Preparation of conjugated linoleic acid from safflower oil   总被引:5,自引:0,他引:5  
Synthetically prepared mixtures of conjugated linoleic acid (CLA) are widely used in animal and cell culture studies to investigate the potential effects of the Δ9c, 11t-18:2 isomer found in food products from ruminant animals. Alkali isomerization of linoleic acid is a common method used in the synthesis of a mixture of CLA isomers containing predominantly the Δ9c, 11t-18:2 and Δ10t, 12c-18:2 isomers. Some biological activity might also be mediated by the Δ10t, 12c-18:2 isomer. Currently few published methodologies exist describing procedures for the enrichment of these two isomers. A method is described herein to take advantage of an inexpensive oil, safflower oil, for use in synthesis of CLA and a procedure to enrich the Δ10t, 12c-18:2 isomer.  相似文献

Aspergillus awamori glucoamylase (GA) contains globular catalyticand starch-binding domains (residues 1–471 and 509–616,respectively). A heavily O-glycosylated sequence comprises twoparts. The first (residues 441–471) in the crystal structurewraps around an /-barrel formed by residues 1–440. Thesecond (residues 472–508) is an extended, semi-rigid linkerbetween the two domains. To investigate the functional roleof this linker, we made internal deletions to remove residues466–512 (GA1), 485–512 (GA2) and 466–483 (GA3).GA2 and GA3 were expressed in Saccharomyces cerevisiae culturesupernatants at 60 and 20% the wild-type level, respectively,while GA1 was almost undetectable. Western blots comparing extracellularand intracellular fractions indicated that the region deletedin GA3 was critical for secretion, while the region deletedin GA2 contributed to the production of a stable enzyme structure.The activities of purified GA2 and GA3 on soluble and insolublestarch were similar to those of wild-type GA, indicating thatfor soluble starch their deletions did not affect the catalyticdomain and for insoluble starch the linker does not coordinatethe activities of the catalytic and starch-binding domains.The deletions had a significant negative effect on GA2 and GA3thermos tabilities.  相似文献

The recent EU Commission proposal for promoting the supply of power from renewable energy sources was originally based on a pan-European, harmonised tradable green certificate (TGC) scheme. We suggest, on the basis of a multi-disciplinary analysis, that a pan-EU TGC system is not the way forward for Europe. It is vital that the Commission (and the majority of Member States) avoids implementation of such policy designs put forward by a coalition of vested interests. They should instead look at, and act upon, the available evidence from those countries that have experimented with TGCs (e.g. Flanders, UK and Sweden) and design policies that stand a better chance of meeting the criteria of effectiveness, efficiency and equity. In particular, the policies must enable EU to meet the immense innovation/industrialisation challenge by inducing the development of a capital goods industry that can, eventually, diffuse a broad range of technologies that use renewable energy sources. Only then we can acquire an ability to implement an industrial revolution in the energy system in a way that broadly meets the criteria of effectiveness and dynamic efficiency.  相似文献

This study examined the longitudinal effects of 2 first-grade universal preventive interventions on academic outcomes (e.g., achievement, special education service use, graduation, postsecondary education) through age 19 in a sample of 678 urban, primarily African American children. The classroom-centered intervention combined the Good Behavior Game (H. H. Barrish, Saunders, & Wolfe, 1969) with an enhanced academic curriculum, whereas a second intervention, the Family–School Partnership, focused on promoting parental involvement in educational activities and bolstering parents’ behavior management strategies. Both programs aimed to address the proximal targets of aggressive behavior and poor academic achievement. Although the effects varied by gender, the classroom-centered intervention was associated with higher scores on standardized achievement tests, greater odds of high school graduation and college attendance, and reduced odds of special education service use. The intervention effects of the Family–School Partnership were in the expected direction; however, only 1 effect reached statistical significance. The findings of this randomized controlled trial illustrate the long-term educational impact of preventive interventions in early elementary school. This study examined the relationship of early alliance ruptures and their resolution to process and outcome in a sample of 128 patients randomly assigned to 1 of 3 time-limited psychotherapies for personality disorders: cognitive–behavioral therapy, brief relational therapy, or short-term dynamic psychotherapy. Rupture intensity and resolution were assessed by patient- and therapist-report after each of the first 6 sessions. Results indicated that lower rupture intensity and higher rupture resolution were associated with better ratings of the alliance and session quality. Lower rupture intensity also predicted good outcome on measures of interpersonal functioning, while higher rupture resolution predicted better retention. Patients reported fewer ruptures than did therapists. In addition, fewer ruptures were reported in cognitive–behavioral therapy than in the other treatments. Depth‐sensing or instrumented indentation is an experimental characterization approach well‐suited for high‐throughput investigation of mechanical properties of polymeric materials. This is due to both the precision of force and displacement, and to the small material volumes required for quantitative analysis. Recently, considerable progress in the throughput (number of distinct material samples analyzed per unit time) of indentation experiments has been achieved, particularly for studies of elastic properties. Future challenges include improving the agreement between various macroscopic properties (elastic modulus, creep compliance, loss tangent, onset of nonlinear elasticity, energy dissipation, etc.) and their counterpart properties obtained by indentation. Sample preparation constitutes a major factor for both the accuracy of the results and the speed and efficiency of experimental throughput. It is important to appreciate how this processing step may influence the mechanical properties, in particular the onset of nonlinear elastic or plastic deformation, and how the processing may affect the agreement between the indentation results and their macroscopic analogues.  相似文献

Among diversified industrial uses, see for instance Koch [Koch, Appl. Clay Sci., (21) 2002], and following positive in situ experiments, compacted bentonite blocks are potential candidates for sealing nuclear waste repositories, thanks to their swelling ability in a wet environment.As requested by Andra (French Agency for Nuclear Waste Management) and complementarily to in situ experiments, an original experimental laboratory set-up was designed in order to reproduce the introduction and swelling of bentonite plugs inside an argillite host rock. Once the argillite/bentonite interface is established, an increase in storage tunnel gas pressure is simulated and the interface gas migration pressure (or gas critical pressure) is evaluated. More precisely, a first experimental set-up provides bentonite swelling pressure and kinetics (i.e. mainly hydraulic cut-off, time to reach asymptotic swelling pressure and value of asymptotic swelling pressure) at given initial compaction and saturation rate. This phase is preparatory to reproducing the introduction and subsequent swelling of a bentonite plug inside the argillite host rock, which uses a similar test rig. Experimental results of water permeability and gas critical pressure are provided for MX80 compacted bentonite associated to Bure Callovo-Oxfordian argillite.  相似文献

The total energies of intermetallic compounds in the Sn–Ti system are calculated employing electronic density-functional theory (DFT) using pseudopotentials constructed by the projector augmented waves (PAW) method in the generalized gradient (GGA) approximation for the exchange and correlation energy. The calculations are performed for the experimentally observed compounds at their ideal stoichiometry as well as for structures which are stable in systems of early transition metals or rare earth elements with p-elements of columns IIIB, IVB, and VB. The calculated formation enthalpy of the hexagonal Sn5Ti6 compound is slightly less exothermic than the value obtained by direct reaction calorimetry. For the stable intermetallic compounds, the calculated zero-temperature lattice parameters agree well with those obtained experimentally at ambient temperature. More, for stable phases with unit cell-internal degree(s) of freedom, the results of ab initio calculations show good agreement when compared with data obtained by structural analysis of X-ray diffraction. The composition dependence of the enthalpies of formation is slightly asymmetric. The electronic densities of state of the D88- Sn3Ti5 compound have been computed; the curve shows the hybridization of Sn 5p states with Ti 3d states. The stability of the intermetallic compounds in the Ti–Sn system is due to this hybridization.  相似文献

Ohio River water was treated by settling, sand filtration, and granular activated carbon filtration. It was then irradiated by low-pressure (monochromatic) and medium-pressure (polychromatic) UV lamps to investigate the effects of UV irradiation on the extracted organic matter (EOM). When the EOM, collected by solid phase extraction cartridges, was analyzed by conventional UV spectroscopy and size exclusion chromatography (SEC), no significant changes in the EOM were revealed for various UV doses. Positive and negative electrospray ionization mass spectrometry (ESI-MS) of the EOM produced mass spectra that vary significantly with UV dose. The UV dosage conditions also appear to affect the reactivity of the EOM to subsequent chlorination. The magnitude of the spectral changes is generally greater for medium-pressure lamps than for low pressure and increases with UV exposure. Based on the observed MS peaks, the changes may be due to the presence of lignin, resulting perhaps from photooxidation and/or photo rearrangement of macromolecules in the sample. When chlorination is used for secondary disinfection, these results suggest that it may be important to consider the effects of UV irradiation on the organic matter in the water before applying UV disinfection technology to a particular source water.  相似文献

Leaching of particle-bound metals affects the ability of settling ponds and other engineered structures to remove metallic pollutants, and leaching behavior is related to particle size. In this investigation, water borne soil particles were leached and fractionated with a split-flow thin cell, and the metal loadings were quantified as a function of particle size. For comparison of the metal-loading curves, different empirical modeling procedures were investigated to convert the data to a precise functional form suitable for quantitative comparison of changes in differential loading as a function of particle size. Results of this investigation are presented for a soil sample before and after leaching caused by simulated acid rain conditions. Following simulated acid rain leaching, the shape of the differential distribution curves change, and these changes reflect the particle size mediated leaching behavior. For the soil used in this demonstration, simulated acid rain leaching shifted the differential loading toward larger particle sizes, and the magnitude of the shift varied significantly among the metals. Because settling rate decreases as the square of particle size, this could potentially affect management decisions for settling ponds receiving these particles. The high precision afforded by the analysis allows the development of insight into the leaching mechanisms through comparing "partial" acid rain leaching with "total recoverable" leaching by EPA Method 3050.  相似文献
