The establishment and maintenance of axonal patterning is crucial for neuronal function. To identify the molecular systems that operate locally to control axonal structure, it is important to manipulate molecular functions in restricted subcellular areas for a long period of time. Microfluidic devices can be powerful tools for such purposes. In this study, we demonstrate the application of a microfluidic device to clarify the function of local Ca2+ signals in axons. Membrane depolarization significantly induced axonal branch-extension in cultured cerebellar granule neurons (CGNs). Local application of nifedipine using a polydimethylsiloxane (PDMS)-based microfluidic device demonstrated that Ca2+ entry from the axonal region via L-type voltage-dependent calcium channels (L-VDCC) is required for branch extension. Furthermore, we developed a method for locally controlling protein levels by combining genetic techniques and use of a microfluidic culture system. A vector for enhanced green fluorescent protein (EGFP) fused to a destabilizing domain derived from E. coli dihydrofolate reductase (ecDHFR) is introduced in neurons by electroporation. By local application of the DHFR ligand, trimethoprim (TMP) using a microfluidic device, we were able to manipulate differentially the level of fusion protein between axons and somatodendrites. The present study revealed the effectiveness of microfluidic devices to address fundamental biological issues at subcellular levels, and the possibility of their development in combination with molecular techniques.  相似文献

The article describes the development of a gait phase time-based split-belt treadmill measurement system. Conventional methods of measuring gait phase, such as the foot switch and force plate, require significant preparation and are costly. In this article, we propose a simple, cheap, and accurate gait phase measurement system that utilizes only the treadmill motor current value. Comparison of this algorithm with conventional methods reveals that the proposed algorithm is as accurate as the foot switch. Moreover, the proposed algorithm can estimate stance phase within a 0.2?s error of the measured value of the force plate in most cases (four out of five healthy subjects). This accuracy is higher than that of the foot switch which is widely used in the clinical field.  相似文献

For geographical information systems (GIS) to be useful in the management of Japanese paddy fields, it is vital to be able to prepare base maps that define sections of field. We attempted to prepare a base map by detecting the edges of submerged paddy fields from a SPOT image with the use of multi-resolution wavelet transform. The raw image of SPOT band 3 was decomposed into a low frequency approximation image and a set of high frequency detailed images to five levels, and then reconstructed. The reconstructed image at the fifth level was obtained from all of the high frequency images except for the low frequency approximation image. The image reconstructed up to full scale was then applied to a zero-crossing scheme and three post-processings—line thinning, removal of isolated pixels, and connection of pixels with their neighbours—in order to obtain clear edges. The spatial features of the image indicating the edges obtained by multi-resolution wavelet transform were compared quantitatively with those obtained by using a difference of Gaussian (DOG) filter. The multi-resolution wavelet transform was better than the DOG filter in that the base map obtained by the wavelet transform method represented a ‘field block’, which is typically composed of 10 holdings of paddy field, more precisely than with the DOG filter, and in that the number of open polygons on this map was smaller, indicating that the wavelet transform method is more suitable for producing the base map in GIS for the management of paddy fields.  相似文献

Nanostructured double ferromagnetic tunnel junctions (MTJs) are indispensable for investigation of spin-dependent single-electron transport at low temperature. A new fabrication process that enables us to reduce the size of MTJs down to nanometer scale by using the side edge of a patterned film were developed. The multilayers of MTJ partially replaced by thick Al2O3/Cu double layer were prepared by using electron beam lithography and lift-off, then Pt film was vacuum-evaporated onto the side edge of Al2O3/Cu film, which masked MTJ during following Ar ion milling. As a result, the double MTJs with the dimension of 10 nm £ 10 mm were formed beneath the Pt film. The large tunnel magnetoresistive ratio of 35% and symmetrical I–V characteristics were obtained at room temperature.  相似文献

The maximum measurable range of a spectral interference microscope depends on the coherence length of the light transmitted by its tunable spectral filter. To achieve a large range in step-height measurement we have developed a new tunable spectral filter that uses tandem liquid-crystal Fabry-Perot interferometers (LC-FPIs), which can simultaneously attain both a high spectral resolution and a large tuning range. Fringe visibility measurements were carried out, and it was found that the coherence length of the light transmitted through tandem LC-FPIs is two times larger than that transmitted through a single LC-FPI. Using this novel tunable spectral filter, we developed a new spectral interference microscope for the measurement of three-dimensional shapes of discontinuous objects. Experimental results of step-height measurements both with a single LC-FPI and with tandem LC-FPIs are presented for a combination of standard steel gauge block sets with 1-, 99-, and 100-microm steps. A large range (1-100 microm) of measurement with submicrometer resolution was achieved with tandem LC-FPIs that was not possible with our previous system in which a single LC-FPI was used.  相似文献

We have investigated the degradation mechanism of Al0.48In0.52As/In0.53Ga0.47As/ InP high electron mobility transistors (HEMTs) using WSi ohmic electrodes. Cross-sectional transmission electron microscopy (TEM) observation and en-ergy dispersive x-ray (EDX) analysis reveal impurities diffusion of gate electrode (titanium: Ti) and fluorine (F) in the AlInAs layer after a high temperature (Ta = 170°C operating life test for 500 h. The decrease of drain current (Ids) during life test shows linear dependence on square root of aging time. It suggests that the degradation is controlled by a diffusion mechanism. Hence, the estimated degradation mechanism of this device is related with decrease of carrier concentration in the epitaxial layer by these diffused impurities. On the other hand, TEM and EDX show no degradation of WSi/InGaAs interface after aging. Therefore, the WSi electrode for this type of HEMT demonstrates excellent high stability under the accelerated operating life test.  相似文献

Kawaguchi and Hatogaya City are located on the northern edge of Tokyo. We analysed between air pollution and prevalence rate of allergic diseases among elementary school children in this area. A prevalence rate of allergic diseases in 1996 May and June was as follows; bronchial asthma 13.5%, atopic dermatitis 24.5%, allergic rhinitis and/or conjunctivitis 22.8%, urticaria 12.4%, food allergy 7.8% and drug allergy 2.2%, respectively. Air pollution of this area was analysed to check the levels of nitrogen dioxide (NO2), sulfur dioxide (SO2) and suspended particulate matters (SPM). NO2 pollution was relatively high in urban area, and SPM pollution was especially high around the highways. SO2 pollution was lower than the environmental standard. No relationship was found between the prevalence rate of bronchial asthma, atopic dermatitis, allergic rhinitis and/or conjunctivitis and air pollution, but it was found that these diseases are slightly related to population density (p < 0.1, p < 0.01, p < 0.1, respectively).  相似文献

A new and efficient scheme is presented for evaluating the kernel function K, which occurs in the linearized theory of oscillatory airloads on a planar lifting surface in subsonic flow. Among many previous efforts of this sort, the present paper was inspired by Ueda's method [10] for computing the integral function B(X, r, k) appearing in the kernel.Firstly we point out that important are not only B but also the remaining part A of the kernel, since singularities are latent in A as well as B. Moreover, it is quite desirable to express the singularities, which often must be treated analytically, by using the original variable x0 rather than the modified one X.The second important problem concerns with the finite continuous part K?F, which must be numerically evaluated efficiently. A new method for this purpose is presented.This paper is restricted to the case where the frequency is real, namely the oscillation is neutrally stable.  相似文献
