The taxonomic assemblage and functions of the plant bacterial community are strongly influenced by soil and host plant genotype. Crop breeding, especially after the massive use of nitrogen fertilizers which led to varieties responding better to nitrogen fertilization, has implicitly modified the ability of the plant root to recruit an effective bacterial community. Among the priorities for harnessing the plant bacterial community, plant genotype-by-microbiome interactions are stirring attention. Here, we analyzed the effect of plant variety and fertilization on the rhizosphere bacterial community. In particular, we clarified the presence in the bacterial community of a varietal effect of N and P fertilization treatment. 16S rRNA gene amplicon sequence analysis of rhizospheric soil, collected from four wheat varieties grown under four N-P fertilization regimes, and quantification of functional bacterial genes involved in the nitrogen cycle (nifH; amoA; nirK and nosZ) were performed. Results showed that variety played the most important role and that treatments did not affect either bacterial community diversity or bacterial phyla abundance. Variety-specific response of rhizosphere bacterial community was detected, both in relation to taxa (Nitrospira) and metabolic functions. In particular, the changes related to amino acid and aerobic metabolism and abundance of genes involved in the nitrogen cycle (amoA and nosZ), suggested that plant variety may lead to functional changes in the cycling of the plant-assimilable nitrogen.  相似文献

Endocrine-disrupting chemicals (EDCs) are different natural and synthetic chemicals that may interfere with several mechanisms of the endocrine system producing adverse developmental, metabolic, reproductive, and neurological effects in both human beings and wildlife. Among pesticides, numerous chemicals have been identified as EDCs. MicroRNAs (miRNAs) can regulate gene expression, making fine adjustments in mRNA abundance and regulating proteostasis. We hypothesized that exposure to low doses of atrazine, cypermethrin, and vinclozolin may lead to effects on miRNA expression in SH-SY5Y cells. In particular, the exposure of SH-SY5Y cells to subtoxic concentrations of vinclozolin is able to downregulate miR-29b-3p expression leading to the increase in the related gene expression of ADAM12 and CDK6, which may promote a pro-oncogenic response through the activation of the PI3K/Akt/mTOR pathway and counteracting p53 activity. A better understanding of the molecular mechanisms of EDCs could provide important insight into their role in human disease.  相似文献

The detection of ROS1 and ALK rearrangements is performed for advanced-stage non-small cell lung cancer. Several techniques can be used on cytological samples, such as immunocytochemistry (ICC), fluorescence in situ hybridization (FISH) and, more recently, next-generation sequencing (NGS), which is gradually becoming the gold standard. We performed a retrospective study to compare ALK and ROS1 rearrangement results from immunocytochemistry, FISH and NGS methods from 131 cytological samples. Compared to NGS, the sensitivity and specificity of ICC were 0.79 and 0.91, respectively, for ALK, and 1 and 0.87 for ROS1. Regarding FISH, the sensitivity and specificity were both at 1 for ALK and ROS1 probes. False-positive cases obtained by ICC were systematically corrected by FISH. When using ICC and FISH techniques, results are very close to NGS. The false-positive cases obtained by ICC are corrected by FISH, and the true-positive cases are confirmed. NGS has the potential to improve the detection of ALK and ROS1 rearrangements in cytological samples; however, the cost of this technique is still much higher than the sequential use of ICC and FISH.  相似文献

Herein, a simple, solvent-free method to fabricate polymer-encapsulated halide perovskite photoconductors is described. Dry mechanochemical synthesis is used to prepare CsPbBr3 in the presence of poly(butyl methacrylate) (PBMA). The resulting composite powder is then heated and pressed into a free-standing disk with a thickness controlled by a metallic spacer ring. The disk can be laminated on a glass substrate patterned with interdigitated electrodes, resulting in a planar photoconductor device. The best photoconductive performance is obtained for disks that consist of 75 wt.% CsPbBr3 in PBMA, reaching a detectivity of ≈2 × 1011 Jones. Moreover, by adjusting the thickness of the disk, narrowband detectors can be obtained due to charge collection narrowing. Depending on the thickness of the pressed disk, the position and width of the detectivity peak can be tuned. At last, the disks are tested as possible absorber materials for X-ray detectors, where ow detection limit, and fast and linear response are measured for perovskite-polymer disks with 50 wt.% perovskite content. This work shows a simple and versatile approach toward the fabrication of halide perovskite photodetectors, which can be carried out in air and without the use of solvents.  相似文献

A rapid, selective and effective method of extraction, clean-up and concentration of organophosphorous pesticides from wheat followed by electrospray (ESI) LC-MS/MS analysis was developed. The μ-SPE (micro-solid-phase extraction) procedure resulted in good analytical performance and reduced at the same time matrix effects, analysis time and solvent consumption. Limits of detection (LODs) and quantification (LOQs) were in the range of 0.3–10 and 1–30 μg kg?1, respectively, with good reproducibility (RSD ≤ 13.8) and recoveries between 75% and 109%. Coefficients of determination (r2) were greater than 0.996 for the studied pesticides. Despite the reduced sorbent bed mass of μ-SPE tips (4.2 mg), the analytical data showed that no saturation phenomena occurs in the tested range of concentration both for single compounds and mixtures. Several real samples were analysed and the concentrations of the selected pesticides were found to be below the respective maximum residue limit (MRLs).  相似文献

Students identified by talent search programs were studied to determine whether spatial ability could uncover math-science promise. In Phase 1, interests and values of intellectually talented adolescents (617 boys, 443 girls) were compared with those of top math-science graduate students (368 men, 346 women) as a function of their standing on spatial visualization to assess their potential fit with math-science careers. In Phase 2, 5-year longitudinal analyses revealed that spatial ability coalesces with a constellation of personal preferences indicative of fit for pursuing scientific careers and adds incremental validity beyond preferences in predicting math-science criteria. In Phase 3, data from participants with Scholastic Aptitude Test (SAT) scores were analyzed longitudinally, and a salient math-science constellation again emerged (with which spatial ability and SAT-Math were consistently positively correlated and SAT-Verbal was negatively correlated). Results across the 3 phases triangulate to suggest that adding spatial ability to talent search identification procedures (currently restricted to mathematical and verbal ability) could uncover a neglected pool of math-science talent and holds promise for refining our understanding of intellectually talented youth. The relationship between mental rotation ability and gender differences in Scholastic Aptitude Test-Math (SAT-M) across diverse samples was investigated. Talented preadolescents, college students and high- and low-ability college-bound youths, totaling 760, were administered the Vandenberg Mental Rotation Test. Gender comparisons showed male outperforming female students in both mental rotation and SAT-M for all 3 high-ability groups but not for the low-ability group. For all female samples, mental rotation predicted math aptitude even when SAT-Verbal was entered first into the regression. For male samples, the relationship varied as a function of sample. When mental rotation ability was statistically adjusted for, the significant gender difference in SAT-M was eliminated for the college sample and the high-ability college-bound students. This suggests that spatial ability may be responsible in part for mediating gender differences in math aptitude among these groups. Paper-and-pencil tasks showed that many university students believed that when laterally approaching a mirror, they would see a reflection in the mirror before it was geometrically possible. Participants failed to adequately factor in the observer's location in the room. However, when asked about the behavior of a ray of light, participants knew about the law of reflection. No differences between psychology and physics students were detected, suggesting that the phenomenon is widespread and refractory to training. The findings were replicated with observers making judgments about image locations in a real room using a pretend mirror. Possible heuristics about mirror reflection that might explain the data are discussed. Naive optics is a promising venue to further knowledge of how observers understand basic laws of physics. Two studies were conducted to determine (1) whether differential educational experiences contribute to differential growth on Scholastic Aptitude Test (SAT) scores and (2) whether such experiences must occur over a long rather than a short duration to have impact. Specific content knowledge in mathematics/science and verbal areas taught during a short time interval did not increase SAT-M and SAT-V scores even when the content was of the type required to solve SAT problems. Exposure to academically rigorous educational experiences over a long time period (5 yrs) did relate to the development of abilities measured by SAT. In addition, students who experienced very large gains on SAT over this 5-yr period, in comparison with students with small gains, were achieving better in a more rigorous program of high school courses in mathematics and science for the SAT-M and in verbal areas for the SAT-V.
