Detergents are an absolute requirement for studying the structure of membrane proteins. However, many conventional detergents fail to stabilise denaturation‐sensitive membrane proteins, such as eukaryotic proteins and membrane protein complexes. New amphipathic agents with enhanced efficacy in stabilising membrane proteins will be helpful in overcoming the barriers to studying membrane protein structures. We have prepared a number of deoxycholate‐based amphiphiles with carbohydrate head groups, designated deoxycholate‐based glycosides (DCGs). These DCGs are the hydrophilic variants of previously reported deoxycholate‐based N‐oxides (DCAOs). Membrane proteins in these agents, particularly the branched diglucoside‐bearing amphiphiles DCG‐1 and DCG‐2, displayed favourable behaviour compared to previously reported parent compounds (DCAOs) and conventional detergents (LDAO and DDM). Given their excellent properties, these agents should have significant potential for membrane protein studies.  相似文献

Mullite (3Al2O3·2SiO2) coatings on SiC substrates and SiC precoated carbon/carbon composite (C/C-Si-SiC) substrates were produced by pulsed laser deposition (PLD) using pressed mullite powder targets. The layers can be characterized efficiently by IR reflection spectroscopy in the spectral range between 650 and 5000 cm−1. The deposited coatings turn into mullite upon oxidation in air at temperatures between 1400° and 1600°C. Fabry-Perot interferences indicate a high quality and homogeneity of the mullite coating/SiC substrate interface. Amorphous SiO2 gradually forms during prolonged heating or at higher temperatures.  相似文献

The use of microcapsules containing healing agents, which can serve as source of self-healing for coatings, was investigated. The appropriate self-repairing coatings are designed to be used for heavy-duty corrosion protection or for the protection of moisture-sensitive substrates like wood. Microcapsules based on thermoset resins (urea–formaldehyde and melamine–formaldehyde) which contained components of different chemical natures (resins or inhibitors) were fabricated using emulsion techniques. The subsequent characterization included optical, fluorescence, and electron microscopy as well as adapted microindentation techniques to determine capsule break forces. Different model coatings were formulated making use of varying amounts of healing agent-charged microcapsules. The coated samples were then injured under controlled conditions (by Erichsen cupping in the case of steel, by simulated hail in the case of wood) and then subjected to salt spray testing (natural and accelerated), weathering, or even real-life exposure, always in comparison with a blank. The results obtained demonstrate the superiority of microcapsule-based self-healing coatings compared to classical protection formulations, and appropriate coating systems have in the meantime been successfully brought to the market.  相似文献

At the Keck Smart Materials Integration Laboratory at Penn State University, low-temperature co-fired ceramic (LTCC) material systems have been used to fabricate a number of devices for a variety of applications. This article presents an overview of the integration of the concepts and materials that we have used to achieve miniaturization and unique device function. Examples of microwave filters, metamaterial antennas, and a dielectrophoretic cell sorter will be presented, with emphasis on device modeling and design, prototype construction methods, and test results.  相似文献

The growing production of biodiesel as a renewable source-based fuel leads to an increased amount of glycerol. Thus, it is a favorable starting material to obtain highly functionalized products. From a variety of catalytic reactions three examples, namely glycerol oxidation, glycerol hydrogenolysis and aqueous-phase reforming, were chosen for detailed studies in our group. The experimental focus for the oxidation of glycerol was set on preparation and detailed examination of supported Pt–Bi catalysts in batch reactions as well as in continuous experiments using a trickle bed reactor. For aqueous-phase reforming of glycerol to hydrogen the addition of tin to supported platinum catalysts was investigated. Ruthenium and copper based catalysts could be successfully applied in the hydrogenolysis of glycerol to 1,2-propanediol.  相似文献

The enantioselective synthesis of an analogue of scyphostatin, a potent inhibitor of the neutral sphingomyelinase, is described. The synthesis starts with cyclohexanone and a protected D-serine derivative. The key step is an asymmetric hydroxylation to access a hydroxycyclohexanone, which is transformed into a substituted hydroxycyclohexenone. This is converted into the scyphostatin analogue 14, a chemically and metabolically stabilised compound lacking the epoxy function of the natural congener and carrying a palmitic acid group instead of the native trienoyl residue. An evaluation of the biological activity of 14 revealed neutral sphingomyelinase inhibition in several in vivo test systems (monocytes, macrophages, hepatocytes) monitoring antiapoptotic effects and the inversion of phorbolester-induced translocation of green fluorescent protein labelled kinase (protein kinase C-alpha).  相似文献

A single-phase problem is solved rather than a multiphase problem for numerical simplicity: and the solution is based on the assumption that the region of gas or plasma can be treated as a void because solid or liquid steel has a greater density level than gas or plasma. The volume-of-fluid method, which can calculate the free surface shape of the keyhole, is used in conjunction with a ray-tracing algorithm to estimate the multiple reflections. Fresnel's reflection model is simplified by the Hagen-Rubens relation for handling a laser beam interaction with materials. Factors considered in the simulations include buoyancy force, Marangoni force and recoil pressure; furthermore, pore generation is simulated by means of an adiabatic bubble model, which can also lead to the phenomenon of a keyhole collapse. Models of the shear stress on the keyhole surface and of the heat transfer to the molten pool via a plasma plume are introduced in simulations of the weld pool dynamics. Analysis of the temperature profile characteristics of the weld bead and molten pool flow in the molten pool is based on the results of the numerical simulations. The simulation results are used to estimate the weld fusion zone shape; and the results of the simulated fusion zone formation are compared with the results of the experimental fusion zone formation and found to be in good agreement. The effects of laser beam profile (Gaussian vs. measured), vapor shear stress, vapor heat source and sulfur content on the molten pool behavior and fusion zone shape are analyzed.  相似文献

A set of fiber-optic sensing experiments that can be performed by senior undergraduates in a fiber optics laboratory is presented. The experiments are based on two stable phase-modulated sensors: a miniaturized Michelson-type fiber-optic interferometer that is relatively insensitive to temperature drifts; and a two-mode fiber used in a differential interferometric sensing scheme. Methods of construction are described, and several possible experiments are presented. The creation of graft copolymers by selectively grafting a second polymer to the amorphous fraction of a semi-crystalline polymer in supercritical CO2 is demonstrated herein. The graft copolymer is synthesized by free radical polymerization of a vinyl monomer within the semi-crystalline polymer below its melt temperature. Such conditions afford selective grafting on the amorphous regions (block “B”) while leaving the crystalline domains (block “A”) unmodified. Accordingly, unique A-B, A-B-A, A-B-A-B-A, and so forth. block structures are formed. In this work, styrene is polymerized within polyamide 6, polyethylene terephthalate, and isotactic polypropylene. Purification of these material is performed to remove the un-grafted homopolymer, allowing for determination of the graft yield, the portion of polymer which covalently bonds to the semi-crystalline matrix. Grafting yields achieved in polyamide 6, polyethylene terephthalate, and isotactic polypropylene were 98%, 59%, and 15%, respectively. Property enhancements were observed upon further characterization of polystyrene-polyamide 6 copolymers, including high glass transition temperatures, the ability to be remelted, and tunable grafting molecular weight. Additionally, hydrophobicity is controlled by varying polystyrene composition. The remarkable range of accessed properties demonstrates this as a potential route to upcycling plastics.  相似文献
