Breast cancer (BC) is a malignant neoplasia with the highest incidence and mortality rates in women worldwide. Currently, therapies include surgery, radiotherapy, and chemotherapy, including targeted therapies in some cases. However, treatments are often associated with serious adverse effects. Looking for new options in BC treatment, we evaluated the therapeutic potential of cold atmospheric plasma (CAP) in two cell lines (MCF7 and HCC1806) with distinct histological features. Apoptosis seemed to be the most prevalent type of death, as corroborated by several biochemical features, including phosphatidylserine exposure, the disruption of mitochondrial membrane potential, an increase in BAX/BCL2 ratio and procaspase 3 loss. Moreover, the accumulation of cells in the G2/M phase of the cell cycle points to the loss of replication ability and decreased survival. Despite reported toxic concentrations of peroxides in culture media exposed to plasma, intracellular peroxide concentration was overall decreased accompanying a reduction in GSH levels shortly after plasma exposure in both cell lines. In HCC1806, elevated nitric oxide (NO) concentration accompanied by reduced superoxide levels suggests that these cells are capable of converting plasma-derived nitrites into NO that competes with superoxide dismutase (SOD) for superoxide to form peroxinitrite. The concomitant inhibition of the antioxidative activity of cells during CAP treatment, particularly the inhibition of cytochrome c oxidase with sodium azide, synergistically increased plasma toxicity. Thus, this in vitro research enlightens the therapeutic potential of CAP in the treatment of breast cancer, elucidating its possible mechanisms of action.  相似文献

Electrospun fibers of poly[(9,9‐dioctylfluorenyl‐2,7‐diyl)‐co‐bithiophene] (F8T2) with exceptional electro‐optical performance are obtained. The I/T characteristics measured in fibers with 7–15 µm diameter and 1 mm length show a semiconductor behavior; their thermal activation energy is 0.5 eV and the dark conductivity at RT is 5 × 10?9 (Ω cm)?1. Besides exhibiting a photosensitivity of about 60 under white light illumination with a light power intensity of 25 mW · cm?2, the fibers also attain RT photoluminescence in the cyan, yellow, and red wavelength range under ultraviolet, blue, and green light excitation, respectively. Optical microscope images of F8T2 reveal homogeneous electrospun fibers, which are in good agreement with the uniformly radial fluorescence observed.

Isothermal storage and reduction of NO2 with CO, C3H6 and H2 as reducing agents on a lean NO x adsorber was investigated by temperature programmed desorption (TPD) and temperature programmed reduction (TPR) studies. The reduction of NO x was clearly favoured with H2 as reducing agent. Carbon monoxide and C3H6 showed fairly low reduction of NO x . The NO x reduction at low temperatures with H2 as reducing agent was found to be effective, clearly much more effective than for CO.  相似文献

Scratching of thin film/substrate structures is studied theoretically and numerically. The results are discussed in connection to delamination initiation and in particular subsequent growth at scratching. The material behavior of the film is described by classical elastoplasticity accounting for large deformations. The deformation of the substrate is neglected indicating that the results are pertinent to soft thin films. The numerical investigation is performed using the finite element method (FEM) and the numerical strategy is discussed in some detail. The results from this study show that delamination growth at thin film scratching is a stable feature with crack arrest occurring at a decreasing load.  相似文献

Isl-1 is a member of a family of Homeodomains containing proteins that possess an N-terminal pair of zinc binding LIM domains. The Isl-1 gene in rat codes for a protein that binds to the insulin gene enhancer and is also involved in regulation of amylin and proglucagon genes. A DNA sequence coding for 66 amino acid residues containing the C-terminal homeodomain fragment of Isl-1 was expressed as a soluble protein in Escherichia coli. Here, we describe a procedure which allows the rapid native purification of recombinant homeodomain protein fused to an N- terminal tag of six histidines. The purified homeodomain showed DNA- binding activity to its cognate DNA sequence. An enhanced binding activity is observed in the presence of a reducing agent in electrophoretic mobility shift assays. The DNA binding was further characterized by circular dichroism spectroscopy. Addition of DNA to the homeodomain did not change the overall secondary structure content, but the thermal and chemical denaturing profiles were altered. A stabilization of the secondary structure was observed upon DNA binding. The free energy of unfolding at 23 degrees C was 7 kJ mol(-1) in absence of DNA and 29 kJ mol(-1) in the presence of DNA.   相似文献

Corrosion of steel rebars in reinforced concrete is one of the major problems in the construction industry. Carbonation reactions of concrete with carbon dioxide and, mainly, the chloride salts action are the main causes responsible for concrete degradation. Protective coatings help to improve the durability of concrete structures by acting as a physical barrier against the corrosion agents. Waterborne paints are usually used for concrete protection rather than solvent-based paints since they are less pollutant. The aim of this work is to investigate the influence of the pore size and porosity on the permeability of the paints films toward sodium chloride. Three characterization methods from membrane science were implemented to characterize paint coatings. The time-lag method was used to determine the permeability toward the sodium chloride and toward helium and argon, these for approximately 100% relative humidity. From the seven waterborne paints formulated, only one was found to be suitable for surface protection of reinforced concrete, since its permeability toward NaCl was smaller than 10−14 m2 s−1, the threshold value required by National Laboratory of Civil Engineering (LNEC) in Portugal. For the formulated paints, it was observed that the average pore size correlates well with the permeability toward sodium chloride. This is an important result since obtaining the permeability toward sodium chloride of corrosion protective paints is very time consuming, while the average pore size can be obtained in a much shorter time.  相似文献

Previous attempts to use polylactide (PLA) latex particles and nanofibrillated cellulose (NFC) in papermaking processing have been limited to low NFC content. In the present study, a bionanocomposite material was successfully produced using a PLA latex and NFC. The components were mixed using a wet mixing method and bionanocomposite films were made by filtration followed by hot pressing. In composite materials, the dispersion of the reinforcing component in the matrix is critical for the material properties. Biopolymers such as PLA are non-polar and soluble only in organic solvents; NFC is, however, highly hydrophilic. By utilizing latex, i.e., an aqueous dispersion of biopolymer micro-particles, wet mixing is possible and the problem of aggregation of the hydrophilic nanocellulose in organic solvent is avoided. The properties of the resulting NFC/PLA latex bionanocomposite films were analyzed. Thorough blending resulted in good dispersion of the reinforcing component within the matrix. Adding increasing amounts of NFC improved the Young's modulus, tensile strength, and strain at break of the bionanocomposite material. The increase in the tensile properties was linear with increasing NFC content as a result of the good dispersion. Pt/Al2O3 catalysts with platinum contents of 0.1, 1.0 and 3.0 wt% were used for the low temperature combustion of methanol in the absence and the respective presence of a trace amount of ammonia. It is observed that ammonia inhibits the combustion of methanol, which is due to competition between methanol and ammonia for the same sites. For a fixed space velocity and without ammonia in the gas, the performances of the catalysts increase up to a Pt loading of 1.0 wt%. In the presence of ammonia, however, no upper limit of performance is observed with increased Pt content. The results are discussed in terms of both Pt–support interface and the Pt surface being active.  相似文献
