OBJECTIVES--(1) to evaluate regional cerebral blood flow (rCBF) with single photon emission computed tomography and 99mTc-hexamethylpropyleneamine oxime in patients with the idiopathic adult hydrocephalus syndrome (IAHS); (2) to examine regional cerebral blood flow (rCBF), gait, and psychometric functions before and after CSF removal (CSF tap test); (3) to assess abnormalities in subcortical white matter by MRI. METHODS--Thirty one patients fulfilling the criteria for IAHS (according to history and clinical and neuroradiological examination) were studied. Quantified gait measurements, psychometric testing, and rCBF before and after removal of CSF were obtained. Pressure of CSF and CSF outflow conductance were investigated with a constant pressure infusion method. Brain MRI was used to quantify the severity of white matter lesions and periventricular hyperintensities. In IAHS a widespread rCBF hypoperfusion pattern was depicted, with a caudal frontal and temporal grey matter and subcortical white matter reduction of rCBF as the dominant feature. Removal of CSF was not accompanied by a concomitant increase in rCBF. Significant white matter lesions were detected only in a minority of patients by MRI. An altered CSF hydrodynamic state with a higher CSF pressure and lower conductance was confirmed. IAHS is characterised by an abnormal CSF hydrodynamic state, associated with a widespread rCBF reduction with preference for subcortical white matter and frontal-temporal cortical regions. Furthermore in most patients MRI did not show white matter changes suggestive of a coexistent subcortical arteriosclerotic encephalopathy. At least in the idiopathic group of patients with AHS, measurements of rCBF before and after temporary relief of the CSF hydrodynamic disturbance will not provide additional information that would be helpful in the preoperative evaluation but is suggestive of a preserved autoregulation of rCBF.  相似文献

Recent advances in Wireless Mesh Networks (WMNs) have overcome the drawbacks of traditional wired and ad-hoc networks and now they are seen as a means of allowing last mile communications with quality level assurance in Future Multimedia Systems. However, new routing schemes are needed to provide end-to-end Quality of Service (QoS) and Quality of Experience (QoE) support for delay/loss/jitter-sensitive multimedia applications. The well-known OLSR (Optimized Link State Routing) protocol with ETX (Expected Transmission Count) metric brings many benefits to the path selection process, but has a drawback with regard to queue availability management, which reduces the system performance. This problem is caused when OLSR-EXT control messages are exchanged and the queues of mesh routers along the end-to-end communication path are overloaded. As a result, multimedia-related packets will suffer from loss/delay/jitter and the overall system performance will decrease. This paper proposes the Optimized Link State Routing-Fuzzy ETX Queue (OLSR-FEQ) protocol to overcome the limitations of OLSR-ETX regarding queue availability, QoS and QoE assurance. OLSR-FEQ optimizes network and user-based parameters by coordinating queue availability, QoS and fuzzy issues in the routing decision process as a way of allocating the best paths for multimedia applications. Performance evaluations were carried out with the Network Simulator (NS-2.34) to show the benefits of the proposed solution when compared with existing routing schemes, namely OLSR-ETX, OLSR-FLC, OLSR-MD and HWMP (IEEE 802.11s standard), regarding QoS (unsuccessful packet delivery and throughput) and QoE (PSNR, SSIM, VQM and MOS) parameters.  相似文献

Experimental investigations have been made in which single crystals of solid explosives such as cyclotrimethylene trinitramine, RDX, pentaerythritol tetranitrate, PETN, AgN3 and Pb(N3)2, were irradiated with a focused laser beam of wavelength 266 nm and duration 5 nsec in an ultra high vacuum system housing a fast and sensitive time-of-flight mass spectrometer. From the mass spectra (both +ve and -ve ions) of the chemical species obtained, possible decomposition schemes of RDX and PETN have been proposed. For silver and lead azides, the laser irradiation did not cause explosions if the power density was sufficiently low, but micrometre size craters were formed. In the mass spectra of AgN3 products N3 and silver cluster ions, Ag n +, have been found for the first time. It has been suggested that such investigations may possibly throw some light on our understanding of the reactions taking place in a deflagration/detonation wave.  相似文献

About 50% of patients with arrhythmogenic cardiomyopathy (ACM) carry a pathogenic or likely pathogenic mutation in the desmosomal genes. However, there is a significant number of patients without positive familial anamnesis. Therefore, the molecular reasons for ACM in these patients are frequently unknown and a genetic contribution might be underestimated. Here, we used a next-generation sequencing (NGS) approach and in addition single nucleotide polymor-phism (SNP) arrays for the genetic analysis of two independent index patients without familial medical history. Of note, this genetic strategy revealed a homozygous splice site mutation (DSG2–c.378+1G>T) in the first patient and a nonsense mutation (DSG2–p.L772X) in combination with a large deletion in DSG2 in the second one. In conclusion, a recessive inheritance pattern is likely for both cases, which might contribute to the hidden medical history in both families. This is the first report about these novel loss-of-function mutations in DSG2 that have not been previously identi-fied. Therefore, we suggest performing deep genetic analyses using NGS in combination with SNP arrays also for ACM index patients without obvious familial medical history. In the future, this finding might has relevance for the genetic counseling of similar cases.  相似文献

Bioluminescent tools have been used for decades to image processes in complex tissues and preclinical models. However, few distinct probes are available to probe multicellular interactions. We and others are addressing this limitation by engineering new luciferases that can selectively process synthetic luciferin analogues. In this work, we explored naphthylamino luciferins as orthogonal bioluminescent probes. Three analogues were prepared using an optimized synthetic route. The luciferins were found to be robust emitters with native luciferase in vitro and in cellulo. We further screened the analogues against libraries of luciferase mutants to identify unique enzyme-substrate pairs. The new probes can be used in conjunction with existing bioluminescent tools for multi-component imaging.  相似文献

A number of lipid products are formed during the desaturation of stearyl CoA by hen liver microsomes. This article presents an analysis of the products formed when [1-14C] stearyl CoA is incubated with hen liver microsomes for various time periods. [1-14C] Oleyl CoA was the first radioactive unsaturated product formed. Synthesis of phospholipids containing [1-14C] oleic acid occurs only after the desaturase activity has begun to decline. The specific radioactivity of [1-14C] oleyl CoA was similar to the specific radioactivity of [1-14C] stearyl CoA at all time periods tested. The specific radioactivities of [1-14C] oleic acid and phospholipids containing [1-14C] oleic acid were much lower than that of the [1-14C] stearyl CoA.  相似文献

Summary In this article, cellulose acetate produced through the homogeneous acetylation of sugar cane bagasse cellulose was used to produce membranes, using poly(ethyleneglycol) 600 (PEG 600) as an admixture. The membranes were characterized using water flux measurements (Payne’s cup), differential scanning calorimetry (DSC) and neutral red uptake (cytotoxicity). The results showed that PEG 600 acts as a crystallinity inductor and/or pore former in the cellulose acetate matrix. The induction of crystallinity is important for this system since it had not been reported on the literature yet. The results also demonstrated that the studied membranes present a nontoxic behavior.  相似文献

The aim of this study was to characterize the fatty acid composition of the core and membrane of differently sized milk fat globules separated by microfiltration, which can now be used to manufacture dairy products. Native milk fat globules of various mean diameters, ranging from d43 = 2.3 µm to 8.0 µm, were obtained using microfiltration of raw whole milk. After milk fat globule washing, the milk fat globule membrane (MFGM) and the triglyceride core (TC) were separated by manual churning. After total lipid extraction from each fraction, their fatty acid composition was characterized using methyl ester analysis by gas chromatography. Regardless of season, no significant differences were observed in the fatty acid composition of the MFGM phospholipids. Conversely, significant differences were found in the fatty acid composition of TC; particularly, small fat globule TC contained more medium‐chain fatty acids and less stearic acid than large fat globule TC. These results show that the previously observed differences in total fatty acid composition among differently sized milk fat globules are due to their triglyceride composition and MFGM amount rather than to the composition of the MFGM.  相似文献
