cell proliferation is sensitive to an inhibitor of estrogen synthesis; this suggests that these two waves are differently regulated.  相似文献

Geologic carbon sequestration represents a promising option for carbon mitigation. Injected CO(2), however, can potentially leak into water systems, increase water acidity, and mobilize metals. This study used column experiments to quantify the effects of environmental controls on cadmium desorption during CO(2) leakage in subsurface systems without ambient flow. Results show that fast leakage rates are responsible for earlier and larger amounts of Cd desorption. Long weathering time of Cd laden clay leads to low Cd desorption. Calcite content as low as 10% can mitigate the effect of pH reduction and result in zero Cd desorption. Increasing the salinity of the leaking fluid has a relatively minor effect, primarily due to the offsetting impacts of an increased extent of ion exchange and the decrease in CO(2) solubility (and therefore acidity). This work systematically quantifies, for the first time, the effects of environmental controls on Cd desorption and points to key parameters for risk assessment associated with metal mobilization during CO(2) leakage.  相似文献

The modulation of the yield distribution of intermediates formed in the photocatalytic degradation of organic pollutants is of extreme importance for the application of photocatalysis in environmental cleanup, as different intermediates usually exhibit distinct biological toxicity and secondary reactivity. In this paper, we report that the distribution of monohydroxylated intermediates (m-, p- and o-) formed during the photocatalytic oxidation of aromatic compounds changes with the variation of reaction conditions, such as O(2) partial pressure and substrate concentration. By detailed product analysis, theoretical calculation, and oxygen isotope labeling experiments, we show that these changes are due to the selective reduction of HO-adduct radicals (the precursors of hydroxylated intermediates) by conduction band electrons (e(cb)(-)) back to the original substrate, that is, p- and o-HO-adduct radicals are more susceptible to e(cb)(-) than the m- one. Our experiments give an example that, even under oxidative conditions, the yield distribution of isomeric intermediates can be modulated by e(cb)(-)-initiated reduction. This study also illustrates that the unique redox characteristics of photocatalysis, that is, both oxidation and reduction reactions take place on or near the surface of a single nanoparticle, can provide opportunities for the reaction control.  相似文献

Cryptosporidium parvum oocysts have been found on the surface of vegetables in both developed and developing countries. C. parvum can contaminate vegetables via various routes, including irrigation water. This study investigated the effect of individual treatments of chlorine, blanching, blast freezing, and microwave heating, as well as combined treatments of chlorine and freezing, and chlorine and microwave heating on the viability of C. parvum oocysts inoculated on green peppers. The viability of the oocysts after the treatments was assessed using propidium iodide and a flow cytometer. Based on the propidium iodide staining, the chlorine treatments did not affect the viability of the oocysts. Blast freezing significantly inactivated 20% of the oocysts. Microwave heating and blanching significantly inactivated 93% of oocysts. Treatment with chlorine followed by blast freezing did not affect the viability of the oocysts significantly. Treatment with chlorine and microwave heating was significantly more effective than microwave heating alone and inactivated 98% of the oocysts. The study indicates that C. parvum oocysts are sensitive to heat and, to some extent, to blast freezing, but are resistant to chlorine. Therefore, the use of chlorine during vegetable processing is not a critical control point for C. parvum oocysts, and the consumption of raw or minimally processed vegetables may constitute a health risk as C. parvum oocysts can still be found viable on ready-to-eat, minimally processed vegetables.  相似文献

A new strain of denitrifying bacterium, Pseudomonas mendocina LR, was isolated from rhizosphere soil of aquatic plants living in a river contaminated with industrial wastewater and domestic sewage. The isolate was found to fully remove as much as 613.2 mg nitrate in 60 h under stationary culture conditions. The effects of carbon sources and nitrogen sources on nitrogen removal were investigated using a modified denitrification medium (DM). Sodium citrate was identified as the most effective source of carbon. The ability of LR to adapt to different nitrogen sources, including nitrite, indicated that LR could be used in the purification of wastewater containing different forms of nitrogen. The optimal C/N ratio was 7 for LR, and it was resistant to antibiotics Amp, Chl, Ery, and Str. Plant-microbe bioaugmentation was performed to remove nitrogen dissolved in Hoagland medium and natural wastewater. An increased rate of nitrogen removal was observed when root exudates of Cyperus alternifolius L. were added simultaneously with LR. LR was not able to survive in the natural wastewater unless root exudates from umbrella grass were added. LR cultured with umbrella grass exhibited a maximal nitrogen reduction rate of 95.9% and 97.3% in Hoagland medium and wastewater, respectively. This shows that bioaugmentation utilizing plant-microbe interactions can be an effective and exhaustive means of removing nitrogen and may be an attractive approach to nitrogen reduction in natural environments and wastewater treatment factories.  相似文献

The yeast Dekkera/Brettanomyces bruxellensis can cause enormous economic losses in wine industry due to production of phenolic off-flavor compounds. D. bruxellensis is a distant relative of baker's yeast Saccharomyces cerevisiae. Nevertheless, these two yeasts are often found in the same habitats and share several food-related traits, such as production of high ethanol levels and ability to grow without oxygen. In some food products, like lambic beer, D. bruxellensis can importantly contribute to flavor development. We determined the 13.4 Mb genome sequence of the D. bruxellensis strain Y879 (CBS2499) and deduced the genetic background of several "food-relevant" properties and evolutionary history of this yeast. Surprisingly, we find that this yeast is phylogenetically distant to other food-related yeasts and most related to Pichia (Komagataella) pastoris, which is an aerobic poor ethanol producer. We further show that the D. bruxellensis genome does not contain an excess of lineage specific duplicated genes nor a horizontally transferred URA1 gene, two crucial events that promoted the evolution of the food relevant traits in the S. cerevisiae lineage. However, D. bruxellensis has several independently duplicated ADH and ADH-like genes, which are likely responsible for metabolism of alcohols, including ethanol, and also a range of aromatic compounds.  相似文献
