This technical note describes a novel method for forming external threads in Superinvar. Superinvar [Interferometric measurement of the dimensional stability of Superinvar, UCRL-53787, 1988] is a nickel–cobalt steel (64% Fe, 31% Ni, and 5% Co) with a low coefficient of expansion (0.5 ppm K−1). This alloy finds application in precision assemblies where thermally induced dimensional changes are of concern. This technical note describes a novel process for the manufacture of high-quality adjustment screws (1/4 in. diameter, 2 in. long, 80 threads per inch—TPI) in Superinvar for use in precision adjustment mechanisms. The quality of the threads produced by this technique is superior to that of threads produced by single-point cutting and compares favorably with commercially produced rolled threads. This technique is best suited to economical production of small quantities.  相似文献

Tertiary wastewater treatment plant effluent before and after ozonation (0.6-1.1 g O3/g DOC) was tested for aquatic ecotoxicity in a battery of standardised microbioassays with green algae, daphnids, and zebrafish eggs. In addition, unconjugated estrogen and 17β-hydroxyandrogen immunoreactive substances were quantified by means of enzyme immunoassays, and endocrine effects were analysed in a 21-day fish screening assay with adult male and female medaka (Oryzias latipes). Ozonation decreased estrogen-immunoreactivity by 97.7 ± 1.2% and, to a lesser extent, androgen-immunoreactivity by 56.3 ± 16.5%. None of the short-term exposure ecotoxicity tests revealed any adverse effects of the tertiary effluent, neither before nor after the ozonation step. Similarly in the fish screening assay, reproductive fitness parameters showed no effects attributed to micropollutants, and no detrimental effects of the effluents were observed. Based on the presented screening, ozonation effectively reduced steroid hormone levels in the wastewater treatment plant effluent without increasing the effluent's ecotoxicity.  相似文献

Subclinical mastitis is a costly disease and its diagnosis is difficult. Besides the somatic cell count (SCC) and bacteriology, the differential inflammatory cell count (DICC) is a meaningful tool for mastitis detection. As microscopy is very subjective because of the low number of events to be counted, flow cytometry has often been proposed for the differentiation of milk cells. The objective of this study was to determine whether it is possible to identify subclinical mastitis in cattle at an early stage by a simple and fast flow cytometric method. The aim was to identify the main leucocyte populations in flow cytometric dotplots (polymorphonuclear neutrophils (PMN), lymphocytes and macrophages) and, with these, to elaborate a method of mastitis prognostics. Milk from 15 German Holstein cows was sampled in cross-sectional studies and SCC determined. After preparation, the milk cells were incubated with different specific antibodies that bind to different cell types and also to propidium iodide (PI), which differs between viable and non-viable cells. This procedure made it possible to localize cell types in a flow cytometric dot plot and to differentiate between viable and non-viable PMN. Percentages of viable PMN can be determined by a procedure consisting of a simple centrifugation, incubation with PI, and flow cytometric measurement. So it is possible to quickly determine the stage of the inflammation even in quarters with a low SCC.  相似文献

Drug load plays an important role in the development of solid dosage forms, since it can significantly influence both processability and final product properties. The percolation threshold of the active pharmaceutical ingredient (API) corresponds to a critical concentration, above which an abrupt change in drug product characteristics can occur. The objective of this study was to identify the percolation threshold of a poorly water-soluble drug with regard to the dissolution behavior from immediate release tablets. The influence of the API particle size on the percolation threshold was also studied. Formulations with increasing drug loads were manufactured via roll compaction using constant process parameters and subsequent tableting. Drug dissolution was investigated in biorelevant medium. The percolation threshold was estimated via a model dependent and a model independent method based on the dissolution data. The intragranular concentration of mefenamic acid had a significant effect on granules and tablet characteristics, such as particle size distribution, compactibility and tablet disintegration. Increasing the intragranular drug concentration of the tablets resulted in lower dissolution rates. A percolation threshold of approximately 20% v/v could be determined for both particle sizes of the API above which an abrupt decrease of the dissolution rate occurred. However, the increasing drug load had a more pronounced effect on dissolution rate of tablets containing the micronized API, which can be attributed to the high agglomeration tendency of micronized substances during manufacturing steps, such as roll compaction and tableting. Both methods that were applied for the estimation of percolation threshold provided comparable values.  相似文献

Lysine acylation constitutes a major group of post‐translational modifications of proteins, and is found in the proteomes of organisms from all kingdoms of life. Sirtuins are considered the main erasers of these modification marks, and thus contribute to acylation‐dependent regulation of enzyme activity, and potentially of protein quality control. We have established a substrate scaffold to enable the analysis of sirtuin activity with a broad range of acyl‐lysine modifications, including hydrophobic fatty acids. Characterization of the deacylase activity of the bacterial SrtN, which is encoded by the yhdZ gene of Bacillus subtilis, showed that this enzyme is capable of removing a broad range of acyl groups. These investigations further showed that SrtN and human SIRT1 are efficient lysine‐deformylases, thereby providing a first clue as to how this nonenzymatic modification might be removed from affected proteins.  相似文献

Visual analysis is widely used to study the behavior of molecules. Of particular interest are the analysis of molecular interactions and the investigation of binding sites. For large molecules, however, it is difficult to detect possible binding sites and paths leading to these sites by pure visual inspection. In this paper, we present new methods for the computation and visualization of potential molecular paths. Using a novel filtering method, we extract the significant paths from the Voronoi diagram of spheres. For the interactive visualization of molecules and their paths, we present several methods using deferred shading and other state-of-the-art techniques. To allow for a fast overview of reachable regions of the molecule, we illuminate the molecular surface using a large number of light sources placed on the extracted paths. We also provide a method to compute the extension surface of selected paths and visualize it using the skin surface. Furthermore, we use the extension surface to clip the molecule to allow easy visual tracking of even deeply buried paths. The methods are applied to several proteins to demonstrate their usefulness.  相似文献

The authors tested the hypothesis that the effects of nicotine on affect are moderated by the presence or absence of emotionally positive and negative stimuli and by attentional choice to avoid attending to emotionally negative stimuli. Thirty-two habitual smokers were assigned to tasks allowing attentional freedom to look back and forth at 2 simultaneously presented pictures, whereas another 32 habitual smokers viewed single pictures without attentional choice. Picture contents in both tasks were 1 of 4 combinations: emotionally negative + neutral, negative + positive, positive + neutral, or neutral + neutral. Participants wore a nicotine patch on 1 day and placebo patch on another day. Nicotine reduced anxiety most when negative pictures were presented in combination with neutral pictures, but it had no effect on anxiety when negative pictures were presented in combination with positive pictures and when negative pictures were not presented. In contrast, nicotine only reduced depressive affect when the participant had attentional choice between positive and negative pictures. Nicotine also enhanced positive affect and reduced negative affect as measured by the Positive and Negative Affect Schedule, but these effects were not moderated by task manipulations. Overall, the findings support the view that nicotine's ability to reduce specific negative affects is moderated by emotional context and attentional freedom. Nicotine tended to enhance eye-gaze orientation to emotional pictures versus neutral pictures in women, but it had no significant effect on eye-gaze in men. This paper describes approaches to topologically segmenting 2D time-dependent vector fields. For this class of vector fields, two important classes of lines exist: stream lines and path lines. Because of this, two segmentations are possible: either concerning the behavior of stream lines or of path lines. While topological features based on stream lines are well established, we introduce path line oriented topology as a new visualization approach in this paper. As a contribution to stream line oriented topology, we introduce new methods to detect global bifurcations like saddle connections and cyclic fold bifurcations as well as a method of tracking all isolated closed stream lines. To get the path line oriented topology, we segment the vector field into areas of attracting, repelling, and saddle-like behavior of the path lines. We compare both kinds of topologies and apply them to a number of test data sets.  相似文献

We present an approach to analyze mixing in flow fields by extracting vortex and strain features as extremal structures of derived scalar quantities that satisfy a duality property: they indicate vortical as well as high-strain (saddletype) regions. Specifically, we consider the Okubo-Weiss criterion and the recently introduced MZ-criterion. While the first is derived from a purely Eulerian framework, the latter is based on Lagrangian considerations. In both cases high values indicate vortex activity whereas low values indicate regions of high strain. By considering the extremal features of those quantities, we define the notions of a vortex and a strain skeleton in a hierarchical manner: the collection of maximal 0D, 1D and 2D structures assemble the vortex skeleton; the minimal structures identify the strain skeleton. We extract those features using scalar field topology and apply our method to a number of steady and unsteady 3D flow fields.  相似文献
