This work evaluates the hydrolysis of sugarcane bagasse using supercritical carbon dioxide to obtain fermentable sugars in a single‐step process. The effects of temperature, pressure, despressurization rate, moisture content and reaction time on the amount of fermentable sugars released were assessed. Results obtained showed the feasibility of adopting the present procedure since it produced about 287 g of fermentable sugars per kg of dry bagasse, which corresponds to 60% yield on the basis of total cellulose. Such a result is very attractive from an industrial viewpoint because there is no use of toxic solvent, solvent removal is completed (no residues), and the method has very low water consumption since the process is a solid–gas reaction. © 2012 Society of Chemical Industry  相似文献

The ρ‐containing γ‐aminobutyric acid type A receptors (GABAARs) play an important role in controlling visual signaling. Therefore, ligands that selectively target these GABAARs are of interest. In this study, we demonstrate that the partial GABAAR agonist imidazole‐4‐acetic acid (IAA) is able to penetrate the blood–brain barrier in vivo; we prepared a series of α‐ and N‐alkylated, as well as bicyclic analogues of IAA to explore the structure–activity relationship of this scaffold focusing on the acetic acid side chain of IAA. The compounds were prepared via IAA from l ‐histidine by an efficient minimal‐step synthesis, and their pharmacological properties were characterized at native rat GABAARs in a [3H]muscimol binding assay and at recombinant human α1β2γ2S and ρ1 GABAARs using the FLIPR? membrane potential assay. The (+)‐α‐methyl‐ and α‐cyclopropyl‐substituted IAA analogues ((+)‐ 6 a and 6 c , respectively) were identified as fairly potent antagonists of the ρ1 GABAAR that also displayed significant selectivity for this receptor over the α1β2γ2S GABAAR. Both 6 a and 6 c were shown to inhibit GABA‐induced relaxation of retinal arterioles from porcine eyes.  相似文献

Resveratrol is a polyphenol that is associated with numerous health benefits related to heart disease, cancer, diabetes, and neurological function. The addition of this compound to food products would help to deliver these health benefits to the consumer. However, bitterness associated with resveratrol may impart negative sensory qualities on the food products into which resveratrol is added; thus, decreasing consumer acceptability. This concern may be resolved by encapsulating resveratrol through spray drying, an innovative processing technique. The objectives of this research were to (1) compare taste detection thresholds of unencapsulated resveratrol and encapsulated resveratrol and (2) determine if the inclusion of anhydrous milk fat in the formulation of the encapsulation wall material affects the taste detection threshold of resveratrol within the microcapsules. Resveratrol microcapsules were produced by encapsulating resveratrol in a protein matrix through spray drying. R‐index measure by the rating method was used to determine the average taste detection threshold and the pooled group taste detection threshold. The average and pooled group taste detection thresholds of unencapsulated resveratrol, sodium‐caseinate‐based resveratrol microcapsule without fat (SC), and sodium‐caseinate‐based resveratrol microcapsule with fat (SCAMF) were 90 and 47 mg resveratrol/L (unencapsulated), 313 and 103 mg resveratrol/L (SC), 334 and 108 mg resveratrol/L (SCAMF), respectively. The findings demonstrate that the encapsulation of resveratrol decreased the detection of the compound and provided a means to incorporate resveratrol into food products without imparting negative sensory properties.  相似文献

In the context of optimization by evolutionary algorithms (EAs), epistasis, deception, and scaling are well-known examples of problem difficulty characteristics. The presence of one such characteristic in the representation of a search problem indicates a certain type of difficulty the EA is to encounter during its search for globally optimal configurations. In this paper, we claim that the occurrence of symmetry in the representation is another problem difficulty characteristic and discuss one particular form, spin-flip symmetry, characterized by fitness invariant permutations on the alphabet. Its usual effect on unspecialized EAs, premature convergence due to synchronization problems, is discussed in detail. We discuss five different ways to specialize EAs to cope with the symmetry: adapting the genetic operators, changing the fitness function, using a niching technique, using a distributed EA, and attaching a highly redundant genotype-phenotype mapping.  相似文献

Until recently, genes from the iron-sulfur (Fe-S) cluster pathway were not known to have a role in plant disease resistance. The Nitrogen Fixation S (NIFS)-like 1 (NFS1) and Mitochondrial Ferredoxin-1 (MFDX1) genes are part of a set of 27 Fe-S cluster genes induced after infection with host and nonhost pathogens in Arabidopsis. A role for AtNFS1 in plant immunity was recently demonstrated. In this work, we showed that MFDX1 is also involved in plant defense. More specifically, Arabidopsis mfdx1 mutants were compromised for nonhost resistance against Pseudomonas syringae pv. tabaci, and showed increased susceptibility to the host pathogen P. syringae pv. tomato DC3000. Arabidopsis AtMFDX1 overexpression lines were less susceptible to P. syringae pv. tomato DC3000. Metabolic profiling revealed a reduction of several defense-related primary and secondary metabolites, such as asparagine and glucosinolates in the Arabidopsis mfdx1-1 mutant when compared to Col-0. A reduction of 5-oxoproline and ornithine metabolites that are involved in proline synthesis in mitochondria and affect abiotic stresses was also observed in the mfdx1-1 mutant. In contrast, an accumulation of defense-related metabolites such as glucosinolates was observed in the Arabidopsis NFS1 overexpressor when compared to wild-type Col-0. Additionally, mfdx1-1 plants displayed shorter primary root length and reduced number of lateral roots compared to the Col-0. Taken together, these results provide additional evidence for a new role of Fe-S cluster pathway in plant defense responses.  相似文献

This study deals with the development of a method for total Hg, inorganic Hg and methylmercury (MeHg) determination in red wine by using flow injection-cold vapour generation–inductively coupled plasma mass spectrometry (FI-CVG-ICP-MS) and gas chromatography-ICP-MS (GC-ICP-MS). For Hg speciation analysis, a derivatization step was carried out using a 1% (m/v) sodium tetraphenylborate (NaBPh4) solution, followed by extraction of Hg species and their quantification by GC-ICP-MS. The main parameters evaluated were the make-up gas flow rate, volume of the NaBPh4 solution, time for derivatization reaction/analyte extraction and solvent used for Hg species extraction. Accuracy was evaluated by analyte recovery, whereas recoveries ranged from 99% to 104% for Hg(II) and MeHg. The limits of detection (LODs) for Hg(II) and MeHg were 0.77 and 0.80 μg L−1, respectively. Wine from Argentina, Brazil, Chile and Uruguay were analysed. The wine samples were also acid digested for total Hg determination by FI-CVG-ICP-MS. The LOD of the method used for total Hg determination was 0.01 μg L−1. The concentrations of Hg species in red wine measured by GC-ICP-MS were lower than the respective LODs. Only total Hg was detected in the analysed samples, where the highest concentration of Hg found was 0.55 ± 0.02 μg L−1.  相似文献
