Nellore beef cattle, a Bos indicus (Zebu) breed, is well adapted to tropical conditions and has allowed Brazil to become one of the largest producers of red meat. Nevertheless, B. indicus breeds are reported to have less tender meat than Bos taurus. This study was designed to identify genes associated with meat tenderness and thus provides important information for breeding programs. A group of 138 animals was evaluated for longissimus thoracis muscle shear force (SF). Animals with the highest and lowest SF values (six animals each) were then selected for protein abundance studies. Samples were subjected to two-dimensional gel electrophoresis (2-DE) followed by peptide sequencing through mass spectrometry (MS) to identify differentially expressed proteins associated with SF values. Seventeen differentially expressed spots were observed (p < 0.05) between the two groups. The 13 proteins identified included structural proteins (alpha actin-1, MLC1, MLC3, MLC2F and tropomyosin), related to cell organization (HSPB1 and HSP70), metabolism (beta-LG, ACBD6 and Complex III subunit I) and some uncharacterized proteins. Results confirm the existence of differentially expressed proteins associated with SF, which can lead to a better understanding of mechanisms involved in meat tenderness.  相似文献

Cereals are a very important part of the human and animal diets. However, agricultural products can be contaminated by moulds and their mycotoxins. Plant extracts, particularly those of Equisetum arvense and Stevia rebaudiana have been reported previously to contain antioxidant compounds which may have antifungal properties. In this study, E. arvense and S. rebaudiana extracts were tested for their control of mycotoxigenic fungi in maize. The extracts were tested separately and as a mixture for their effect on growth of Aspergillus flavus and Fusarium verticillioides. Extracts were added to unsterilised inoculated maize at different water activity (a(w)) levels (0.85-0.95). Moulds were inoculated and incubated for 30 days. Results confirmed that the extract of E. arvense and a mixture 1:1 of Equisetum-Stevia may be effective for the inhibition of both growth of A. flavus and aflatoxin production at high water activity levels (pre-harvest conditions). In general, growth of the F. verticillioides was reduced by the use of plant extracts, especially at 0.95 a(w). However, fumonisin presence was not significantly affected. E. arvense and S. rebaudiana extracts could be developed as an alternative treatment to control aflatoxigenic mycobiota in moist maize.  相似文献

To design a specific lactic culture for the controlled manufacture of coalho cheese, 13 Lactobacillus rhamnosus, two Lactobacillus fermentum and one Lactobacillus plantarum strains isolated from artisanal coalho cheeses were identified and characterised. Two Lbrhamnosus, one Lbplantarum and one Lb. fermentum were selected and grouped in pairs designing four different culture formulations that demonstrated a good performance in cheesemaking experiments at pilot scale. Further studies to adjust the balance of strains used are necessary to attain adequate sensorial and technological attributes as expected for artisanal cheeses.  相似文献

ABSTRACT

For academic design programmes and research councils, finding mechanisms to measure impact and valuate co-design research projects is very difficult. The metric parameters – number of attendees, volume, shape, length – have shown to be insufficient. The subject matter, the subjects and matters, do not easily fit within these parameters, which fail to translate the value of a ‘constellation of affections’ that emerge throughout co-design processes. Emphasis on quantifiable impact derives from particular ways of understanding production, reproduction, and dissemination of knowledge. This paper understands academic design programmes as a technology to discuss axiology in co-design practices. This paper re-configures a techno-logic of value; from chain of value to value constellation, intersecting a co-design case study with readings of Simondon’s philosophy of technology and Barad’s agential realism. Ageing Playfully was a knowledge exchange project from The Creative Exchange programme, that involved people living with dementia and their caregivers co-designing interventions to boost mobility and well-being. At the core, this paper, explores an axiological move that informs a framework of emergence, a way of thinking in methods to account for the value of co-design as site of emergence.  相似文献

Cell therapies offer exciting new opportunities for effectively treating many human diseases. However, delivery of therapeutic cells by intravenous injection, while convenient, relies on the relatively inefficient process of homing of cells to sites of injury. To address this limitation, a novel strategy has been developed to load cells with superparamagnetic iron oxide nanoparticles (SPIOs), and to attract them to specific sites within the body by applying an external magnetic field. The feasibility of this approach is demonstrated using human mesenchymal stem cells (hMSCs), which may have a significant potential for regenerative cell therapies due to their ease of isolation from autologous tissues, and their ability to differentiate into various lineages and modulate their paracrine activity in response to the microenvironment. The efficient loading of hMSCs with polyethylene glycol‐coated SPIOs is achieved, and it is found that SPIOs are localized primarily in secondary lysosomes of hMSCs and are not toxic to the cells. Further, the key stem cell characteristics, including the immunophenotype of hMSCs and their ability to differentiate, are not altered by SPIO loading. Through both experimentation and mathematical modeling, it is shown that, under applied magnetic field gradients, SPIO‐containing cells can be localized both in vitro and in vivo. The results suggest that, by loading SPIOs into hMSCs and applying appropriate magnetic field gradients, it is possible to target hMSCs to particular vascular networks.  相似文献

This work shows heat transfer and friction characteristics for water single-phase flow in micro-fin tubes. The analysis of thermal and hydraulic behavior from a laminar to a turbulent flow was carried out in an experimental setup with a 9.52 mm diameter micro-fin tube. The tube was wrapped up with an electrical resistance tape to supply a constant heat flux to its surface. Different operational conditions were considered in the heating tests. The inlet and outlet temperatures, differential wall temperatures along the tube, pressure drop and flow rate were measured. The relationships of heat flux and flow rate with heat transfer coefficient and pressure drop were analyzed. Under the same conditions, comparative experiments with an internally smooth tube were conducted. The micro-fin tube provides higher heat transfer performance than the smooth tube (in turbulent flow hmicro-fin/hsmooth=2.9). In spite of the increase in pressure drop (Δpmicro-finpsmooth=1.7) the heat transfer results were significantly higher (about 80%). This shows the advantages of this enhanced configuration in thermal performance related to conventional tubes. The smooth tube results were validated by the comparison with the Dittus–Boelter and Gnielinski correlations. For the micro-fin tube an empirical correlation to the heat transfer coefficient adjusted from the set of measured data is proposed. The values obtained are in conformity with experimental results.  相似文献

In this study chick embryo optic cups at HH stage 13 of development were analyzed under normal conditions and after inoculation with colchicine for 1, 2, 4, and 8 h. Several changes were seen after these periods of treatment: 1) modifications of the structure, with thicker regions in the cup and a general decrease in the total volume according to the duration of exposure to the drug (about 4 times less than normal, 5,035 x 10(3) microns 3 vs 1,334 x 10(3) microns 3 after 8 h of treatment); 2) enlargement of the ventricular cavity and its closure, due to failure of approximation of retinal and pigmentary layers; 3) failure of lens development, with delay and impairment of pit formation and deformation of all structures; lens volume was less than normal (about 4 times less, 2,148 x 10(3) microns 3 vs 658 x 10(3) microns 3 after 8 h of treatment); 4) a general segregation of the cells making up the structure, principally in the more active proliferating zones.
