BACKGROUND: The occasional presence of H-deficient red cells among both referred and donor blood samples prompted the mass screening of donated blood in Hong Kong for H-deficient phenotypes; 96 percent of the donors tested are Chinese from the southern province of Kwongtung. STUDY DESIGN AND METHODS: Donor blood was screened for H-deficient red cells with the use of Ulex europaeus. Lewis phenotyping was carried out on all H-deficient individuals, and saliva testing was performed on most such individuals. The thermal amplitude and potency of their anti-H and anti-HI in the serum were also estimated. RESULTS: Between 1984 and 1993, 28 H-deficient blood donors were identified; 16 H-deficient patient samples were also identified, and family studies revealed an additional 7 H-deficient subjects. The H-deficient red cells did not react with anti-H lectin, the levels of ABH substances in saliva were normal or near-normal, normal levels of A or B transferase were found in plasma, minute quantities of A or B (in persons who were genetically group A or B) were detected on the red cells, and anti-H or anti-HI was detected in the serum (about 66.7% of which reacted at 37 degrees C). Atypical anti-A or anti-B was demonstrated in 81.8 percent of the cases. CONCLUSION: The H-deficient phenotype among the Hong Kong Chinese seems to represent a homogeneous group. Despite the presence of normal quantities of ABH substance in the saliva, anti-H or anti-HI that was active at 37 degrees C was detected in most cases. PURPOSE: It remains a challenge to predict which women with axillary node-negative (ANN) breast cancer at greatest risk of relapse may benefit most from adjuvant therapy. Increases in neu/erbB-2 have been implicated in breast cancer prognosis. Although overexpression has been investigated extensively, this study represents the first prospective assessment of the prognostic value of neu/erbB-2 DNA amplification in a cohort of women with newly diagnosed ANN. METHODS: A consecutive series of women was monitored for recurrence (median follow-up duration, 36 months) and tumors from 580 individuals were analyzed for amplification. The association of amplification with risk of recurrence was examined in survival analyses with traditional and histologic markers as prognostic factors. RESULTS: Neu/erbB-2 was amplified in 20% of cases. We found an increased risk of disease recurrence when neu/erbB-2 was amplified > or = twofold that persisted with adjustment for other prognostic factors (relative risk, 2.36; P = .002). We found some evidence that amplification was more important in patients who received chemotherapy compared with untreated patients. CONCLUSION: neu/erbB-2 amplification is an independent prognostic factor for risk of recurrence in ANN breast cancer. Women with tumors without neu/erbB-2 amplification have a good prognosis; aggressive therapy in this group is therefore difficult to justify. On the other hand, even with adjuvant chemotherapeutic treatment, women whose tumors exhibit neu/erbB-2 amplification have an increased risk of recurrence. We encourage a randomized trial to compare more aggressive adjuvant chemotherapy versus standard chemotherapy for ANN women whose tumors exhibit neu/erbB-2 amplification.  相似文献

The leukocyte integrin LFA-1 plays an important role in leukocyte trafficking and the immune response. Using LFA-1-deficient mice, we demonstrate that LFA-1 regulates the trafficking of lymphocytes to peripheral lymph nodes, and, to a lesser degree, to mesenteric lymph nodes and acute inflammatory sites. LFA-1, either because of its role in initial adhesion and/ or the passage of leukocytes across endothelial cells, plays a vital role in T lymphocyte and neutrophil transendothelial migration. Neutrophils and activated T lymphocytes from LFA-1-deficient mice were unable to cross endothelial cell monolayers in response to a chemokine gradient, whereas wild-type (WT) T lymphocytes and neutrophils were capable of migration. By contrast, LFA-1-deficient T lymphocytes displayed normal chemotaxis to the same chemokine. Our studies with LFA-1-deficient monocytes indicate that LFA-1 acts in concert with complement receptor 3 to mediate transendothelial migration of these cells, as anti-CD18 monoclonal antibodies (mAb) blocked both WT and LFA-1-deficient monocyte transendothelial migration, whereas anti-CD11 b mAb preferentially blocked transendothelial migration of LFA-1-deficient monocytes. Finally, whereas anti-CD31 mAb blocked WT monocyte and neutrophil transendothelial cell migration they did not block LFA-1-deficient monocyte and neutrophil transendothelial migration.  相似文献

An oversized-cavity millimetre-wave IMPATT oscillator has been constructed which can be easily coupled to a groove guide via a simple transformer. A resonant-cap structure is used to determine the frequency of oscillation. The oversized-cavity results in the Q of the oscillator being greater than that of a conventional impatt oscillator, but the output power and frequency are nevertheless stable. As silicon manufacturing processes scale to and beyond the 65-nm node, process variations are consuming an increasingly larger portion of design and test budgets. Such variations play a significant part in subthreshold leakage and other important device performance metrics. The rise in inherent systematic and random nonuniformity as we scale our silicon devices to the level of atomic scaling will have far-reaching effects on every aspect of design, manufacturing, test, and overall reliability. Previous studies regarding interunit dispersion used Reynolds‐averaged Navier–Stokes (RANS) models and thus obtained only mean dispersion routes and re‐entry ratios. Given that the envelope flow around a building is highly fluctuating, mean values could be insufficient to describe interunit dispersion. This study investigates the wind‐induced interunit dispersion around multistory buildings using the large eddy simulation (LES) method. This is the first time interunit dispersion has been investigated transiently using a LES model. The quality of the selected LES model is seriously assured through both experimental validation and sensitivity analyses. Two aspects are paid special attention: (i) comparison of dispersion routes with those provided by previous RANS simulations and (ii) comparison of timescales with those of natural ventilation and the survival times of pathogens. The LES results reveal larger dispersion scopes than the RANS results. Such larger scopes could be caused by the fluctuating and stochastic nature of envelope flows, which, however, is canceled out by the inherent Reynolds‐averaged treatment of RANS models. The timescales of interunit dispersion are comparable with those of natural ventilation. They are much shorter than the survival time of most pathogens under ordinary physical environments, indicating that interunit dispersion is a valid route for disease transmission.  相似文献

Carbon nanotubes were easily coated with a reactive biopolymer - polydopamine (Pdop) - by dispersing them in a dopamine solution and mildly stirring at room temperature. The coating did not ruin the nanotube sidewalls, and its thickness was finely controlled on the nanometer scale by adjusting the reaction time, pH and temperature. The resulted coating is a versatile platform for both functional organic ad-layers and electroless metallization. A high concentration of gold nanoparticles was deposited on the Pdop layer, by simply stirring within HAuCl4 solution at room temperature.  相似文献

Effective solutions to the cell formation and the production scheduling problems are vital in the design of virtual cellular manufacturing systems (VCMSs). This paper presents a new mathematical model and a scheduling algorithm based on the techniques of genetic algorithms for solving such problems. The objectives are: (1) to minimize the total materials and components travelling distance incurred in manufacturing the products, and (2) to minimize the sum of the tardiness of all products. The proposed algorithm differs from the canonical genetic algorithms in that the populations of candidate solutions consist of individuals of different age groups, and that each individual's birth and survival rates are governed by predefined aging patterns. The condition governing the birth and survival rates is developed to ensure a stable search process. In addition, Markov Chain analysis is used to investigate the convergence properties of the genetic search process theoretically. The results obtained indicate that if the individual representing the best candidate solution obtained is maintained throughout the search process, the genetic search process converges to the global optimal solution exponentially.

The proposed methodology is applied to design the manufacturing system of a company in China producing component parts for internal combustion engines. The performance of the proposed age-based genetic algorithm is compared with that of the conventional genetic algorithm based on this industrial case. The results show that the methodology proposed in this paper provides a simple, effective and efficient method for solving the manufacturing cell formation and production scheduling problems for VCMSs.  相似文献

Trucks are the most popular transport equipment in most mega-terminals, and scheduling them to minimize makespan is a challenge that this article addresses and attempts to resolve. Specifically, the problem of scheduling a fleet of trucks to perform a set of transportation jobs with sequence-dependent processing times and different ready times is investigated, and the use of a genetic algorithm (GA) to address the scheduling problem is proposed. The scheduling problem is formulated as a mixed integer program. It is noted that the scheduling problem is NP-hard and the computational effort required to solve even small-scale test problems is prohibitively large. A crossover scheme has been developed for the proposed GA. Computational experiments are carried out to compare the performance of the proposed GA with that of GAs using six popular crossover schemes. Computational results show that the proposed GA performs best, with its solutions on average 4.05% better than the best solutions found by the other six GAs.  相似文献
