Glioblastoma multiforme (GBM) is a particularly malignant primary brain tumor. Despite enormous advances in the surgical treatment of cancer, radio- and chemotherapy, the average survival of patients suffering from this cancer does not usually exceed several months. For obvious ethical reasons, the search and testing of the new drugs and therapies of GBM cannot be carried out on humans, and for this purpose, animal models of the disease are most often used. However, to assess the efficacy and safety of the therapy basing on these models, a deep knowledge of the pathological changes associated with tumor development in the animal brain is necessary. Therefore, as part of our study, the synchrotron radiation-based X-ray fluorescence microscopy was applied for multi-elemental micro-imaging of the rat brain in which glioblastoma develops. Elemental changes occurring in animals after the implantation of two human glioma cell lines as well as the cells taken directly from a patient suffering from GBM were compared. Both the extent and intensity of elemental changes strongly correlated with the regions of glioma growth. The obtained results showed that the observation of elemental anomalies accompanying tumor development within an animal’s brain might facilitate our understanding of the pathogenesis and progress of GBM and also determine potential biomarkers of its extension. The tumors appearing in a rat’s brain were characterized by an increased accumulation of Fe and Se, whilst the tissue directly surrounding the tumor presented a higher accumulation of Cu. Furthermore, the results of the study allow us to consider Se as a potential elemental marker of GBM progression. 相似文献

The global urgency to uncover medical countermeasures to combat the COVID-19 pandemic caused by the severe acute respiratory syndrome-coronavirus 2 (SARS-CoV-2) has revealed an unmet need for robust tissue culture models that faithfully recapitulate key features of human tissues and disease. Infection of the nose is considered the dominant initial site for SARS-CoV-2 infection and models that replicate this entry portal offer the greatest potential for examining and demonstrating the effectiveness of countermeasures designed to prevent or manage this highly communicable disease. Here, we test an air–liquid-interface (ALI) differentiated human nasal epithelium (HNE) culture system as a model of authentic SARS-CoV-2 infection. Progenitor cells (basal cells) were isolated from nasal turbinate brushings, expanded under conditionally reprogrammed cell (CRC) culture conditions and differentiated at ALI. Differentiated cells were inoculated with different SARS-CoV-2 clinical isolates. Infectious virus release into apical washes was determined by TCID50, while infected cells were visualized by immunofluorescence and confocal microscopy. We demonstrate robust, reproducible SARS-CoV-2 infection of ALI-HNE established from different donors. Viral entry and release occurred from the apical surface, and infection was primarily observed in ciliated cells. In contrast to the ancestral clinical isolate, the Delta variant caused considerable cell damage. Successful establishment of ALI-HNE is donor dependent. ALI-HNE recapitulate key features of human SARS-CoV-2 infection of the nose and can serve as a pre-clinical model without the need for invasive collection of human respiratory tissue samples. 相似文献

In this study, the conversion of silver ions into ∼30.74 nm sized silver nanoparticles (AgNPs) was achieved in 30 min at a reaction temperature of 80–90°C in aqueous leaf extract of Artemisia afra. The synthesised AgNPs showed surface plasmon resonance in the range of 423–438 nm. Spherical and face‐centred cubic nanoparticles were confirmed by transmission electron microscope (TEM) and X‐ray diffraction (XRD) analysis, respectively. Fourier transform infra‐red (FTIR) results indicated that the obtained nanoparticles were stabilised and capped through the carbonyl and carboxylate ion groups possibly from flavonoids, terpenoids, phenolics and esters content of the extracts. In addition, the AgNPs were assessed for their biological potentials against some microbes and, also, their free radical scavenging ability was established. The AgNPs exhibited interesting antimicrobial and antioxidant properties better than the aqueous extract of A. afra. We present further results of the simulation research on the constrained regularized least squares (CRLS) solution of the ill-conditioned inverse problem in spectral extinction (turbidimetric) measurements, which we originally presented in this journal [Appl. Opt. 49, 4591 (2010)]. The inverse problem consists of determining the particle size distribution (PSD) function of a particulate system on the basis of a measured extinction coefficient as a function of wavelength. In our previous paper, it was shown that under assumed conditions the problem can be formulated in terms of the discretized Fredholm integral equation of the first kind. The CRLS method incorporates two constraints, which the PSD sought will satisfy: nonnegativity of the PSD values and normalization of the PSD to unity when integrated over the whole range of particle size, into the regularized least squares (RLS) method. This leads to the quadratic programming problem, which is solved by means of the active set algorithm within the research. The simulation research that is the subject of the present paper is a continuation and extension of the research described in our previous paper. In the present research, the performance of the CRLS method variants is compared not only to the corresponding RLS method variants but also to other regularization techniques: the truncated generalized singular value decomposition and the filtered generalized singular value decomposition, as well as nonlinear iterative algorithms: The Twomey algorithm and the Twomey-Markowski algorithm. Moreover, two methods of selecting the optimum value of the regularization parameter are considered: The L-curve method and the generalized cross validation method. The results of our simulation research provide even stronger proof that the CRLS method performs considerably better with reconstruction of PSD than other inversing methods, in terms of better fidelity and smaller uncertainty. 相似文献

The relationship between speed and crashes has been well established in the literature, with the consequence that speed reduction through enforced or other means should lead to a reduction in crashes. The extent to which the public regard speeding as a problem that requires enforcement is less clear. Analysis was conducted on public perceptions of antisocial behaviors including speeding traffic. The data was collected as part of the British Crime Survey, a face-to-face interview with UK residents on issues relating to crime. The antisocial behavior section required participants to state the degree to which they perceived 16 antisocial behaviors to be a problem in their area. Results revealed that speeding traffic was perceived as the greatest problem in local communities, regardless of whether respondents were male or female, young, middle aged, or old. The rating of speeding traffic as the greatest problem in the community was replicated in a second, smaller postal survey, where respondents also provided strong support for enforcement on residential roads, and indicated that traveling immediately above the speed limit on residential roads was unacceptable. Results are discussed in relation to practical implications for speed enforcement, and the prioritization of limited police resources. 相似文献

We report the changes in dispersion relations of hypersonic acoustic phonons in free-standing silicon membranes as thin as ~8 nm. We observe a reduction of the phase and group velocities of the fundamental flexural mode by more than 1 order of magnitude compared to bulk values. The modification of the dispersion relation in nanostructures has important consequences for noise control in nano- and microelectromechanical systems (MEMS/NEMS) as well as opto-mechanical devices. 相似文献

In this paper, we present the theoretical studies of a refractive index map to implement a Gauss to a J(0)-Bessel-Gauss convertor. We theoretically demonstrate the viability of a device that could be fabricated on a Si/Si(1-y)O(y)/Si(1-x-y)Ge(x)C(y) platform or by photo-refractive media. The proposed device is 200 μm in length and 25 μm in width, and its refractive index varies in controllable steps across the light propagation and transversal directions. The computed conversion efficiency and loss are 90%, and -0.457 dB, respectively. The theoretical results, obtained from the beam conversion efficiency, self-regeneration, and propagation through an opaque obstruction, demonstrate that a two-dimensional (2D) graded index map of the refractive index can be used to transform a Gauss beam into a J(0)-Bessel-Gauss beam. To the best of our knowledge, this is the first demonstration of such beam transformation by means of a 2D index-mapping that is fully integrable in silicon photonics based planar lightwave circuits (PLCs). The concept device is significant for the eventual development of a new array of technologies, such as micro optical tweezers, optical traps, beam reshaping and nonlinear beam diode lasers. 相似文献

The performance of silicon heterojunction (SHJ) solar cells is discussed in this paper in regard to their dependence on the applied amorphous silicon layers, their thicknesses and surface morphology. The emitter system investigated in this work consists of an n-doped, hydrogenized, amorphous silicon carbide a-SiC:H(n) layer with or without a pure, hydrogenized, intrinsic, amorphous silicon a-Si:H(i) intermediate layer. All solar cells were fabricated on p-type FZ-silicon and feature a high-efficiency backside consisting of a SiO2 passivation layer and a diffused local boron back surface field, allowing us to focus only on the effects of the front side emitter system. The highest solar cell efficiency achieved within this work is 18.5%, which is one of the highest values for SHJ-solar cells using p-type substrates. A dependence of the passivation quality on the surface morphology was only observed for solar cells including an a-Si:H(i) layer. It could be shown that the fill factor suffers from a reduction due to a reduced pseudo fill factor for emitter thicknesses below 11 nm due to a lower passivation quality and/or a higher potential for shunting thorough the a-Si emitter to the crystalline wafer with the conductive indium tin oxide layer. Furthermore, the influence of a variation of the doping gas flow (PH3) during the plasma enhanced chemical vapor deposition of the doped amorphous silicon carbide a-SiC:H(n) on the solar cell current-voltage characteristic-parameter has been investigated. We could demonstrate that a-SiC:H(n) shows in principle the same dependence on PH3-flow as pure a-Si:H(n). 相似文献
