Actinobacillus actinomycetemcomitans is considered to be an aetiological agent in various forms of periodontitis, with serotype b-specific carbohydrate being the immunodominant antigen of A. actinomycetemcomitans Y4 in high-responder patients. Lipopolysaccharide (LPS) of the organism may also be an important antigen. The purpose of the present study was to clarify the importance of LPS as an antigen of A. actinomycetemcomitans. Twenty patients who had high antibody titres to strain Y4 were selected, and the reactivity of their sera with LPS was determined by ELISA and Western blotting. Two groups of patients were observed: group 1 had high IgG titres only to serotype b strain, whereas group 2 had high IgG titres to serotypes a, b and c strains. The results of adsorption tests showed that anti-A. actinomycetemcomitans Y4 antibody in group 1 patients mostly consisted of antibody reactive with the serotype b-specific carbohydrate, whereas the antibody in group 2 patients mostly consisted of antibody reactive with the LPS of all serotypes. These data show that anti-LPS antibody is present and predominant in anti-A. actinomycetemcomitans Y4 antibody from some high-responder patients, and indicate an important role for LPS as an antigen in the humoral immune response to the organism.  相似文献

A series of amidine substituted phenyl-, benzyl-, and phenethylimidazoles based on the known H3 agonist SK&F 91606 (4) has been synthesized and tested as ligands for the histamine H3 receptor. Insertion of a phenyl ring between the imidazole ring and the amidine moiety produces antagonists. The benzyl series was found to be the most potent and was further investigated. Compounds 9c and 18 (entries 5 and 12, Table 1) are potent ligands for the H3 receptor with K(i) values of 16 nM and 7.2 nM respectively. In vivo, both compounds were shown to be equipotent to thioperamide (2), the standard H3 antagonist.  相似文献

Blood osmolality was altered in guinea pigs by intravenous administration of glycerin (glycerol). After glycerin administration, serum osmolality increased rapidly, reaching a plateau within 15 minutes. Perilymph osmolality lagged significantly behind the increase of serum osmolality and equalled serum osmolality only after one hour. While perilymph osmolality responded to changes of serum osmolality, there was some time lag noted in the response of perilymph, suggesting the existence of a blood-labyrinth barrier. It was observed that this barrier is permeably to glycerin and water, suggesting the possibility of transient reduction of hydrostatic pressure in the labyrinth by the injection of an osmotic agent into the systemic circulation.  相似文献

The influence of retained austenite on the rotary bending fatigue endurance of carbonitrided specimens of two different steels is investigated. Results obtained in this work support the existence of a value of retained austenite for which the fatigue limit is maximum. A model for the influence of retained austenite, based on that generally accepted for superficial residual compressive stresses, is presented. According to this model, fatigue cracks could nucleate at different sites, surface or case bottom, in specimens with austenite contents at each side of the fatigue maximum, and this maximum should occur at lower values of retained austenite in notched specimens.  相似文献

A total of 231 pigs were anesthetized and then scanned by DXA using a Lunar DPXL instrument. The weight of the pigs ranged from 5 to 97 kg (av. = 37.7 kg). Of the total, 98 pigs were scanned using the pediatric mode and 133 pigs were scanned using the adult mode. After scanning, the pigs were euthanized, the entire body homogenized by grinding, and analyzed chemically for fat, water, protein and total body mineral content. The mean value for the DXA total tissue mass (37.1 kg) was not significantly (P > 0.05, n = 231) different from the mean body weight. The mean DXA value for percent fat was 14.6 and was significantly less (P < 0.05) than the mean value (17.6%) measured by CHEM analysis. The greatest discrepancy occurred in pigs with < 15% body fat. From a prediction equation using the DXA R value [% fat = 493-349(DXA R value)], the DXA estimated percent fat was 18.0%, compared to 17.6% by CHEM analysis (P > 0.05). Using a prediction equation [g protein = -1.062 + 0.22(g DXA lean)], the DXA estimate for body protein content was 17.8%, compared to 17.1% (P < 0.05, n = 131) by CHEM analysis. The DXA estimate for body water content [g water = 508 + 0.74 (g DXA lean)] was 63.8%, compared to 62.6% (P < 0.05, n = 231) by CHEM analysis. The bone mineral content of 83 of the pigs measured by DXA was 2.40%, compared to 2.54% (P < 0.05, n = 83) estimated from CHEM analysis of total body ash [g bone mineral = g total body ash -0.0085(g DXA lean)].  相似文献

We sought to establish predictive response models of milk fatty acid (FA) yields or concentrations from their respective duodenal flow, rumen digestive parameters, or diet characteristics in dairy cows, with a special focus on cis and trans isomers of C18:1, C18:2, odd- and branched FA, and mammary de novo synthesized FA. This meta-analysis was carried out using data from trials with nature of forage, percentage of concentrate, supplementation of diets with vegetable oils or seeds, and marine products' animal fats as experimental factors. The data set included 34 published papers representing 50 experiments with 142 treatments. Increasing duodenal C18 FA flow induced a quadratic increase in milk total C18 yield and a linear decrease in milk C4:0 to C14:0 concentration. Intra-experimental predictive response models of individual milk cis C18:1 isomers (Δ 11 to 15 position) from their respective duodenal flows had coefficients of determination (R2) ranging from 0.74 to 0.99, with root mean square error varying from 0.19 to 0.96 g/d, 0.02 to 0.10% of total FA, and 0.03 to 0.29% of C18 FA. Models predicting milk trans C18:1 isomer yields or concentrations had R2 greater than 0.90 (except for trans-4 and trans-10 C18:1) with root mean square error varying from less than 0.1 to 5.2 g/d. Linear regressions for C18:2n-6, trans-10,cis-12 CLA, and trans-11,trans-13 CLA were calculated according to their respective duodenal flows. Quadratic models of milk C18:3n-3 yield or concentration from its duodenal flow had R2 values above 0.97. Models of amounts desaturated from C18:0 into cis-9 C18:1 and trans-11 C18:1 into cis-9,trans-11 CLA indicated that the contribution of C18:0 and trans-11 C18:1 desaturation to respective cis-9 C18:1 and cis-9,trans-11 CLA yields in milk fat was 83.8% (±0.75) and 86.8% (±2.8). Furthermore, when cows were fed marine products, our results could indicate a lower mammary uptake of C18:0 and trans-11 C18:1 in proportion to their respective duodenal flow, with no associated change in mammary Δ9-desaturase activity. Yields or concentrations of C15:0, C17:0, iso-C15:0, iso-C17:0, anteiso-C15:0, and anteiso-C17:0 were dependent on their respective duodenal flow or concentration at duodenum, but synthesis of these FA from C3 units for linear-chain odd FA, and from C2 units for branched-chain FA was suggested, respectively. Several milk C18 FA concentrations were closely related to their duodenal concentrations with slopes of the linear models close to the bisector; this could reflect a priority for the use of these duodenal C18 FA by the mammary gland to favor their high concentration in plasma triglycerides and nonesterified FA, which are preferentially taken up by the mammary gland.  相似文献
