Cells have the ability to sense different environmental signals and position themselves accordingly in order to support their survival. Introducing analogous capabilities to the bottom‐up assembled minimal synthetic cells is an important step for their autonomy. Here, a minimal synthetic cell which combines a multistimuli sensitive adhesion unit with an energy conversion module is reported, such that it can adhere to places that have the right environmental parameters for ATP production. The multistimuli sensitive adhesion unit senses light, pH, oxidative stress, and the presence of metal ions and can regulate the adhesion of synthetic cells to substrates in response to these stimuli following a chemically coded logic. The adhesion unit is composed of the light and redox responsive protein interaction of iLID and Nano and the pH sensitive and metal ion mediated binding of protein His‐tags to Ni2+‐NTA complexes. Integration of the adhesion unit with a light to ATP conversion module into one synthetic cell allows it to adhere to places under blue light illumination, non‐oxidative conditions, at neutral pH and in the presence of metal ions, which are the right conditions to synthesize ATP. Thus, the multistimuli responsive adhesion unit allows synthetic cells to self‐position and execute their functions.  相似文献

Light-driven ATP regeneration systems combining ATP synthase and bacteriorhodopsin have been proposed as an energy supply in the field of synthetic biology. Energy is required to power biochemical reactions within artificially created reaction compartments like protocells, which are typically based on either lipid or polymer membranes. The insertion of membrane proteins into different hybrid membranes is delicate, and studies comparing these systems with liposomes are needed. Here we present a detailed study of membrane protein functionality in different hybrid compartments made of graft polymer PDMS-g-PEO and diblock copolymer PBd-PEO. Activity of more than 90 % in lipid/polymer-based hybrid vesicles could prove an excellent biocompatibility. A significant enhancement of long-term stability (80 % remaining activity after 42 days) could be demonstrated in polymer/polymer-based hybrids.  相似文献

The natural sweeteners erythritol and xylitol might be helpful to reduce sugar consumption and therefore prevent obesity and diabetes. The aim of the present study was to determine the absorption and metabolization into erythronate of different concentrations of erythritol and xylitol. Seventeen healthy lean participants received intragastric solutions of 10, 25, or 50 g erythritol or 7, 17, or 35 g xylitol on three study days in a randomized order. The study was double blinded with respect to the doses administered. We assessed plasma concentrations of erythritol, xylitol, and erythronate at fixed time intervals after administration with gas chromatography-mass spectrometry. We found: (i) a dose-dependent and saturable absorption of erythritol, (ii) a very low absorption of xylitol, (iii) a dose-dependent metabolization of erythritol into erythronate, and (iv) no metabolization of xylitol into erythronate. The implications of the metabolization of erythritol into erythronate for human health remain to be determined and more research in this area is needed.  相似文献

In 3 experiments, the authors examined factors that, according to the source-monitoring framework, might influence false memory formation and true/false memory discernment. In Experiment 1, combined effects of warning and visualization on false childhood memory formation were examined, as were individual differences in true and false childhood memories. Combining warnings and visualization led to the lowest false memory and highest true memory. Several individual difference factors (e.g., parental fearful attachment style) predicted false recall. In addition, true and false childhood memories differed (e.g., in amount of information). Experiment 2 examined relations between Deese/Roediger-McDermott task performance and false childhood memories. Deese/Roediger-McDermott performance (e.g., intrusion of unrelated words in free recall) was associated with false childhood memory, suggesting liberal response criteria in source decisions as a common underlying mechanism. Experiment 3 investigated adults' abilities to discern true and false childhood memory reports (e.g., by detecting differences in amount of information as identified in Experiment 1). Adults who were particularly successful in discerning such reports indicated reliance on event plausibility. Overall, the source-monitoring framework provided a viable explanatory framework. Analysis and design of interconnects in high speed integrated circuits and systems involves models in the form of multiconductor transmission lines. The fundamental parameters of those models are matrices of capacitance, (C), inductance, (L), resistance, (R), and conductance (G). We present a methodology for measurement of entries in capacitance matrix. The entries of capacitance matrices can be calculated using numerical solvers of electrostatic fields established under the assumption of suitable biasing of interconnect structures. Numerical calculations of complete field equations are very complex and expensive in terms of computer time, therefore several approximations are made in constructing interconnect dedicated software packages available on the market. Because of these approximations it is necessary to validate the calculations via measurements. Calculation of the off-diagonal entries of capacitance matrix from measurements of "two-terminal" capacitances is strongly corrupted by the measuring errors. The method involves direct capacitance measurement in multi-conductor structures and provides analysis of accuracy.  相似文献

This paper presents a simple, physics-based, and continuous model for the quantum effects and polydepletion in deep-submicrometer MOSFETs with very thin gate oxide thicknesses. This analytical design-oriented MOSFET model correctly predicts inversion and depletion charges, transcapacitances, and drain current, from weak to strong inversion and from nonsaturation to saturation. One single additional parameter is used for polysilicon doping concentration, while the quantum correction does not introduce any new parameter. Comparison to experimental data of deep-submicrometer technologies is provided, showing accurate fits both for I-V and C-V data. The model offers simple relationships among effective electrical parameters and physical device parameters, providing insight into the physical phenomena. This new model thereby supports device engineering, analog circuit design practice, as well as efficient circuit simulation.  相似文献

We report the synthesis of alumina/stishovite nano‐nano composite ceramics through a pressure‐induced dissociation in Al2SiO5 at a pressure of 15.6 GPa and temperatures of 1300°C‐1900°C. Stishovite is a high‐pressure polymorph of silica and the hardest known oxide at ambient conditions. The grain size of the composites increases with synthesis temperature from ~15 to ~750 nm. The composite is harder than alumina and the hardness increases with reducing grain size down to ~80 nm following a Hall–Petch relation. The maximum hardness with grain size of 81 nm is 23 ± 1 GPa. A softening with reducing grain size was observed below this grain size down to ~15 nm, which is known as inverse Hall–Petch behavior. The grain size dependence of the hardness might be explained by a composite model with a softer grain‐boundary phase.  相似文献

Correlation optimized warping (COW) based on the total ion current (TIC) is a widely used time alignment algorithm (COW-TIC). This approach works successfully on chromatograms containing few compounds and having a well-defined TIC. In this paper, we have combined COW with a component detection algorithm (CODA) to align LC-MS chromatograms containing thousands of biological compounds with overlapping chromatographic peaks, a situation where COW-TIC often fails. CODA is a variable selection procedure that selects mass chromatograms with low noise and low background (so-called "high-quality" mass chromatograms). High-quality mass chromatograms selected in each COW segment ensure that the same compounds (based on their mass and their retention time) are used in the two-dimensional benefit function of COW to obtain correct and optimal alignments (COW-CODA). The performance of the COW-CODA algorithm was evaluated on three types of complex data sets obtained from the LC-MS analysis of samples commonly used for biomarker discovery and compared to COW-TIC using a new global comparison method based on overlapping peak area: trypsin-digested serum obtained from cervical cancer patients, trypsin-digested serum from a single patient that was treated with varying preanalytical parameters (factorial design study), and urine from pregnant and nonpregnant women. While COW-CODA did result in minor misalignments in rare cases, it was clearly superior to the COW-TIC algorithm, especially when applied to highly variable chromatograms (factorial design, urine). The presented algorithm thus enables automatic time alignment and accurate peak matching of multiple LC-MS data sets obtained from complex body fluids that are often used for biomarker discovery.  相似文献
