Activation of P2X7 signaling, due to high glucose levels, leads to blood retinal barrier (BRB) breakdown, which is a hallmark of diabetic retinopathy (DR). Furthermore, several studies report that high glucose (HG) conditions and the related activation of the P2X7 receptor (P2X7R) lead to the over-expression of pro-inflammatory markers. In order to identify novel P2X7R antagonists, we carried out virtual screening on a focused compound dataset, including indole derivatives and natural compounds such as caffeic acid phenethyl ester derivatives, flavonoids, and diterpenoids. Molecular Mechanics/Generalized Born Surface Area (MM/GBSA) rescoring and structural fingerprint clustering of docking poses from virtual screening highlighted that the diterpenoid dihydrotanshinone (DHTS) clustered with the well-known P2X7R antagonist JNJ47965567. A human-based in vitro BRB model made of retinal pericytes, astrocytes, and endothelial cells was used to assess the potential protective effect of DHTS against HG and 2′(3′)-O-(4-Benzoylbenzoyl)adenosine-5′-triphosphate (BzATP), a P2X7R agonist, insult. We found that HG/BzATP exposure generated BRB breakdown by enhancing barrier permeability (trans-endothelial electrical resistance (TEER)) and reducing the levels of ZO-1 and VE-cadherin junction proteins as well as of the Cx-43 mRNA expression levels. Furthermore, HG levels and P2X7R agonist treatment led to increased expression of pro-inflammatory mediators (TLR-4, IL-1β, IL-6, TNF-α, and IL-8) and other molecular markers (P2X7R, VEGF-A, and ICAM-1), along with enhanced production of reactive oxygen species. Treatment with DHTS preserved the BRB integrity from HG/BzATP damage. The protective effects of DHTS were also compared to the validated P2X7R antagonist, JNJ47965567. In conclusion, we provided new findings pointing out the therapeutic potential of DHTS, which is an inhibitor of P2X7R, in terms of preventing and/or counteracting the BRB dysfunctions elicited by HG conditions.  相似文献

Accelerated storage tests are frequently used to assess the oxidative stability of foods and related systems due to its reproducibility. Various methods and experimental conditions are used to measure lipid oxidation. Differences between laboratories make it necessary to determine the repeatability and reproducibility of oxidation tests performed under the same conditions. The objective of the present interlaboratory study was to evaluate the outcome of a storage test for two different bulk oils, sunflower oil (SFO) and rapeseed oil (RSO), during a period of 9 weeks at 20°C, 30°C, 40°C, and 60°C. Sixteen laboratories were provided with bottled oils and conducted the storage tests according to a detailed protocol. Lipid oxidation was monitored by the formation of conjugated dienes (CD) and the activation energy (Ea) was determined for comparative purposes and statistically evaluated. An increase in CD formation was observed for both oils when the storage temperature was increased in all laboratories. The Ea,1 ranged from 47.9 to 73.3 kJ mol−1 in RSO and from 27.8 to 62.6 kJ mol−1 in SFO, with average values of 58.2 and 46.8 kJ mol−1, respectively. The reproducibility coefficients were 10.9% and 18.2% for RSO and SFO, respectively. Practical applications: In order to compare results on oxidative stability of foods derived from different studies, the reproducibility of storage tests and methods employed to evaluate the oxidation level should be considered. This study provides fundamental data on the reproducibility of lipid oxidation under accelerated storage conditions and defines important parameters to be considered for the conduction of experiments.  相似文献

Styrene polymerization was carried out with Ni(acac)2/MAO and Ni(acac)2/SiO2/MAO. The influence of reaction parameters (Al/Ni mole ratio, catalyst concentration, temperature and time polymerization) on styrene polymerization was evaluated. It was observed that both catalytic systems were affected by reaction parameters and that the heterogeneous catalyst presented higher activity than the homogeneous one. Polystyrenes with different molecular weight, stereoregularity and polydispersity were obtained. These results suggest that different active catalyst species could have been present. In addition, two types of methylaluminoxane (MAO) with different molecular weights were also evaluated as cocatalyst. As a result, the catalyst activity and stereospecificity were strongly affected by the MAO type.  相似文献

The production of porous polymeric particles is attractive for a large number of applications and can be achieved by various techniques. Although numerous production schemes exist for glassy polymers, difficulties arise for soft, rubbery materials that need a chemical crosslinking step, such as elastomers. This is particularly true for poly(dimethylsiloxane) (PDMS), which shows the lowest glass‐transition temperature among the polymers. Recent studies suggest in situ hydrogen bubble formation or vacuum drying of water droplets dispersed in the polymer matrix in order to generate porous PDMS structures. In this work we report early results based on the chemical crosslinking of water in PDMS emulsion droplets in a mechanically stirred thermostated water vessel. This approach is shown to lead to high porosity PDMS beads (ca. 10?3 m particle diameter) with an open structure whose properties (diameter and porosity) are strongly influenced by the starting composition (solvent, surfactant, and polymer types and ratios), as well as the operating parameters (agitation and temperature). We discuss an analytical model for the evaluation of radiation absorption in a tubular photocatalytic reactor. The model has no adjustable parameters and takes into account scattering in all directions. We compare the results of this model with those of Monte Carlo (MC) simulations and of a Lambert–Beer (LB) approximation, for a reactor illuminated by a parabolic solar concentrator. A good correspondence is found with the MC simulations. In particular, the model displays the correct saturation behavior of absorption for large catalyst particle concentrations, which is not obtained with the LB approximation. We have carried out experiments for the degradation of carbaryl in a solar parabolic collector (PC). The model is used to calculate the rate constant for this degradation from the experimental data. The theoretical model predictions reproduce well, the trends observed in the experiments.  相似文献

Summary The preparation of a chelating ion-exchange network based on acrylonitrile was carried out by chemical modification with hydroxylamine. The beads of resin were synthesized by aqueous suspension copolymerization of acrylonitrile (AN), styrene (STY) and divinylbenzene (DVB). The influence of diluent used in the suspension polymerization on the structure of the resulting copolymers was evaluated. The diluents employed were heptane (HEP), toluene (TOL) and anisole (ANI). It was found that the AN incorporation into copolymer structure was dependent on the diluent used. Conversion of nitrile groups into the amidoxime was conducted by treatment with hydroxylamine under alkaline solution. The resins were characterized by apparent density, surface area, average pore diameter, elemental analysis (CHN), FTIR and optical microscopy. Based on the results obtained, it was possible to control the porosity by diluent employed in the synthesis and to modify chemically a resin containing nitrile groups by hydroxylamine reaction. A conformational energy analysis of the isolated chain of syndiotactic poly(m-methylstyrene) under the constraint of a crystalline field is reported. Two different minimum energy conformations having similar energy have been found; the trans-planar conformation with tcm symmetry and the two-fold helical conformation with s(2/1)2 symmetry, according with the observed polymorphic behavior of this polymer. The calculated chain axes are in agreement with the experimental axes of 5.1 and 7.9 Å found for the different polymorphic forms of syndiotactic poly(m-methylstyrene). However, only a metastable disordered modification (form III) having chains in trans-planar conformation has been described. This indicates that, even though the trans-planar conformation is, in the isolated chain as stable as the helical conformation, the packing of the chains in helical conformation is probably more efficient than that of the trans-planar chains.  相似文献

The coating of soybean seeds with bacteria and micronutrients favours the vigorous growth of the plant, dispensing with the use of ammoniacal fertilizers. The optimum thickness of the coating should allow the fundamental gaseous interchanges for the germination and the ideal conditions for the activity of the bacteria. The objective of this work was to simulate dynamic mass distribution of soybean seeds covered with bacteria and macronutrients in spouted bed through a specific program developed in Maple V®. Background: Pathological complete response (pCR) to neoadjuvant chemoradiotherapy (CRT) in locally advanced rectal cancer (LARC) is still ascribed to a minority of patients. A pathway based-approach could highlight the predictive role of germline single nucleotide polymorphisms (SNPs). The primary aim of this study was to define new predictive biomarkers considering treatment specificities. Secondary aim was to determine new potential predictive biomarkers independent from radiotherapy (RT) dosage and cotreatment with oxaliplatin. Methods: Thirty germ-line SNPs in twenty-one genes were selected according to a pathway-based approach. Genetic analyses were performed on 280 LARC patients who underwent fluoropyrimidine-based CRT. The potential predictive role of these SNPs in determining pathological tumor response was tested in Group 1 (94 patients undergoing also oxaliplatin), Group 2 (73 patients treated with high RT dosage), Group 3 (113 patients treated with standard RT dosage), and in the pooled population (280 patients). Results: Nine new predictive biomarkers were identified in the three groups. The most promising one was rs3136228-MSH6 (p = 0.004) arising from Group 3. In the pooled population, rs1801133-MTHFR showed only a trend (p = 0.073). Conclusion: This exploratory study highlighted new potential predictive biomarkers of neoadjuvant CRT and underlined the importance to strictly define treatment peculiarities in pharmacogenetic analyses.  相似文献
