Sesquiterpene lactones are of pharmaceutical interest due their cytotoxic and antitumor properties, which are commonly found within plants of several genera from the Asteraceae family such as the Decachaeta genus. From Decachaeta incompta four heliangolide, namely incomptines A-D have been isolated. In this study, cytotoxic properties of incomptine A (IA) were evaluated on four lymphoma cancer cell lines: U-937, Farage, SU-DHL-2, and REC-1. The type of cell death induced by IA and its effects on U-937 cells were analyzed based on its capability to induce apoptosis and produce reactive oxygen species (ROS) through flow cytometry with 4′,6-diamidino-2-phenylindole staining, dual annexin V/DAPI staining, and dichlorofluorescein 2′,7′-diacetate, respectively. A differential protein expression analysis study was carried out by isobaric tags for relative and absolute quantitation (iTRAQ) through UPLC-MS/MS. Results reveal that IA exhibited cytotoxic activity against the cell line U-937 (CC50 of 0.12 ± 0.02 μM) and the incubation of these cells in presence of IA significantly increased apoptotic population and intracellular ROS levels. In the proteomic approach 1548 proteins were differentially expressed, out of which 587 exhibited a fold-change ≥ 1.5 and 961 a fold-change ≤ 0.67. Most of these differentially regulated proteins are involved in apoptosis, oxidative stress, glycolytic metabolism, or cytoskeleton structuration.  相似文献

The catalytic oxidative coupling of methane (OCM) to C2 hydrocarbons (C2H6 and C2H4) represents one of the most effective ways to convert natural gas to more useful products,which can be performed effec-tively using La0.6Sr0.4Co0.2Fe0.8O3-δ (LSCF) perovskite hollow fiber membrane microreactor.In this work,the effects of adding a thin BaCe0.8Gd0.2O3-δ (BCG) catalyst film onto the inner LSCF fiber surface as the OCM catalyst and a porous Ba0.5Sr0.5Co0.8Fe0.2O3-δ (BSCF) perovskite layer onto the outer LSCF surface to improve the oxygen permeation were evaluated.Between 700 ℃ and 1000 ℃,methane conversion increased in the order of uncoated,BCG and BSCF-coated,and BCG-coated LSCF hollow fiber while C2-selectivity and C2-yield increased in the order of BCG and BSCF-coated,uncoated,and BCG-coated LSCF hollow fiber.Oxygen permeation flux at the same temperature range,on the other hand,was enhanced in the order of uncoated,BCG-coated,and BCG and BSCF-coated LSCF hollow fiber,This finding demon-strates the complex interplay between oxygen permeation and OCM performance.The BCG and BSCF-coated hollow fiber was also subjected to thermal cycles between 850 ℃ and 900 ℃ for up to 175 hours during which the fiber showed minor degradation in oxygen permeation fluxes and relatively stable OCM performance.  相似文献

Understanding the effects of N application or the introduction of a legume on N cycling is critical for achieving productive and sustainable grassland systems. This 2-year study assessed the N cycling of three pasture treatments: (1) mixed Marandu palisadegrass (Brachiaria brizantha) and forage peanut (Arachis pintoi) without N fertiliser (GRASS?+?LEGUME); (2) monoculture Marandu palisadegrass fertilised with 150 kg N ha?1 year?1 (GRASS?+?N); and (3) monoculture Marandu palisadegrass without N fertiliser (GRASS). Continuous stocking was used with a target canopy height of 0.20 to 0.25 m. Litter responses, forage and N intake, N livestock excretion and N cycling were measured. Existing litter and litter deposition rate were greatest in GRASS pasture (3030 and 84.3 vs. 2140 kg ha?1 and 64.8 kg OM ha?1 d?1; average of GRASS?+?N and GRASS?+?LEGUME pastures, respectively; P?<?0.10). Litter decomposition rate in GRASS pasture was smaller 30.4 and 36.0% compared to GRASS?+?N and GRASS?+?LEGUME pastures, respectively (P?<?0.10). The GRASS?+?N obtained greatest (P?<?0.10) faecal N excretion (21.7 vs. 13.8 kg N ha?1 season?1), and urinary N excretion (32.0 vs. 14.2 kg N ha?1 season?1). In the GRASS?+?N and GRASS?+?LEGUME pastures, there was a positive overall change of N in the soil–plant–animal system of 13 and 33 kg N ha?1 year?1, respectively. In the GRASS pasture, there was an overall negative change of N in the soil–plant–animal system of ??41 kg N ha?1 year?1. Nitrogen application or the integration of forage peanut in a grass pasture increased the conservation of soil N reserves.

Focused ion beam scanning electron microscopy (FIB-SEM) tomography is a serial sectioning technique where an FIB mills off slices from the material sample that is being analysed. After every slicing, an SEM image is taken showing the newly exposed layer of the sample. By combining all slices in a stack, a 3D image of the material is generated. However, specific artefacts caused by the imaging technique distort the images, hampering the morphological analysis of the structure. Typical quality problems in microscopy imaging are noise and lack of contrast or focus. Moreover, specific artefacts are caused by the FIB milling, namely, curtaining and charging artefacts. We propose quality indices for the evaluation of the quality of FIB-SEM data sets. The indices are validated on real and experimental data of different structures and materials.  相似文献

SiC-fiber-reinforced Si3N4 composites were fabricated by hot pressing. The indentation-strength technique was applied to study the mechanical properties of these composites. This enabled the investigation of short-crack behavior of continuous-fiber ceramic composites (CFCCs). The flaw tolerance of composite ultimate strength, matrix-cracking stress, and work-of-fracture were investigated. Scanning electron microscopy was used to examine crack–fiber interactions. The ultimate strength was found to be independent of indentation load at a fiber volume fraction f = 0.29, while at f = 0.14 it exhibited a transition from flawsensitive to flaw-independent. The work-of-fracture was found to be independent of indentation load at both fiber volume fractions. The matrix-cracking stress was found to correspond to the first load-drop on the load–displacement curve. It decreased with increasing flaw size and therefore is the steady-state matrix-cracking stress. A failure mechanism transition from catastrophic failure to non-catastrophic failure, coupled with the transition from flawsensitive to flaw-tolerant behavior, was observed by varying the preexisting flaw size and the fiber volume fraction. These transitions were explained by analyzing the relations between ultimate strength, matrix-cracking stress, fiber volume fraction, and preexisting flaw size of the composite materials. Experimental results were compared with predictions from available models.  相似文献

A novel device that uses friction between one or more pairs of elastic conical rings to dissipate the energy from an impacting body is presented. The device consists of one moving and one stationary cylinders coupled to each other using two pairs of conical rings and a spring. The spring is used to restore the system to its original configuration after the impact. The dynamic response of the system to the impact forces during impact events is analysed numerically and experimentally. The effects of several governing parameters, such as the mass ratio between the cylinders, the duration of the transient response of the device, the magnitude of the rest zone of the moving element and the peak impact force are investigated. The proposed system is applicable in sequential impact scenarios, in which remarkable improvements were observed over traditional solid-rod impact absorbers. The present study may serve as a guide for the design of improved damping devices for impact applications.  相似文献

The objective in this study was to evaluate the antifungal activity of essential oils from native and commercial aromatic plants grown in Argentina against corn postharvest fungi and to link the essential oil bioactivity with lipid oxidation and morphological changes in fungus cell membrane. Essential oil (EO) of oregano variety Mendocino (OMen), Cordobes (OCor), and Compacto (OCom), mint variety Inglesa (Mi), and Pehaujo (Mp), Suico (Sui); rosemary (Ro), and Aguaribay (Ag) were tested in vitro against 4 corn fungi: A. flavus (CCC116–83 and BXC01), P. oxalicum (083296), and P. minioluteum (BXC03). The minimum fungicidal concentration (MFC) and the minimum inhibitory concentration (MIC) were determined. The chemical profiles of the EOs were analyzed by GC‐MS. Lipid oxidation in cell membrane of fungi was determined by hydroperoxides and related with essential oil antifungal activity. The major compounds were Thymol in OCor (18.66%), Omen (12.18%), and OCom (9.44%); menthol in Mi and Mp; verbenone in Sui; dehydroxy‐isocalamendiol in Ag; and eucaliptol in Ro. OCor, Omen, and OCom showed the best antifungal activity. No antifungal activity was observed in Ag and Ro EO. The hydroperoxide value depended on the fungi (P < 0.001) and the antimicrobial agent (P < 0.001).Membrane lipids were oxidized by Sui EO in A. flavus BXC01 and A. flavus CCC116–83 (0.021 and 0.027 meqO2/kg, respectively). The results suggest that the EOs of OCor, OMen, OCom, Mi, Mp, and Sui grown in Argentina can be used as natural alternatives to control fungi that produce mycotoxin in maize.  相似文献

An antioxidant active packaging was prepared by coating a citrus extract, consisting of a mixture of carboxylic acids and flavanones, on polyethylene terephthalate trays. The effect of the packaging in reducing lipid oxidation in cooked turkey meat and on meat pH, colour characteristics and sensorial parameters was investigated. An untrained sensory panel evaluated the odour, taste, tenderness, juiciness and overall acceptability of the meat, using triangle, paired preference and quantitative response scale tests. A comparison between the antioxidant effects of the different components of the extract was also carried out. The packaging led to a significant reduction in lipid oxidation. After 2 days of refrigerated storage the sensory panel detected differences in odour and, after 4 days, rated the meat stored in the active packaging higher for tenderness and overall acceptability. Citric acid appeared to be the most important component of the extract with regard to its antioxidant potency.  相似文献
