Six synthetic heparin-like oligosaccharides have been used to investigate the effect of the oligosaccharide sulfation pattern on the stimulation of acidic fibroblast growth factor (FGF-1) induced mitogenesis signaling and the biological significance of FGF-1 trans dimerization in the FGF-1 activation process. It has been found that some molecules with a sulfation pattern that does not contain the internal trisaccharide motif, which has been proposed for high affinity for FGF-1, stimulate FGF-1 more efficiently than those with the structure of the regular region of heparin. In contrast to regular region oligosaccharides, in which the sulfate groups are distributed on both sides of their helical three-dimensional structures, the molecules containing this particular sulfation pattern display the sulfate groups only on one side of the helix. These results and the fact that these oligosaccharides do not promote FGF-1 dimerization according to sedimentation-equilibrium analysis, confirm the importance of negative-charge distribution in the activation process and strongly suggest that FGF dimerization is not a general and absolute requirement for biological activity. 相似文献

This paper evaluated mechanical and thermal stability of alkali-activated materials obtained from metakaolin and alternative silica sources, such as rice husk ash (RHA) and silica fume (SF), and were reinforced with recycled ceramic particles (RP) obtained by grinding bricks. Specimens were produced, and after 7 days of curing, they were exposed to temperatures between 300 and 1200°C to determine the influence that different percentages of RP had on the mechanical behavior and microstructure of the produced composites. The results showed a reduction in the linear contraction by 10.22% with 20 wt% RP and that the reinforcing materials improved the mechanical performance of the geopolymers after exposure to high temperatures; the compressive strengths reached 137.7 (±11.4) MPa after being exposed to 1200°C for the matrix based on RHA and 180.6 (±19.15) MPa after being reinforced with 20 wt% RP. The improvement was mainly due to densification and the formation of crystalline products such as leucite, kalsilite, and mullite. 相似文献

A series of interpenetrating polymer networks (IPNs) with different compositions were prepared from emulsions of silylated soybean oil (SilSoy) and hydrophilic polysiloxanes (HPPS). Silylation of the soybean oil was achieved via the “Ene reaction” and the water soluble polysiloxanes were prepared by hydrolysis and condensation polymerization of 3-aminopropyldiethoxymethylsilane followed by a reaction with ethylene carbonate. Unlike the triglyceride structure of SilSoy, HPPS are water soluble, linear polysiloxanes having a pendant hydroxyl group on every siloxane. Evaporation of the water phase from emulsions of SilSoy in aqueous solutions of HPPS yielded crosslinked IPN films composed of silanols and carbinol condensations that formed stable siloxanes (Si O Si) crosslink linkages as well as hydrolysable silicone-carbinol (Si O C) linkages. The entangled network that was obtained is characterized by intimate mixing of these incompatible phases with a typical IPN microphase separation. The crosslink density between the dispersed phase and the continuous phase was correlated with the swell-gel data. The mechanical and thermal properties were also studied and were correlated with the composition and the crosslinks density. These IPNs can be used as convenient environmentally degradable control release systems via the hydrolysis of the Si O C linkages. © 2018 Wiley Periodicals, Inc. J. Appl. Polym. This work describes the preparation of thermosensitive chitosan-graft-poly(N-vinylcaprolactam) nanoparticles by ionic gelation and their potential use as a controlled drug delivery system, using doxorubicin as a model drug. A systematic study of the effect of the main processing parameters on both the size and thermoresponsive behavior of nanoparticles was investigated. The size of the particles is strongly dependent on the length of the poly(N-vinylcaprolactam) grafted chains and the concentration of the copolymer and crosslinking agent solutions. The molecular structure of the copolymer plays an essential role in the phase transition temperature of the particles, which decreases with the length of PVCL grafted chain. The system displayed proper drug-association parameters, and the drug-loaded nanoparticles exhibited dose-dependent cytotoxicity. A significant increase in the doxorubicin delivery rate was observed above the phase transition temperature (40 °C). These features indicate that these nanoparticles are suitable for the development of a new thermally controlled anti-cancer drug delivery system. © 2019 Wiley Periodicals, Inc. J. Appl. Polym. The use of macroporous monolithic matrices in the purification of biocompounds is constantly growing and developing. In this work, the objective was to optimize the quantity of N-acetyl-D-glucosamine (D-GlcNAc) immobilized on the surface of macroporous polymeric cryogels for capture of lectins from less clarified solutions. Surface response methodology was applied and it was observed that the immobilization temperature of the glutaraldehyde (GLU) and the D-GlcNAc concentration influenced the amount of sugar immobilized. The matrices produced with 1.1% of allyl glycidyl ether were functionalized by GLU. Optimal maximum condition was obtained with mean value of 160.39 ± 26.38 mg of D-GlcNAc immobilized per gram of dry cryogel. Characterization analyses of the matrices showed that the activation process was effective, maintaining the macroporous structure and physical characteristics. The adsorbents produced were tested for capture of lectins from a crude protein solution of barley. At tested conditions, adsorbent capture around 11% of protein in solution but reduce the hemaglutinating capacity in 40%, demonstrating its selectivity. Quadruplex nucleic acids are promising targets for cancer therapy. In this study we used a fragment-based approach to create new flexible G-quadruplex (G4) DNA-interactive small molecules with good calculated oral drug-like properties, based on quinoline and triazole heterocycles. G4 melting temperature and polymerase chain reaction (PCR)-stop assays showed that two of these compounds are selective G4 ligands, as they were able to induce and stabilize G4s in a dose- and DNA sequence-dependent manner. Molecular docking studies have suggested plausible quadruplex binding to both the G-quartet and groove, with the quinoline module playing the major role. Compounds were screened for cytotoxicity against four cancer cell lines, where 4,4′-(4,4′-(1,3-phenylene)bis(1H-1,2,3-triazole-4,1-diyl))bis(1-methylquinolin-1-ium) ( 1 d ) showed the greater activity. Importantly, dose–response curves show that 1 d is cytotoxic in the human colon cancer HT-29 cell line enriched in cancer stem-like cells, a subpopulation of cells implicated in chemoresistance. The ubiquitin-proteasome system (UPS) is an established therapeutic target for approved drugs to treat selected hematologic malignancies. While drug discovery targeting the UPS focuses on irreversibly binding epoxyketones and slowly-reversibly binding boronates, optimization of novel covalent-reversibly binding warheads remains largely unattended. We previously reported α-ketoamides to be a promising reversible lead motif, yet the cytotoxic activity required further optimization. This work focuses on the lead optimization of phenoxy-substituted α-ketoamides combining the structure-activity relationships from the primed and the non-primed site of the proteasome β5 subunit. Our optimization strategy is accompanied by molecular modeling, suggesting occupation of P1′ by a 3-phenoxy group to increase β5 inhibition and cytotoxic activity in leukemia cell lines. Key compounds were further profiled for time-dependent inhibition of cellular substrate conversion. Furthermore, the α-ketoamide lead structure 27 does not affect escape response behavior in Danio rerio embryos, in contrast to bortezomib, which suggests increased target specificity. 相似文献

Medium-chain-length poly-3-hydroxyalkanoates (PHA) and carboxyl group-functionalized multiwalled carbon nanotubes (MC) were used to fabricate a composite membrane for application in a double-chambered microbial fuel cell (MFC). MC was composited into PHA at 5%, 10%, and 20% w/w via ultrasound dispersion blending method. PHA-MC composite was compared with Nafion 117 as proton exchange membrane in MFC operated with palm oil mill effluent (POME) wastewater. The composite exhibited prerequisite separator membrane characteristics. The dispersion of MC in the polymer matrix increased its interfacial surface area and water uptake properties. PHA-MC10% membrane in MFC showed maximum power density of 361?mW/m2, which was comparable with Nafion 117 (372?mW/m2). Internal resistance decrease, chemical oxygen demand (COD) removal, coulombic efficiency (CE), and conductivity of the PHA-MC10% were superior to Nafion 117. The environmental-friendly material could provide an alternative towards realizing practical MFC application. 相似文献

Solanum paludosum extracts obtained by maceration (SP-EtOHCRUDE and SP-EtOHMARC) and supercritical fluid extraction (SP-SFECO2) were evaluated. Three extracts with distinct chromatographic profiles were obtained: SP-EtOHCRUDE, with a complex phenolic composition; SP-SFECO2, containing methoxylated flavonoids; and SP-EtOHMARC, which bears mainly polar polyphenols. All extracts were strong antioxidants, as measured using the diphenylpicrylhydrazyl (DPPH) method, xanthine/xanthine oxidase/luminol and iron-induced lipid peroxidation systems. Extracts were also evaluated as modifiers of antibiotic activity in strains of Staphylococcus aureus expressing fluoroquinolone, macrolide and tetracycline efflux proteins. Despite their lack of antibacterial activity, all extracts were able to potentiate antibiotics activities by as much as eight fold. Both the maceration and supercritical fluid extraction processes were adequate to obtain extracts with different biological activities according to their patterns of phenolic compounds. The data suggest that S. paludosum could be a valuable source of antioxidants and putative efflux pumps inhibitors. 相似文献
