The enzymatic hydrolysis still stands as a challenge regarding the use of renewable feedstocks, as well as of lignocellulosic materials. The present work reports optimization of swollenin addition to pretreated sugarcane bagasse, in a step prior to the enzymatic hydrolysis, named pre-conditioning. Also, it describes further fermentation of the hydrolyzed material to d-lactic acid. Trichoderma harzianum enzymatic cocktail was produced from pretreated sugarcane bagasse, while swollenin was produced using Aspergillus niger as a host for recombinant protein expression. For pre-conditioning optimization, a Central Composite Rotatable Design was applied and validated. The optimized conditions were used to produce the hydrolysate that was fermented to D-lactic acid by Lactobacillus coryniformis subsp. torquens. It could be observed that pre-conditioning effectively leads to improvements in glucose release: 62% of hydrolysis efficiency. Besides, it was possible to use the produced hydrolysate to obtain d-lactic acid, with a yield of 1.34?g/g and a productivity of 1.59?g/L h?1. 相似文献

In-cylinder pressure analysis is a key tool for engine research and diagnosis; however, it normally requires to process the experimental signal for providing valuable information. Usual four-step data processing consists on level correction, angle referencing, cycle averaging, and filtering. Concerning the last two issues, ad-hoc methods and experience-based algorithms are mostly used, and there is not a consensus in the scientific community about the optimal way to proceed. In-cylinder pressure measurement has historically been used for off-line combustion diagnosis, but online application for real-time combustion control has become of great interest. This work considers low computing-cost methods for analysing the instant variation of the chamber pressure, directly obtained from the electric signal provided by a traditional piezoelectric sensor. Presented methods are based on the detection of sudden changes in the chamber pressure, which are amplified by the pressure derivative, and which are due to thermodynamic phenomena within the cylinder. Signal analysis tools both in time and in time-frequency domains are used for detecting the start of combustion, the end of combustion and the heat release peak. The need to ensure the microbiological quality and safety of food products has stimulated interest in the use of mathematical models for quantifying and predicting microbial behaviour. For 20 years, predictive microbiology has been developed for predicting the occurrence of food-borne pathogens, although these tools are dedicated to bacteria. Recently, the situation has changed and a growing number of studies are available in the literature dealing with the predictive modelling approach of fungi. Class 1 myosins (Myo1s) were the first unconventional myosins identified and humans have eight known Myo1 isoforms. The Myo1 family is involved in the regulation of gene expression, cytoskeletal rearrangements, delivery of proteins to the cell surface, cell migration and spreading. Thus, the important role of Myo1s in different biological processes is evident. In this study, we have investigated the effects of pentachloropseudilin (PClP), a reversible and allosteric potent inhibitor of Myo1s, on angiogenesis. We demonstrated that treatment of cells with PClP promoted a decrease in the number of vessels. The observed inhibition of angiogenesis is likely to be related to the inhibition of cell proliferation, migration and adhesion, as well as to alteration of the actin cytoskeleton pattern, as shown on a PClP-treated HUVEC cell line. Moreover, we also demonstrated that PClP treatment partially prevented the delivery of integrins to the plasma membrane. Finally, we showed that PClP caused DNA strand breaks, which are probably repaired during the cell cycle arrest in the G1 phase. Taken together, our results suggest that Myo1s participate directly in the angiogenesis process. 相似文献

The slag-corrosion behavior of two alumina–magnesia–carbon refractories (AMC) with different antioxidants (Si and Al) has been comparatively studied by means of thermodynamic simulation and laboratory tests. A cup test (static) at 1723 K and 1873 K and dipping test (dynamic) at 1873 K were carried out using a steelmaking ladle slag. An iterative method that considers the change of the liquid's composition while it penetrates the refractory was employed for the thermodynamic calculation. The simulation as well as the static tests showed similar performance between both refractories (5% of wear at 1723 K and 6% at 1873 K). In spite of the type of antioxidant, the material with smaller particles of magnesia exhibited better performance in the dynamic test, with 55% less wear than the other AMC brick. This behavior was attributed to the faster MgAl2O4 spinel formation, which increased the material's cohesion, especially the matrix, during the heating stage. The Si antioxidant improved oxidation resistance, although it did not have a positive effect on the corrosion resistance of the material in any of the performed tests. 相似文献
