The connection between cytoskeleton alterations and diseases is well known and has stimulated research on cell mechanics, aiming to develop reliable biomarkers. In this study, we present results on rheological, adhesion, and morphological properties of primary rat cardiac fibroblasts, the cytoskeleton of which was altered by treatment with cytochalasin D (Cyt-D) and nocodazole (Noc), respectively. We used two complementary techniques: quartz crystal microbalance (QCM) and digital holographic microscopy (DHM). Qualitative data on cell viscoelasticity and adhesion changes at the cell–substrate near-interface layer were obtained with QCM, while DHM allowed the measurement of morphological changes due to the cytoskeletal alterations. A rapid effect of Cyt-D was observed, leading to a reduction in cell viscosity, loss of adhesion, and cell rounding, often followed by detachment from the surface. Noc treatment, instead, induced slower but continuous variations in the rheological behavior for four hours of treatment. The higher vibrational energy dissipation reflected the cell’s ability to maintain a stable attachment to the substrate, while a cytoskeletal rearrangement occurs. In fact, along with the complete disaggregation of microtubules at prolonged drug exposure, a compensatory effect of actin polymerization emerged, with increased stress fiber formation. 相似文献

In many species, interval timing behavior is accurate—appropriate estimated durations—and scalar—errors vary linearly with estimated durations. Whereas accuracy has been previously examined, scalar timing has not been clearly demonstrated in house mice (Mus musculus), raising concerns about mouse models of human disease. The authors estimated timing accuracy and precision in C57BL/6 mice, the most used background strain for genetic models of human disease, in a peak-interval procedure with multiple intervals. Both when timing 2 intervals (Experiment 1) or 3 intervals (Experiment 2), C57BL/6 mice demonstrated varying degrees of timing accuracy. An important finding was that, both at the individual and group levels, their precision varied linearly with the subjective estimated duration. Further evidence for scalar timing was obtained using an intraclass correlation statistic. This is the first report of consistent, reliable scalar timing in a sizable sample of house mice, thus validating the peak-interval procedure as a valuable technique, the intraclass correlation statistic as a powerful test of the scalar property, and the C57BL/6 strain as a suitable background for behavioral investigations of genetically engineered mice modeling disorders of interval timing. A wavelength-independent method for optical gating, based on the optical Kerr effect, has been demonstrated. Using this method, we produced 100-ps, 10-kW, two-wavelength pulses (10.3 and 10.6 microm) with a signal-to-background ratio contrast of 10(5) by slicing a long CO2 pulse. A gas sensor for application in water analysis was developed by combination of a mid-infrared (MIR) hollow waveguide with a Fourier transform infrared (FT-IR) spectrometer and coupling of the hollow waveguide gas sensor module to a supported capillary membrane sampler (SCMS) for continuous liquid-gas extraction. Different hollow waveguides have been characterized in this study for developing an optimized optical configuration. Analysis of industrially relevant compounds has been performed, investigating chlorinated hydrocarbons (CHCs), such as dichloromethane and chloroform, representing highly volatile analytes, and 1,4-dioxane as an example of target compounds with low volatility. The suitability of this spectroscopic IR sensing system for industrial applications is demonstrated under simulated real-world conditions with limits of detection in the ppb (v/v) and ppm (v/v) concentration range for CHCs and 1,4-dioxane, respectively. 相似文献

This paper presents an alternative approach to obtaining crack- and pore-free NiCrBSiFeCuMoC hard coatings on a low alloy steel substrate through coaxial laser cladding, by using Inconel 718 as a buffer layer between the hard coating and the base material. The presence of the buffer layer reduces the overall cracking susceptibility of the hardfacing material by reducing the compressive stresses developed during the cladding process and ensuring a more uniform heat distribution gradient at the surface of the material than the base metal alone, which provides an additional hardness and wear coefficient increase of 7% and improves the corrosion resistance of the obtained materials by 20%, in comparison with the reference sample obtained without an intermediate layer, by using the same operational parameters and minimizes elemental dilution with the substrate. Our method could prove useful in increasing the quality and life cycle of expensive high-performance hard-coated materials, especially those working under demanding operational and environmental conditions. 相似文献

Porous mullite ceramics with unidirectionally oriented pores were prepared by an extrusion method using rayon fibers as the pore formers. Rayon fibers of 8.1, 9.6, 16.8, and 37.6 μm in diameter were used as the pore formers and were kneaded with alumina powder, kaolin clay, China earthen clay, and water to form pastes. These pastes were extruded into cylindrical tubes, dried, and fired at 1500 °C for 4 h. The apparent porosities ranged from 45.7 to 48.2 %. The pore size distributions showed a sharp peak at 9.4, 10.0, 15.6, and 30 μm with increasing fiber diameters. The height of the capillary rise was 1780, 1670, 1320, and 950 mm with increasing fiber diameter. The maximum capillary rise is much higher than previously reported. The contact angle and effective pore radius that determine the capillary rise ability were calculated by fitting the capillary rise curves using the Fries and Dreyer’s equation. 相似文献

In pulsed eddy current testing, repetitive excitation signals with different duty cycles have different spectral representations. This work studies the influence of duty cycle on the ability to detect holes and EDM notches beneath rivet heads in subsurface layers of stratified samples. Feature patterns for the integrity of rivet joints are proposed and verified. The proposed method has the added advantage in that no reference sample is needed while employing multiple pulse measurements, with different pulse widths. Experimental testing and modelling approaches are discussed in connection with defect depth quantification, which can be extended to the quantification of complex defects. 相似文献

Humans and animals process temporal information as if they were using an internal stopwatch that can be stopped and reset, and whose speed is adjustable. Previous data suggest that dopaminergic drugs affect the speed of this internal stopwatch. Using a paradigm in which rats have to filter out the gaps that (sometimes) interrupted timing, the authors found that methamphetamine and haloperidol also affect the stop and reset mechanism of the internal clock, possibly by modulating attentional components that are dependent on the content and salience of the timed events. This is the fist report of both clock and attentional effects of dopaminergic drugs on interval timing in the same experimental setting. Boundary layer modeling of reactive flow over a porous surface with angled injection 总被引:1,自引:0,他引:1
An analytical model was developed to investigate the dynamics of nonpremixed flames in a shear layer established between a mainstream flow of fuel-rich combustion products and a porous surface with an angled injection of air. In the model, a one-step overall chemical reaction was employed, together with boundary layer conservation equations solved using similarity solutions. Parametric studies were performed to understand the effects of equivalence ratio, temperature, and mass flow rate of the fuel and air streams on the flame standoff distance, surface temperature, and heat flux at the surface. The analytical model predictions were compared with computational fluid dynamics results obtained using the FLUENT commercial code for both the laminar and the turbulent flow models. Qualitative agreement in surface temperature was observed. Finally, the flame stability limits predicted by the model were compared with available experimental data and found to agree qualitatively, as well. 相似文献

Previous data suggest that rats (Rattus norvegicus) and pigeons (Columba livia) use different interval-timing strategies when a gap interrupts a to-be-timed signal: Rats stop timing during the gap, and pigeons reset their timing mechanism after the gap. To examine whether the response rule is controlled by an attentional mechanism dependent on the characteristics of the stimuli, the authors manipulated the intensity of the signal and gap when rats and pigeons timed in the gap procedure. Results suggest that both rats and pigeons stop timing during a nonsalient gap and reset timing after a salient gap. These results also suggest that both species use similar interval-timing mechanisms, influenced by nontemporal characteristics of the signal and gap.
