In order to tackle the study of DNA repair pathways, the physical and chemical agents creating DNA damage, the genotoxins, are frequently employed. Despite their utility, their effects are rarely restricted to DNA, and therefore simultaneously harm other cell biomolecules. Methyl methanesulfonate (MMS) is an alkylating agent that acts on DNA by preferentially methylating guanine and adenine bases. It is broadly used both in basic genome stability research and as a model for mechanistic studies to understand how alkylating agents work, such as those used in chemotherapy. Nevertheless, MMS exerts additional actions, such as oxidation and acetylation of proteins. In this work, we introduce the important notion that MMS also triggers a lipid stress that stems from and affects the inner nuclear membrane. The inner nuclear membrane plays an essential role in virtually all genome stability maintenance pathways. Thus, we want to raise awareness that the relative contribution of lipid and genotoxic stresses when using MMS may be difficult to dissect and will matter in the conclusions drawn from those studies.  相似文献

We present a method to control the work function of metallic carbon nanotube transparent electrodes by functionalization of the random network with metallic nanoparticles. Flexible functionalized electrodes with high transparency (~90%) can be obtained with work function values ranging from ~4.6 up to ~5.1 eV, depending on the nature of the nanoparticles. The work function values were obtained by comparative in situ Kelvin probe force microscopy under ultra high vacuum and ultraviolet photoelectron spectroscopy measurements. Interestingly, by appropriate choice of the metal source for functionalization, work function engineering can lead to work function values higher or lower than that for pristine metallic nanotubes. This could be of great interest for adjusting the work function of transparent electrodes to active layers in many optoelectronics devices.  相似文献

Organic fluorescent molecules are infiltrated in the channels of zeolite L nanocrystals, thus creating organic–inorganic fluorescent nanoparticles. Combined with dielectric matrices, these fluorescent nanopigments open the way to the realization of novel optical devices. In this paper, the optical measurement of the quantum yield of fluorescent zeolites by means of a precise and reliable diffuse reflectance technique is presented. Several possible factors that may affect the fluorescence quantum yield are also investigated.  相似文献

Many synthetic polycationic vectors for non-viral gene delivery show high efficiency in vitro, but their usually excessive charge density makes them toxic for in vivo applications. Here we describe the synthesis of a series of high molecular weight terpolymers with low charge density, and show that they exhibit efficient gene delivery, some surpassing the efficiency of the commercial transfection reagents Polyethylenimine and Lipofectamine 2000. The terpolymers were synthesized via enzyme-catalyzed copolymerization of lactone with dialkyl diester and amino diol, and their hydrophobicity adjusted by varying the lactone content and by selecting a lactone comonomer of specific ring size. Targeted delivery of the pro-apoptotic TRAIL gene to tumour xenografts by one of the terpolymers results in significant inhibition of tumour growth, with minimal toxicity both in vitro and in vivo. Our findings suggest that the gene delivery ability of the terpolymers stems from their high molecular weight and increased hydrophobicity, which compensates for their low charge density.  相似文献

This work describes the development, implementation, and assessment of enhanced variants of three different groups of bio‐inspired methodologies: genetic algorithms, particle swarm optimization, and artificial immune system. The algorithms are implemented on a computational tool for the synthesis and optimization of offshore oil production risers that connect a floating platform at the sea surface to the wellheads at the sea bottom. Optimization procedures using bio‐inspired algorithms for such real‐world engineering problems require the calculation of the objective function through a large number of time‐consuming finite element nonlinear dynamic analyses, for the evaluation of the structural behavior of each candidate configuration. Therefore, the performance of the algorithms may be measured by the smaller number of objective function evaluations associated to a given target fitness value. The results indicate that the artificial immune system approach, incorporating some enhancements presented in this work, is more effective than the genetic algorithms and particle swarm optimization methods, requiring a smaller number of evaluations to obtain better solutions. The treatment of oxidized Cu surfaces using an alkanethiol as a reducing agent has been investigated. Exposure to a dilute solution of 1-decanethiol resulted in the complete removal and/or conversion of CuO and subsequent formation of a passivating thiolate film, a so-called self-assembled monolayer (SAM), on the underlying Cu/Cu2O surface as evidenced by x-ray photoelectron spectroscopy (XPS) analysis. Morphological changes, monitored by scanning electron microscopy (SEM) and atomic force microscopy (AFM), revealed transformation of the rough, porous CuO layer into a comparatively smooth Cu/Cu2O surface. Experiments performed on integrated circuit back-end-of-line (BEOL) die structures, comprising Cu/SiO2 bond pads used as substrates for Cu wire bonding, demonstrate the potential application of a thiol-based in-situ cleaning-passivation procedure in microelectronics.  相似文献

In this paper we show that thiolated self-assembled monolayers (SAMs) can be used to anchor source–drain gold electrodes on the substrate, leading to excellent electrical performances of the organic field-effect transistor (OFET) on a par with those using a standard electrode process. Using an amorphous semiconductor and a gate dielectric functionalized with SAMs bearing different dipole moments, we demonstrate that we can tune the threshold voltage alone, while keeping nearly unchanged the other electrical properties (hole carrier mobility, Ion/Ioff ratio, subthreshold swing). This differs from previous studies for which SAMs functionalization induced significant changes in all the OFET electrical performances. This result opens doors to design organic circuits using reproducible amorphous semiconductor based OFETs for which only the threshold voltage can be tuned on demand.  相似文献

The standard modeling framework in functional magnetic resonance imaging (fMRI) is predicated on assumptions of linearity, time invariance and stationarity. These assumptions are rarely checked because doing so requires specialized software, although failure to do so can lead to bias and mistaken inference. Identifying model violations is an essential but largely neglected step in standard fMRI data analysis. Using Lagrange multiplier testing methods we have developed simple and efficient procedures for detecting model violations such as nonlinearity, nonstationarity and validity of the common double gamma specification for hemodynamic response. These procedures are computationally cheap and can easily be added to a conventional analysis. The test statistic is calculated at each voxel and displayed as a spatial anomaly map which shows regions where a model is violated. The methodology is illustrated with a large number of real data examples.  相似文献
